    Human gingival mesenchymal stem cells (hGMSCs) and endothelial committed hGMSCs (e-hGMSCs) have considerable potential to serve as an in vitro model to replicate the inflammation sustained by Porphyromonas gingivalis in periodontal and cardiovascular diseases. The present study aimed to investigate the effect of ascorbic acid (AA) on the inflammatory reverting action of lipopolysaccharide (LPS-G) on the cell metabolic activity, inflammation pathway and reactive oxygen species (ROS) generation in hGMSCs and e-hGMSCs. Cells were treated with LPS-G (5 μg mL−1 ) or AA (50 μg mL−1 ) and analyzed by 3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide (MTT) assay, immunofluorescence and Western blot methods. The rate of cell metabolic activity was decreased significantly in LPS-G-treated groups, while groups co-treated with LPS-G and AA showed a logarithmic cell metabolic activity rate similar to untreated cells. AA treatment attenuated the inflammatory effect of LPS-G by reducing the expression of TLR4/MyD88/NFκB/NLRP3/Caspase-1/IL-1β, as demonstrated by Western blot analysis and immunofluorescence acquisition. LPS-G-induced cells displayed an increase in ROS production, while AA co-treated cells showed a protective effect. In summary, our work suggests that AA attenuated LPS-G-mediated inflammation and ROS generation in hGMSCs and e-hGMSCs via suppressing the NFκB/Caspase-1/IL-1β pathway. These findings indicate that AA may be considered as a potential factor involved in the modulation of the inflammatory pathway triggered by LPS-G in an vitro cellular model

    We adopt a combination of analytical and numerical methods to study the renormalization group flow of the most general field theory with quartic interaction in d=4-ε with N=3 and N=4 scalars. For N=3, we find that it admits only three nondecomposable critical points: The Wilson-Fisher with O(3) symmetry, the cubic with H3=(Z2)3â ŠS3 symmetry, and the biconical with O(2)×Z2. For N=4, our analysis reveals the existence of new nontrivial solutions with discrete symmetries and with up to three distinct field anomalous dimensions

    A Nutrition mathematical model to account for dietary supply and requirements of energy and other nutrients for domesticated small ruminants: the development and evaluation of the Small Ruminant Nutrition System

    A mechanistic model that predicts nutrient requirements and biological values of feeds for sheep (Cornell Net Carbohydrate and Protein System; CNCPS-S) was expanded to include goats and the name was changed to the Small Ruminant Nutrition System (SRNS). The SRNS uses animal and environmental factors to predict metabolisable energy (ME) and protein, and Ca and P requirements. Requirements for goats in the SRNS are predicted based on the equations developed for CNCPS-S, modified to account for specific requirements of goats, including maintenance, lactation, and pregnancy requirements, and body reserves. Feed biological values are predicted based on carbohydrate and protein fractions and their ruminal fermentation rates, forage, concentrate and liquid passage rates, and microbial growth. For sheep, the SRNS accurately predicted gains and losses of shrunk body weight (SBW) of adult sheep (n = 15; mean bias (MB) = 5.8 g/d and root mean square error of prediction (RMSPE) = 30 g/d; and r2 = 0.73) when diets were not deficient in ruminal nitrogen. Several evaluations indicated the SRNS had MB varying from 2.4 to 18 g/d, RMSPE varying from 21.4 to 41 g/d, and r2 varying from 0.70 to 0.84 when predicting average daily gain (ADG) of growing lambs. For goats, the evaluation for lactating does indicated that predictions of ME intake (n = 21; MB = 0.04 Mcal/d; RMSEP = 0.24 Mcal/d; and r2 = 0.99) and energy balance (n = 21; MB = 0.075 Mcal/d; RMSEP = 0.20 Mcal/d; and r2 = 0.87) were adequate. Similarly, the SRNS accurately predicted ADG of kids (n = 31; MB = −6.4 g/d; RMSEP = 32.5 g/d; and r2 = 0.85). In conclusion, the SRNS can accurately predict, ADG of growing lambs, and changes in SBW of mature sheep and ME intake and the energy balance of lactating and non-lactating adult goats and the ADG of kids of dairy, meat, and indigenous breeds.     Search for the decay Bs0→D*∓π±

    A search for the decay Bs0→D*∓π± is presented using a data sample corresponding to an integrated luminosity of 1.0  fb-1 of pp collisions collected by LHCb. This decay is expected to be mediated by a W-exchange diagram, with little contribution from rescattering processes, and therefore a measurement of the branching fraction will help us to understand the mechanism behind related decays such as Bs0→π+π- and Bs0→DD̅ . Systematic uncertainties are minimized by using B0→D*∓π± as a normalization channel. We find no evidence for a signal, and set an upper limit on the branching fraction of B(Bs0→D*∓π±)&#60;6.1(7.8)×10-6 at 90% (95%) confidence level

    CENP-G in neocentromeres and inactive centromeres

    CENP-G is a novel constitutive centromere-specific protein localized to the kinetochore inner plate and subjacent region. It has been identified as associating specifically with the α-1 subfamily of α-satellite DNA. In the present work, the localization of CENP-G was compared with that of other CENPs by immunofluorescence and fluorescence in situ hybridization. Studies were carried out on four abnormal human centromeres: two neocentromeres and two inactive centromeres. CENP-G was detected in one of the two inactive centromeres but not in the other that shows a partial deletion of the alphoid DNA. Interestingly, CENP-G is also present in neocentromeres, which lack alphoid DNA sequences, and in the human Y chromosome, which lacks the α-1 type of satellite DNA. These data provide further evidence that CENP-G may be an essential factor in cen- tromeric function and that in centromeres lacking the α-1 subfamily of alphoid DNA, other DNA sequences are able to bind CENP-G
