IDENTIFIKASI BAKTERI RONGGA MULUT PEROKOK DAN BUKAN PEROKOK DI PEKANBARU
Kebiasaan merokok merupakan salah satu penyebab timbulnya berbagai masalah kesehatan salah satunya penyakit rongga mulut. Kondisi kesehatan mulut yang menurun akibat rokok tersebut dapat menurunkan jumlah bakteri flora normal rongga mulut dan meningkatkan jumlah bakteri potensial pathogen. Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi jenis-jenis bakteri pada rongga mulut perokok dan bukan perokok di Pekanbaru. Penelitian ini dilakukan dengan isolasi bakteri pada medium MSA dan MacConkey, purifikasi, dan uji reaksi biokimia. Hasil penelitian menunjukkan bahwa jumlah isolat bakteri dari sampel rongga mulut perokok lebih tinggi (37 isolat), dibanding bukan perokok (26 isolat). Coagulase Negative Staphylococcus (27,03%), dan Aggregatibacter sp. (21,62%) lebih sering diisolasi dari rongga mulut perokok, sedangkan Coagulase Negative Staphylococcus (30,77%), Pseudomonas sp. (26,92%), dan Klebsiella sp. (19,23%) lebih sering diisolasi dari rongga mulut bukan perokok
PENYULUHAN PERILAKU HIDUP BERSIH DAN SEHAT (ORAL HYGIENE) DI KELURAHAN MUARA FAJAR PEKANBARU
Kesehatan gigi dan mulut dapat dicapai dengan menerapkan perilaku oral hygiene seperti menyikat gigi dengan baik dan benar serta menjaga kebersihan sikat gigi. Kegiatan PKM ini dilakukan dengan tujuan untuk meningkatkan pengetahuan masyarakat di Kelurahan Muara Fajar mengenai praktik oral hygiene yang benar dengan cara menyikat gigi dan pemeliharaan sikat gigi yang baik dan benar. Kegiatan dilakukan diawali dengan pre-test, pemberian penyuluhan oral hygiene, dan evaluasi kegiatan melalui Tanya jawab dan demostrasi cara menyikat gigi yang baik dan benar. Kegiatan ini diikuti oleh 43 peserta dan berdasarkan hasil pre-test diketahui bahwa hampir 80% peserta PKM belum menerapkan perilaku oral hygiene yang baik dan benar. Seluruh peserta mengikuti penyuluhan oral hygiene dengan antusias dan hasil evalusi diketahui bahwa seluruh peserta telah paham tentang perilaku oral hygiene. Peserta mampu melakukan praktik menyikat gigi dengan baik dan benar dan mengetahui cara menjaga kebersihan sikat gigi agar terhindar dari kontaminasi
Pengaruh Obat Kumur Berbasis Fluoride Dan Essential Oils Terhadap Jumlah Angka Kuman Rongga Mulut
Mouthwash is a solution or liquid that is used to clean the oral cavity with the aim of eliminating the bacteria causing tooth decay, working as a refresher and eliminating bad breath. The purpose of this study were to determine the differences of bacterial colony count in the oral cavity before and after using fluoride and essential oils based mouthwash, and to compare the effectiveness of both mouthwashes in reducing the number of oral bacteria. The sample for this study was mouth rinse collected from 10 students of Akademi Kesehatan John Paul II Pekanbaru, before and after using mouthwash. The samples were inoculated on Nutrient agar by pour plate method, and the bacterial colonies (CFU/mL) were counted after 2 days incubation.The results of this study showed that there was a difference of colony count between before and after the used of both mouthwashes. The mean of bacterial colony count after using fluoride-based mouthwash decreased by 314830.00 CFU/mL, while essential oils-based mouthwash reduced the bacterial colony count by 880220.00 CFU/mL. Dependent T-test showed that there were statistically significant reduction of bacterial colony count after the used of fluoride-based mouthwash (P=0.034) and essential oils-based mouthwash (P=0.008). Based on the Independent T-test there was no statistically significant difference (P>0.05) between fluoride and essential oils based mouthwash in reducing oral bacterial count
Isolasi dan Identifikasi Khamir Selulolitik Dari Tanah Rizosfer Anggrek Puser Bumi (Pecteilis susannae L.) di Hutan Wonosadi Gunung Kidul DIY
P. susannae (L.) Raf. is terrestrial orchid grown on the floor of Wonosadi forest. They belong to endangered species of orchid. Those orchids have grown well on degraded leaf litter and surround the grass. So, these plant were interesting to be elucidated especially for their nutrient availability. The was investigate the possibility of yeast associated with terrestrial orchids and cooperate in the provision of nutrients. The research objective was to obtain cellulolytic yeast on soil of root system P. susannae (L.) Raf. The research began with rhizosphere soil sampling on orchids root system P. susannae (L.) Raf. in two place in Forest Wonosadi. Soil samples were analyzed in the laboratory of microbiology, including the determination of the population of cellulolytic yeast and yeast isolated from soil of root system P. susannae (L.). directly from root system using Carboxy methyl cellulose medium (CMC), incubated for 5 days. Colonies were counted for the determination of population growth. Yeast colonies were grown separately isolated as a single culture. Sorting trough growth experiments using a form of cellulose (cellulose microcrystalline, leaf litter and paper) was different. Isolates were grown on microcrystalline cellulose selected for further testing based on the ability to use CMC. Activity was determined by the use of cellulose determine reducing sugar levels (glucose) was released using a spectrophotometer (OD 540 nm). The results showed that the rhizosphere soil P. susannae (L.) Raf. containing a yeast population (17x102 CFU and 2.3 x 102 CFU) and was dominated by the four isolates (K2, K16, A9 and A10) cellulolytic yeasts. The results of the identification of the four isolated of yeasts like Saccharomyces sp. (K2) and Candida sp. (K16, A9 and A10)
POLA KEPEKAAN ANTIBIOTIK ISOLAT BAKTERI ENTEROBACTERIACEAE KONTAMINAN PADA SIKAT GIGI
A toothbrush is a dental instrument used to maintain oral hygiene by removing bacteria on the surface of the teeth. Toothbrushes can be contaminated by Enterobacteriaceae from the bathroom environment and cause reinfection in the oral cavity. The purpose of this study was to determine the antibiotic susceptibility pattern of Enterobacteriaceae isolated from toothbrushes after 5 weeks of use. Bacteria were isolated on MacConkey agar and identified based on biochemical reaction tests. The antibiotic susceptibility test (Chloramphenicol, Gentamicin, Streptomycin, Ofloxacin, and Tetracycline) of the bacterial isolates was carried out by Kirby-Bauer disk diffusion method. Isolation of bacteria from toothbrushes on MC medium obtained Klebsiella sp, Escherichia coli, Serratia marcescens, and Enterobacter sp. The results of the antibiotic sensitivity test showed that Enterobacter sp. were sensitive to all of the antibiotics tested, while the other isolates had variations in the susceptibility pattern to the various antibiotics. Ofloxacin was the only tested antibiotic that can inhibit all bacterial isolates
Pengaruh Kualitas Pelayanan Terhadap Kepuasan nasabah PT Pegadaian (Persero) UPC Titi Kuning Medan
The aim of this research is to determine the effect of pawn service quality on customer satisfaction at PT. Pegadaian (Persero) UPC Titi Kuning Medan. The research object is limited to the service quality variable as the independent variable and customer satisfaction as the dependent variable. The sample taken was 86 respondents. The research method used by the author is the descriptive method, which is a method for researching the status of a group of people, an object, a set of conditions or a set of thoughts. The service quality variable is able to explain changes in customer satisfaction at PT. Pegadaian (Persero) UPC Titi Kuning Medan was 29.2% and the remaining 70.8% was explained by other factors not included in this research model. Based on the results of hypothesis testing with the t test, it was obtained that the t count variable for customer satisfaction with service quality was (5.879 > 1.988) that Ha was accepted and Ho was rejected. So it can be concluded that service quality has a significant effect on customer satisfaction. It is recommended to PT. Pegadaian (Persero) UPC Titi Kuning Medan to maintain and improve the quality of service by influencing, encouraging and mobilizing customers to come back to Pegadaian
Khamir Penghasil Indole-3-Acetic Acid dari Rhizosfer Anggrek Tanah Pecteilis susannae (L.) Rafin
Indole-3-acetic acid (IAA) is a growth hormone produced by plants. Several types of yeast has been reported to produce these hormones, and generally produce IAA in an higher amount than in plants. The purpose of this study is to get the yeast producing indole-3-acetic acid in the rhizosphere soil orchid Pecteilis susannae (L.) Rafin and to test the ability of yeast isolates to form IAA on medium L-tryptophan given as precursors. Yeasts were isolated and tested for their ability to produce IAA in vitro. The results showed that there are four potential yeast isolates that produce IAA which are isolates K2, U5, A3, and A1. Highest concentration of IAA was produced by A3 isolates grown on a medium containing GPB 200 ug / ml of L tryptophan in the amount of 56.262 g/ml.Keywords: Indole-3-acetic acid, rhizosfer yeast, Pecteilis susanna
Analisis Kajian Semantik Pada Antologi Puisi Di Bawah Payung Senja Kita Bercerita Karya Titi Sanaria Dan Lila Saraswaty
This study aims to analyze the denotative meaning, connotative meaning, referential and non-referential meanings contained in the poetry anthology Below Umbrella Senja Kita Tells Stories by Titi Sanaria and Lila Saraswati. Problems in This research is (1) How is the denotative meaning contained in the anthology of Di . poetry? Under Umbrella Senja We Tell Stories by Titi Sanaria and Lila Saraswaty? (2) How the connotative meaning contained in the anthology of poetry Under the Umbrella of Senja Kita Tells By Titi Sanaria and Lila Saraswaty? (3) What is the referential meaning contained in the poetry anthology Under Umbrella Senja Kita Tells Stories by Titi Sanaria and Lila Saraswati? (4) What is the non-referential meaning contained in Di .'s poetry anthology? Under Umbrella Senja We Tell Stories by Titi Sanaria and Lila Saraswaty?. Data This research is in the form of words and phrases that show the expression of referring words. The theory used in this research is Chaer (2013), Pateda (2010), Rokhmansyah (2014), Guntur (2009) and Daniel (2004). Sources of data in this study are 25 titles of poetry contained in the poetry anthology Under the Payung Senja Kita Storytelling by Titi Sanaria and Lila Saraswaty. The methodology used in This research is a method of content analysis, the type of research literature review and approach qualitative. The data collection techniques used in this study were (1) read, (2) note and (3) listen. The results showed that the denotative meaning of the poem rarely used because basically a poem uses a connotative meaning or indirect meaning to beautify the meaning contained in the poem, meaning connotative is often used because it has a connotative meaning verse by stanza every time poetry becomes more beautiful, referential meaning is used more often because in In this anthology of poetry, the author or writer pays more attention to objects and circumstances around him to make it an object of poetry, non-referential meaning is widely used because This non-referential meaning refers to the conjunction used for linking words or clauses so that verse by stanza in the poem becomes more effective. So, these four meanings are very useful for making verse after verse of the poem become more complete and perfect
KHAMIR PENGHASIL INDOLE-3-ACETIC ACID DARI RHIZOSFER ANGGREK TANAH Pecteilis susannae (L.) Rafin
The objective of this study was to obtain the yeasts capable of producing
indole-3-acetic acid isolated from rhizosphere soil of terrestrial orchid Pecteilis
susannae from several location in Wonosadi Forest Gunung Kidul and to investigate
the capability of yeast isolates to produce IAA on medium supplemented with Ltryptophan
as precursor. The capability of P. susannae orchid to grow in nature was
assisted by phytohormone IAA derived from plant itself or environment. Some of
rhizosphere soil yeast species estimated to be able to produce phytohormone IAA.
This research commenced with the isolation of yeasts from rhizosphere soil of
orchid P. susannae using Yeast Malt Agar (YMA) medium. Yeast isolates were
selected based on growth experiment on Glucose Peptone Broth (GPB) supplemented
with 200 µg/ml L-tryptophan. The IAA concentration was determined
colorimetrically on spectrofotometer (OD530 nm) with Salkowsky�s reagent added to
supernatant of yeast culture. The ability of selected yeasts to produce IAA was
elucidated through IAA production experiment on Glucose Peptone Broth (GPB) and
Salt Basal Medium (SBM) supplemented with and without 200 µg/ml L-tryptophan.
Biomass (OD600 nm) and IAA production (OD530 nm) measured every 24 hours for 4
days. IAA produce in culture of selected yeast isolates was extracted and identified
through Thin-Layer Chromatography (TLC) by spotted the culture extract on silica
gel covered-plate. Selected isolates of IAA producing yeasts were identified using
standard methods for yeast.
The results revealed that 29 isolates of yeast had been successfully isolated
from rhizosphere of terrestrial orchid P.susannae (L.) Rafin, only four isolates (K2,
U5, A3 and A1) showed their capability to grow and produce high concentration of
IAA on GPB and SBM. Upon addition of 200 µg/ml L-tryptophan, selected yeast
isolates (K2, U5, A3 and A1) produce 31.191, 51.524, 56.262, and 13.286 µg IAA/ml
respectively in PGB medium. Otherwise, IAA production in SBM medium were
2.789, 5.095, 2.619, and 2.619 respectively. The IAA identification on silica gel-plate
showed that U5 and A3 isolates produce compound with the same Rf value compared
to IAA standard. The fourth selected yeast isolates presumably as strain
Saccharomyces sp.1 (K2), Saccharomyces sp.2 (U5), Cryptococcus sp.1 (A3), and
Candida sp.1 (A1)
