Role of Arbusccular mycorrhizal fungi and mycorrhizal endosymbiotic bacteria in increasing oil palm (Elaeis guineensis Jacq) seedlings adaptation towards biotic stress of fungal pathogen ganoderma boninense Pat.
Basal stem rot caused by Ganoderma boninense is the most serious disease of oil palm (Elaeis guineensis) in Indonesia and it has caused major loss in palm oil production. Under natural conditions, oil palm is often colonized by arbuscular mycorrhizal (AM) fungi and together with mycorrhizosphere bacteria offer possible advantages in increasing plant adaptation against biotic stress of pathogen. This research comprised five interrelated experiments, namely: (1) Exploration, Isolation and Identification of Arbuscular Mycorrhizal Fungi (AMF) and Mycorrhizal Endosymbiotic Bacteria from Oil Palm Rhizosphere and Their Ability in Inducing Oil Palm Adaptation Towards Biotic Stress of Pathogen Ganoderma boninense Pat; (2) Selection of Mycorrhizal Endosymbiotic Bacteria on Spores Germination of AMF Gigaspora margarita and Their Inhibition Towards Fungal Pathogen G. boninense Pat; (3) Identification of Active Compounds from Mycorrhizal Endosymbiotic Bacteria B. subtilis B10 that Inhibit the Growth of Fungal Pathogen G. boninense Pat; (4) Adaptation of Oil Palm Seedlings Inoculated with AMF and Mycorrhizal Endosymbiotic Bacteria B. subtilis B10 Towards Biotic Stress of Pathogen G. boninense Pat; (5) Analyzing the Response of Oil Palm Seedlings Inoculated with AMF and Mycorrhizal Endosymbiotic Bacteria in the Form of Active Compound Profile as Expression of Oil Palm Adaptation Towards Biotic Stress of Pathogen G. boninense Pat. The soil sampels were used in this experiment came from Aek Pancur Plantation belong to Indonesian Oil Palm Research Institute, Medan. The results showed that twenty isolates of mycorrhizal endosymbiotic bacteria were obtained from spores of AMF isolated from oil palm rhizosphere and were dominated by genus of Bacillus sp. Among these isolates, isolate B10 identified based on 16S rDNA as Bacillus subtilis B10, had the highest activity against G. boninense in vitro by producing intracellular active compounds with molecular weight 255.39 and postulated as of 2-(4-aminophenoxy)-6-methyl-tetrahydro-2H-pyran-3,4,5- triol. Dual inoculation of AMF and B. subtilis B10 on oil palm seedlings resulted in the lowest basal stem rot disease incidence caused by G. boninese which indicated disease severity index was only 5% and necrotic area of roots just 10%. Application of AMF together with B. subtilis B10 also increased oil palm seedlings height, root dry weight and stem diameter at 52 weeks after planting. The adsorption of nitrogen (N), phosphorus (P), potassium (K) and magnesium (Mg) increased when seedlings were co-inoculated with AMF and bacteria B. subtilis B10 together. This finding is important in terms of recommendation for application of AMF and bacteria B. subtilis B10 as biocontrol of pathogen G. boninense causal agent of basal stem rot in oil palm.Penyakit busuk pangkal batang yang disebabkan oleh fungi Ganoderma boninense merupakan penyakit yang paling serius pada kelapa sawit (Elaeis guineensis) di Indonesia karena menyebabkan kerugian besar dalam produksi minyak kelapa sawit. Peningkatan Produksi 8-Hydroxy 9,12-Octadecadienoic Acid (8-HODE) dari Kapang Endofit Curvularia lunata BioMCC Fe-00283 dengan Optimasi Media Fermentasi
8-Hydroxy 9,12-octadecadienoic acid (8-HODE) adalah salah satu asam lemak alifatik dari kelompok senyawa oksilipin yang menunjukkan aktivitas biologis. Telah dilaporkan sebelumnya bahwa Curvularia lunata BioMCC FE-00283, kapang endofit yang diisolasi dari tanaman Cibotium barometz, pada kultur fermentasi cair mampu menghasilkan 8-HODE. Glukosa, monosodium glutamat dan minyak jagung adalah komponen media fermentasi yang menunjukkan pengaruh nyata terhadap produksi 8-HODE oleh C. lunata BioMCC FE-00283. Dalam tulisan ini dilaporkan peningkatan produksi 8-HODE dengan menambahkan ekstrak kentang pada komposisi media hasil optimasi sebelumnya. Empat komponen media yang terdiri dari ekstrak kentang, glukosa, monosodium glutamat dan minyak jagung dioptimasi untuk produksi 8-HODE menggunakan Response Surface Metodology (RSM). Rancangan full factorial Central Composite Design (CCD) digunakan dalam percobaan ini agar dapat menjelaskan interaksi antar komponen media fermentasi. Hasil optimasi menunjukkan bahwa produksi 8-HODE maksimal yang diprediksi oleh model kuadratik adalah 16,46 mg/L. Hasil optimal tersebut dicapai pada komposisi media yang terdiri dari ekstrak kentang 36,50%; glukosa 3,60 g/L; monosodium glutamat 15,92 g/L dan minyak jagung 0,96 ml/L.Verifikasi komposisi media hasil optimasi menghasilkan 8-HODE sebesar 16,189 ± 0,526 mg/L. Hasil optimasi dengan menambahkan ekstrak kentang dalam optimasi media fermentasi ternyata meningkatkan konsentrasi 8-HODE tiga kali lipat dibandingkan dengan hasil optimasi dengan 3 peubah
CYTOTOXIC PROTEIN FROM Trichosanthes cucumerina L. var anguina (L.) Haines
Three proteins were discovered, they were: TF2 from fruit, TS3 from seed and TR3 from root of Trichosanthes cucumerina L. var anguina (L.) Haines by extraction with Phosphate Buffer Saline (PBS). The proteins were precipitated using 80% saturated ammonium sulphate and then were dialysed using cellophane. Furthermore, the dialysate was fractionated through gel filtration chromatography. The highest protein yield was 1.109%, resulted from TS3, then 0.356% from TR3, and the lowest was 0.014% from TF2. Characterization of the proteins using SDS-PAGE indicated that all of the protein fractions showed more than one sub unit molecular masses (Mr) in the range of 16 - 29 kDa. The toxicity (LC50) of proteins on brine shrimp lethality test were within range of 19 µg/ml - 25 µg/ml. The cytotoxicities of the TR3 and TS3 proteins on cancer cell line indicated that both of the active proteins inhibit proliferation of HeLa and K-562 cells with IC50 up to 45 µg/ml. Pengaruh Customer Relationship Manajemen dan Kualitas PelayananTerhadap Kepuasan Pasien serta Dampaknya kepada Loyalitas Pasien Non BPJS
Puskesmas as a First Level Health Facility that provides individual health services and community health services, is responsible for its working area. Public health efforts are made to increase public knowledge of the quality of health services it receives. This study aims to determine the activities of Customer Relationship Management and service quality that can affect patient satisfaction and its impact on patient loyalty of non-Social Security Administering Bodies (BPJS) at the Pasar Rebo District Health Center and the Kramat Jati District Health Center. This research method is a survey method. The selection of respondents was carried out using random sampling techniques on non-BPJS patients at the Kramat Jati Subdistrict Health Center and Pasar Rebo District Health Center and primary data was collected using a validated questionnaire. The research approach is quantitative descriptive and statistical data analysis of research results using Structural Equation Modeling. Data analysis from 200 completed questionnaires that meet the inclusion criteria. Based on the results of the study, the path coefficient value on patient satisfaction is Customer Relationship Management of 0.266, service quality of 0.773, while the results of research on patient loyalty are Customer Relationship Management of 0.270, service quality of 0.191 and patient satisfaction of 0.421. Effective Customer Relationship Management, good service quality and high patient satisfaction can increase patient loyalty
ANTIPROLIFERATIVE PROTEIN FROM TRICHOSANTHES CUCUMERINA L. VAR ANGUINA (L.) HAINES
Three proteins were isolated from plant parts of Trichosanthes cucumerina L. var anguina (L.) Haines, they were TF2 from fruit, TS3 from seed and TR3 from root with molecular masses (Mr) approximately 16 - 64 kDa on SDS-PAGE characterization. The proteins were extracted with Phosphate Buffer Saline (PBS), then they were precipitated using 80% saturated ammonium sulphate continued with the dialysis using cellophane. The dialysate was fractionated through gel filtration chromatography. The highest yield of protein was 1.109% from the seed (TS3), then 0.356% from the root (TR3), while the lowest was 0.014% from the fruit (TF2). The Lethal Concentration 50 (LC ) of proteins on brine shrimp lethality test was 50within range of 19 µg/ml - 25 µg/ml. The cytotoxicity test of the TR3 and TS3 proteins on cancer cell lines indicated that both of the proteins could inhibit proliferation of HeLa and K-562 cells with IC up to 45 µg/ml. It is recommended to conduct further researches on 50Trichosanthes plant as a herbal medicine to treat cancer

Aktivitas Sitotoksik Ekstrak Etanol Daun Aglaia elliptica Blume terhadap Galur Sel Kanker Serviks (HeLa)
Cytotoxicity tests on ethanol extract of Aglaia elliptica Blume leaves and its fractions on cervical cancer cell line (HeLa) were carried out. Dried powder of Aglaia elliptica leaves was extracted using ethanol andthen fractionated by n-hexane, ethyl acetate, and buthanol solvents. The cytotoxicity tests using MTT method (prolipheration inhibiton test) on HeLa cell lines were conducted for ethanol extract, n-hexana, ethyl acetate, and buthanol lractions.The results showed that the ethanol extract of Aglaia elliptica and its fractions have inhibition effect against HeLa cell line with IC50 of 282.44 ppm, 177.64 ppm, 16.03 ppm, 211.77 ppm, 181.33 ppm for ethanol extract, n-hexane, ethyl acetate, buthanol, and Water fractions respectively
Characterization of Trypsin-Like Protease of Lactobacillus plantarum FNCC 0270
Trypsin is an enzyme that has a unique mechanism of cutting peptide bonds specifically at the carboxyl side of lysine or arginine amino acids, with another amino acid. This study aims to analyze a trypsin-like protease (TLP) found in Lactobacillus plantarum FNCC 0270, by performing partial proteomic tests, i.e. MALDI-TOF/TOF, and standard bioinformatics tools. SDS-PAGE analysis showed 4 protein bands. Two bands of the (P1 and P2) showed molecular weights equivalent to 47.35 and 38.42 kD, each generating 8 and 11 peptide fragments respectively. According to information in www.ncbi.nlm.nih.gov/genbank/structures, the structure of serine protease HtrA (subs. plantarum L. plantarum ST-DT) consists of three domains. Using Clone Manager® software by aligning two sequences we obtained eleven. The Lactobacillus produces of the trypsin-like serine protease has 40-90% similarity. Using the Clustal W2 software we passed the 11 sequences through multiple alignments, and found that the isolate L. plantarum is closely related to L. buchneri, L. brevis, and L. malefermentans on the phylogenetic tree. Alignment analysis results showed that all 8 peptide fragments of band 1 and 11 peptide fragments of band 2, of the SDS-PAGE, were located in the active domain region of the fourth trypsin-like serine protease producing Lactobacilli
PENGARUH BAURAN PEMASARAN JASA DAN KUALITAS PELAYANAN KESEHATAN PUSKESMAS BOJONG NANGKA TERHADAP KEPUASAN PASIEN BPJS
Penelitian ini bertujuan untuk menganalisa bagaimana pengaruh bauran pemasaran jasa dalam hal ini produk (product), harga (price), promosi (promotion), lokasi (place), orang (people), bukti fisik / fasilitas fisik (physical evidence), dan proses (process), dan kualitas pelayanan kesehatan dalam hal ini bukti langsung (tangibles), kehandalan (reliability), daya tanggap (responsiveness), jaminan (assurance), dan empati (empathy) di Puskesmas Bojong Nangka Kecamatan Gunung Putri Kabupaten Bogor terhadap kepuasan pasien BPJS. Metode penelitian menggunakan analisis deskriptif metode potong lintang atau cross sectional. Tempat penelitian adalah di Puskesmas Bojong Nangka, Gunung Putri, Bogor. Sampel penelitian berjumlah 100 orang. Teknik pengumpulan data yaitu kuesioner dan observasi. Analisis data penelitian yaitu dengan uji Regresi Linier Berganda dengan hasil Y1 = 1,515 + 0,064X1 + 0,073X2. Kesimpulan penelitian adalah bahwa bauran pemasaran dan kualitas pelayanan kesehatan berpengaruh secara parsial maupun bersama-sama terhadap kepuasan pasien BPJS
