    Mechanistic studies of the phenobarbital-type induction of cytochromes P450 : role of AMP-activated protein kinase

    Inside the liver cells there are sophisticated mechanisms that have evolved over millions of years to metabolize toxic substances, many of which are fat-soluble compounds making them difficult for the body to excrete. Cytochromes P450 (CYPs) are drug-metabolizing enzymes predominantly expressed in the liver that catalyze the first step in the metabolism of lipophilic compounds into water-soluble and thus excretable compounds. Transcriptional activation of CYPs and other drug-metabolizing enzymes by drugs and xenobiotics in the liver is mediated by the mammalian constitutive androstane receptor (CAR) and pregnanes X receptor (PXR), whereas the chicken xenobiotic receptor (CXR) mediates drug responses in chicken hepatocytes. This phenomenon, which is called drug induction, is fully reversible and dose-dependent. By increasing the capability for metabolic detoxification and elimination, induction of CYPs is an integral part of the defense mechanism against xenochemical insult. However, it also has important negative clinical consequences, such as altered pharmacokinetics of drugs and carcinogens, drug-drug interactions, and changes in the metabolism of steroids, vitamin D, and other endogenous compounds. For these reasons, it is of great importance to understand the molecular mechanisms leading to drug induced gene expression of hepatic drugmetabolizing enzymes. In this thesis the role of the AMP-activated protein kinase (AMPK) in the phenobarbital-mediated transcriptional regulation of CYPs was studied in hepatoma cells (chicken and human), and in hepatocytes (human and mouse). We show that AMPK activation by phenobarbital occurs via increased reactive oxygen species (ROS) formation in mitochondria and that increased AMPK activity is necessary for drug induction. Furthermore, we demonstrate that phenobarbital-type drugs affect the AMPK upstream kinase LKB1 and the protein kinase C zeta (PKCζ) activities. Importantly, these studies reveal that the signaling cascade involved in drug responses is conserved in evolution from birds to mice and humans. In addition, CXR interacting proteins were identified, which may also bind CAR in mammalian liver. In conclusion, these findings provide new insights into the so far poorly characterized molecular mechanism by which drugs lead to transcriptional regulation of CYP enzymes

    Phenotypic and Functional Characterization of Human Memory T Cell Responses to Burkholderia pseudomallei

This is an open-access article distributed under the terms of the Creative Commons Attribution License, which permits unrestricted use, distribution, and reproduction in any medium, provided the original author and source are credited.Background: Infection with the Gram-negative bacterium Burkholderia pseudomallei is an important cause of community-acquired lethal sepsis in endemic regions in southeast Asia and northern Australia and is increasingly reported in other tropical areas. In animal models, production of interferon-gamma (IFN-γ) is critical for resistance, but in humans the characteristics of IFN-γ production and the bacterial antigens that are recognized by the cell-mediated immune response have not been defined. Methods: Peripheral blood from 133 healthy individuals who lived in the endemic area and had no history of melioidosis, 60 patients who had recovered from melioidosis, and 31 other patient control subjects were stimulated by whole bacteria or purified bacterial proteins in vitro, and IFN-γ responses were analyzed by ELISPOT and flow cytometry. Findings: B. pseudomallei was a potent activator of human peripheral blood NK cells for innate production of IFN-γ. In addition, healthy individuals with serological evidence of exposure to B. pseudomallei and patients recovered from active melioidosis developed CD4+ (and CD8+) T cells that recognized whole bacteria and purified proteins LolC, OppA, and PotF, members of the B. pseudomallei ABC transporter family. This response was primarily mediated by terminally differentiated T cells of the effector–memory (TEMRA) phenotype and correlated with the titer of anti-B. pseudomallei antibodies in the serum. Conclusions: Individuals living in a melioidosis-endemic region show clear evidence of T cell priming for the ability to make IFN-γ that correlates with their serological status. The ability to detect T cell responses to defined B. pseudomallei proteins in large numbers of individuals now provides the opportunity to screen candidate antigens for inclusion in protein or polysaccharide–conjugate subunit vaccines against this important but neglected disease

    The uses of silence : a twentieth-century preoccupation in the light of fictional examples, 1900-1950

    A striking feature of twentieth-century Western cultural history was a preoccupation with silence. This thesis is a survey of the phenomenon across a broad range of literary and theoretical discourses actively engaged in the period in exploring and exploiting silence's expressive and philosophical potential. Its focus, and unifying principle, is the dynamic resourcefulness of the motif-the diversity of its uses and significations. The meaning of silence shifts according to its context and the discourse deploying it. By analysing an array of novels and theoretical formulations-by writers as diverse as James, Chopin, Conrad, H. D., Forster, Lawrence, Faulkner, and Wittgenstein, Benjamin, Blanchot, Hassan, Macherey, Irigaray, Spivak, Derrida-the mobility of silence as a construct is exposed. Silence is identified in the fiction of the period 1900-1950, and its implications are assessed in the light of the various ways in which its uses were understood and interpreted by twentieth-century theorists. Theory provides a heuristic device for the comprehension of the fiction selected for scrutiny whilst further highlighting the extent of the past century's dedication to the motif. Fiction and theory are regarded as two different manifestations of a fascination with silence: fiction dramatizes a commitment to the motif which comes to be formally registered in theoretical discourse as the century progresses. After an introductory chapter outlining the expanse of the phenomenon to be studied, the thesis is divided into two parts illustrating the discrete implications attaching to the motif: 'Social Silences' and 'Ontological Silences'. The project questions whether the multiplicity of silence's usage may work to depotentiate its signifying power; in particular, whether its role in abstract 'ontological' formulations diminishes its force for emancipatory 'social' discourses. In conclusion, by means of the synchronic organization of the thesis, silence's import is shown to lie in its resourcefulness rather than in any intrinsic characteristic it might be thought to possess

    The use of corporate reputation in the development of brand image strategy in the Taiwanese pharmaceutical industry

    This thesis was submitted for the degree of Doctor of Philosophy and awarded by Brunel University.This doctoral research aims to investigate the reputation building process of companies and to examine the applicability of western-developed theories about the uses of corporate reputation in a non-western context. It is the first study that synthesises three theories (value creation, strategic resources and corporate communication) to examine the strategic consequences of the uses of corporate reputation. Corporate reputation is an attribute or a set of attributes ascribed to a firm and inferred from the firm’s past actions. It is the belief of market participants about a firm’s strategic character (Weigelt and Camerer, 1988). Also, corporate reputation is the public’s cumulative judgement of a firm over time (Roberts and Dowling, 2002). The review of theoretical literature indicates the uses of corporate reputation by business organizations can be theorized along six dominant paradigmatic perspectives: 1-public relations; 2-marketing; 3-management, 4-economic; 5-sociological; 6-finance and accounting. The uses of corporate reputation in these six paradigms are comprehensively discussed. The objective of this study is to establish the use of corporate reputation in the development of brand image strategy. A review of the uses of the concept of corporate reputation is discussed in detail in chapter 2. The review of the literature also identified a research gap by showing that scarce research has been conducted on how these three main functions (value creation, strategic resources and corporate communication) affect a company’s brand positioning strategy. The following research question thus is proposed: How do (Taiwanese pharmaceutical) companies use their corporate reputation to develop a brand image strategy? The research hypotheses based on three theories (value-based theory, resource-based theory and integrated marketing communication theory) appear in Chapter 3. The research question is constructed theoretically, and then a conceptual model, which begins with three antecedents of corporate reputation and simultaneously illustrates the outcomes of their use, are discussed. The construct of the uses of corporate reputation has three dimensions: value creation, strategic resources and corporate communication. Each of these three dimensions includes several items. The items were proposed based on the previous researchers’ summaries and the qualitative interview. The researcher will then depict the proposed research conceptual framework and a number of hypotheses that will be further investigated and tested. Then the quantative study was completed by providing the data analysis and the results were explained. A multi-stage procedure was involved in this research. First, data examination and screening to prepare for subsequent quantitative analyses and then the descriptive statistics were presented. Second, a reliability test was performed on measurement scales to ensure that they achieve an acceptable level of reliability for further analysis. The resulting solutions were then re-assessed using confirmatory factor analysis. Finally, PLS (Partial Least Squares) was used to test the hypothesized relationships between the research constructs as postulated in the conceptual model, and to assess the overall goodness-of-fit between the proposed model and the collected data set. The researcher then discusses the validation of the measurement model and the research findings. The findings are then further discussed in terms of the contribution to marketing theory and relevance to marketing managers. Then the items of adapted scales were subjected to several rounds of adjustments and were finally found to possess acceptable measurement properties. Reliability and construct validity tests indicated that all scales satisfied widely accepted criteria such as the minimum reliability of 0.7. The results of scale purification will be discussed. And an evaluation of the research hypotheses and their significance are summarized, the findings of all hypotheses testing will be reviewed and compared with previous research. According to the research findings, the hypotheses that value creation, as one dimension of corporate reputation, has a positive impact on brand segmentation, brand differentiation and brand positioning are all accepted. The hypotheses that strategic resource, as one dimension of corporate reputation, has a positive impact on brand segmentation and brand differentiation are rejected. However, the hypothesis that strategic resource, as one dimension of corporate reputation, has a positive impact on brand positioning is accepted. The hypotheses that corporate communication, as one dimension of corporate reputation, has a positive impact on brand segmentation and brand differentiation are both accepted. The hypothesis that corporate communication, as one dimension of corporate reputation, has a positive impact on brand positioning is partially supported. Finally, the hypothesis that the (see in Table 5.18) moderating effect of price policy on corporate reputation has a positive impact on brand image strategy is partially but negatively supported. This thesis makes a significant contribution to the study of corporate reputation of firms in the Taiwanese pharmaceutical industry from the robustness of the qualitative and quantitative data collection

    Cellulose-Based Bio- and Nanocomposites: A Review

    Cellulose macro- and nanofibers have gained increasing attention due to the high strength and stiffness, biodegradability and renewability, and their production and application in development of composites. Application of cellulose nanofibers for the development of composites is a relatively new research area. Cellulose macro- and nanofibers can be used as reinforcement in composite materials because of enhanced mechanical, thermal, and biodegradation properties of composites. Cellulose fibers are hydrophilic in nature, so it becomes necessary to increase their surface roughness for the development of composites with enhanced properties. In the present paper, we have reviewed the surface modification of cellulose fibers by various methods. Processing methods, properties, and various applications of nanocellulose and cellulosic composites are also discussed in this paper

Wesley Alan MacKay, beloved husband of Mrs. Carle Louise MacKay of Canadian Forces Base Moose Jaw, Sask., and dear son of Mr. Mrs. Alan (Evelyn) MacKay of Warner, passed away suddenly near Latrobe, south of ^^9 Pennsylvania, Saturday, September 24th, 1988 at the age of 28 years. Capt. MacKay, a Canadian Armed rorces Snowbird pilot, died when the car he was driv­ing crashed into a tree in fog on a narrow roadway. TVro other Snowbird pilots were also injured in the accident, one seriously. Wesley was born in Magrath but moved to Camrose with his parents at an early age where he was raised and received his education. He began his military flying career as an instructor pilot at 2 CFFTS. He was then posted to CFB Summerside, P.E.I. where he logged 750 hours on the CP-121 Tracker. He has accumulated some 3500 hours since his enrolment in 1978. He joined the Snowbirds two years ago and was scheduled to finish his Snowbirds assignment in October. He was Lead Solo pilot and flew eighth position. Besides his loving wife and parents, survivors include two brothers, Dennis and Calvin and one sifter Connie Davies. Also his maternal grandmother Mrs. Nellie Seward of Magrath. . A private Memorial service was held Tuesday at Snowbirds hangar, CFB Base, Moose Jaw. Funeral Services will be held in Lethbridge Saturday, October 1st at Martin Bros.
