    No full text
    First evidence of the B 0 → J / ψ ω decay is found and the B s 0 → J / ψ η and B s 0 → J / ψ η ′ decays are studied using a dataset corresponding to an integrated luminosity of 1.0 fb -1 collected by the LHCb experiment in proton-proton collisions at a centre-of-mass energy of sqrt(s) = 7 TeV. The branching fractions of these decays are measured relative to that of the B 0 → J / ψ ρ 0 decay:frac(B (B 0 → J / ψ ω), B (B 0 → J / ψ ρ 0)) = 0.89 ± 0.19 (stat) - 0.13 + 0.07 (syst),frac(B (B s 0 → J / ψ η), B (B 0 → J / ψ ρ 0)) = 14.0 ± 1.2 (stat) - 1.5 + 1.1 (syst) - 1.0 + 1.1 (frac(f d, f s)),frac(B (B s 0 → J / ψ η ′), B (B 0 → J / ψ ρ 0)) = 12.7 ± 1.1 (stat) - 1.3 + 0.5 (syst) - 0.9 + 1.0 (frac(f d, f s)), where the last uncertainty is due to the knowledge of f d / f s, the ratio of b-quark hadronization factors that accounts for the different production rate of B 0 and B s 0 mesons. The ratio of the branching fractions of B s 0 → J / ψ η ′ and B s 0 → J / ψ η decays is measured to befrac(B (B s 0 → J / ψ η ′), B (B s 0 → J / ψ η)) = 0.90 ± 0.09 (stat) - 0.02 + 0.06 (syst)

    No full text
The B 0 s → J/ψK 0 S branching fraction is measured in a data sample corresponding to 0.41 fb−1 of integrated luminosity collected with the LHCb detector at the LHC. This channel is sensitive to the penguin contributions affecting the sin 2β measurement from B 0 → J/ψK 0 S . The time-integrated branching fraction is measured to be B(B 0 s → J/ψK 0 S ) = (1.83±0.28)×10−5 . This is the most precise measurement to date

Modeling neuronal consequences of autism-associated gene regulatory variants with human induced pluripotent stem cells

    No full text
Genetic factors contribute to the development of autism spectrum disorder (ASD), and although non-protein-coding regions of the genome are being increasingly implicated in ASD, the functional consequences of these variants remain largely uncharacterized. Induced pluripotent stem cells (iPSCs) enable the production of personalized neurons that are genetically matched to people with ASD and can therefore be used to directly test the effects of genomic variation on neuronal gene expression, synapse function, and connectivity. The combined use of human pluripotent stem cells with genome editing to introduce or correct specific variants has proved to be a powerful approach for exploring the functional consequences of ASD-associated variants in protein-coding genes and, more recently, long non-coding RNAs (lncRNAs). Here, we review recent studies that implicate lncRNAs, other non-coding mutations, and regulatory variants in ASD susceptibility. We also discuss experimental design considerations for using iPSCs and genome editing to study the role of the non-protein-coding genome in ASD

Rational identification of a colorectal cancer targeting peptide through phage display

    No full text
Colorectal cancer is frequently diagnosed at an advanced stage due to the absence of early clinical indicators. Hence, the identification of new targeting molecules is crucial for an early detection and development of targeted therapies. This study aimed to identify and characterize novel peptides specific for the colorectal cancer cell line RKO using a phage-displayed peptide library. After four rounds of selection plus a negative step with normal colorectal cells, CCD-841-CoN, there was an obvious phage enrichment that specifically bound to RKO cells. Cell-based enzyme-linked immunosorbent assay (ELISA) was performed to assess the most specific peptides leading to the selection of the peptide sequence CPKSNNGVC. Through fluorescence microscopy and cytometry, the synthetic peptide RKOpep was shown to specifically bind to RKO cells, as well as to other human colorectal cancer cells including Caco-2, HCT 116 and HCT-15, but not to the normal non-cancer cells. Moreover, it was shown that RKOpep specifically targeted human colorectal cancer cell tissues. A bioinformatics analysis suggested that the RKOpep targets the monocarboxylate transporter 1, which has been implicated in colorectal cancer progression and prognosis, proven through gene knockdown approaches and shown by immunocytochemistry co-localization studies. The peptide herein identified can be a potential candidate for targeted therapies for colorectal cancer.Portuguese Foundation for Science and Technology (FCT) under the scope of the strategic funding of UID/BIO/04469/2019 unit and BioTecNorte operation (NORTE-01-0145-FEDER-000004) funded by the European Regional Development Fund under the scope of Norte2020 - Programa Operacional Regional do Norte and the Project FCOMP-01–0124-FEDER-021053 (PTDC/SAU-BMA/121028/2010). Measurement of the ratio of prompt χ c to J / ψ production in pp collisions at √s = 7 TeV

    No full text
    The prompt production of charmonium χ c and J / ψ states is studied in proton-proton collisions at a centre-of-mass energy of √s = 7 TeV at the Large Hadron Collider. The χ c and J / ψ mesons are identified through their decays χ c → J / ψ γ and J / ψ → μ + μ - using 36 pb - 1 of data collected by the LHCb detector in 2010. The ratio of the prompt production cross-sections for χ c and J / ψ, σ (χ c → J / ψ γ) / σ (J / ψ), is determined as a function of the J / ψ transverse momentum in the range 2 < p T J / ψ < 15 GeV / c. The results are in excellent agreement with next-to-leading order non-relativistic expectations and show a significant discrepancy compared with the colour singlet model prediction at leading order, especially in the low p T J / ψ region

Measurement of the fragmentation fraction ratio fs/fd and its dependence on B meson kinematics

    No full text
The relative production rate of B0s and B 0 mesons is determined with the hadronic decays B0s→D−sπ+ and B 0 → D − K +. The measurement uses data corresponding to 1.0 fb−1 of pp collisions at a centre-of-mass energy of s√=7 TeV recorded in the forward region with the LHCb experiment. The ratio of production rates, f s /f d , is measured to be 0.238 ± 0.004 ± 0.015 ± 0.021, where the first uncertainty is statistical, the second systematic, and the third theoretical. This is combined with a previous LHCb measurement to obtain f s /f d  = 0.256 ± 0.020. The dependence of f s /f d on the transverse momentum and pseudorapidity of the B meson is determined using the decays B0s→D−sπ+ and B 0 → D −π+. There is evidence for a decrease with increasing transverse momentum, whereas the ratio remains constant as a function of pseudorapidity. In addition, the ratio of branching fractions of the decays B 0 → D − K + and B 0 → D −π+ is measured to be 0.0822 ± 0.0011 (stat) ± 0.0025 (syst)

Discovery of an optical counterpart to the hyperluminous X-ray source in ESO 243-49

    No full text
The existence of black holes of masses similar to 10(2)-10(5)M(circle dot) has important implications for the formation and evolution of star clusters and supermassive black holes. One of the strongest candidates to date is the hyperluminous X-ray source (HLX1), possibly located in the S0- a galaxy ESO 243-49, but the lack of an identifiable optical counterpart had hampered its interpretation. Using the Magellan telescope, we have discovered an unresolved optical source with R = 23.80 +/- 0.25 mag and V = 24.5 +/- 0.3 mag within HLX1's positional error circle. This implies an average X-ray/optical flux ratio similar to 500. Taking the same distance as ESO 243-49, we obtain an intrinsic brightness M-R = -11.0 +/- 0.3 mag, comparable to that of a massive globular cluster. Alternatively, the optical source is consistent with a main-sequence M star in the Galactic halo (for example an M4.4 star at approximate to 2.5 kpc). We also examined the properties of ESO 243-49 by combining Swift/Ultraviolet/Optical Telescope (UVOT) observations with stellar population modelling. We found that the overall emission is dominated by a similar to 5-Gyr-old stellar population, but the UV emission at approximate to 2000 angstrom is mostly due to ongoing star formation at a rate of similar to 0.03M(circle dot) yr(-1). The UV emission is more intense (at least a 9 sigma enhancement above the mean) north-east of the nucleus, in the same quadrant as HLX1. With the combined optical and X-ray measurements, we put constraints on the nature of HLX1. We rule out a foreground star and a background AGN. Two alternative scenarios are still viable. HLX1 could be an accreting intermediate mass black hole in a star cluster, which may itself be the stripped nucleus of a dwarf galaxy that passed through ESO 243-49, an event which might have caused the current episode of star formation. Or, it could be a neutron star in the Galactic halo, accreting from an M4-M5 donor star
