Abschließend wird im Licht von drei konkurrierenden philosophischen Modellen der Frage nachgegangen, was es überhaupt erlaubt, einen bestimmten Charakterzug als Tugend zu bestimmen

    Genetic association study of QT interval highlights role for calcium signaling pathways in myocardial repolarization.

    The QT interval, an electrocardiographic measure reflecting myocardial repolarization, is a heritable trait. QT prolongation is a risk factor for ventricular arrhythmias and sudden cardiac death (SCD) and could indicate the presence of the potentially lethal mendelian long-QT syndrome (LQTS). Using a genome-wide association and replication study in up to 100,000 individuals, we identified 35 common variant loci associated with QT interval that collectively explain ∼8-10% of QT-interval variation and highlight the importance of calcium regulation in myocardial repolarization. Rare variant analysis of 6 new QT interval-associated loci in 298 unrelated probands with LQTS identified coding variants not found in controls but of uncertain causality and therefore requiring validation. Several newly identified loci encode proteins that physically interact with other recognized repolarization proteins. Our integration of common variant association, expression and orthogonal protein-protein interaction screens provides new insights into cardiac electrophysiology and identifies new candidate genes for ventricular arrhythmias, LQTS and SCD

    Ordoliberalism and the evolution of norms

    The first part of the following paper deals with varying points of criticism forwarded against Ordoliberalism. Here, it is not the aim to directly falsify each argument on its own; rather, the author tries to give a precise overview of the spectrum of critique. The second section picks out one argument of critical review – namely that the ordoliberal concept of the state is somewhat elitist and grounded on intellectual experts. Based on the previous sections, the final part differentiates two kinds of genesis of norms: an evolutionary and an elitist one – both (latently) present within Ordoliberalism. In combination with the two-level differentiation between individual and regulatory ethics, the essay allows for a distinction between individual-ethical norms based on an evolutionary genesis of norms and regulatory-ethical norms based on an elitist understanding of norms. A by-product of the author’s argument is a (further) demarcation within neoliberalism

    Implementation of Web Query Languages Reconsidered

    Visions of the next generation Web such as the "Semantic Web" or the "Web 2.0" have triggered the emergence of a multitude of data formats. These formats have different characteristics as far as the shape of data is concerned (for example tree- vs. graph-shaped). They are accompanied by a puzzlingly large number of query languages each limited to one data format. Thus, a key feature of the Web, namely to make it possible to access anything published by anyone, is compromised. This thesis is devoted to versatile query languages capable of accessing data in a variety of Web formats. The issue is addressed from three angles: language design, common, yet uniform semantics, and common, yet uniform evaluation. % Thus it is divided in three parts: First, we consider the query language Xcerpt as an example of the advocated class of versatile Web query languages. Using this concrete exemplar allows us to clarify and discuss the vision of versatility in detail. Second, a number of query languages, XPath, XQuery, SPARQL, and Xcerpt, are translated into a common intermediary language, CIQLog. This language has a purely logical semantics, which makes it easily amenable to optimizations. As a side effect, this provides the, to the best of our knowledge, first logical semantics for XQuery and SPARQL. It is a very useful tool for understanding the commonalities and differences of the considered languages. Third, the intermediate logical language is translated into a query algebra, CIQCAG. The core feature of CIQCAG is that it scales from tree- to graph-shaped data and queries without efficiency losses when tree-data and -queries are considered: it is shown that, in these cases, optimal complexities are achieved. CIQCAG is also shown to evaluate each of the aforementioned query languages with a complexity at least as good as the best known evaluation methods so far. For example, navigational XPath is evaluated with space complexity O(q d) and time complexity O(q n) where q is the query size, n the data size, and d the depth of the (tree-shaped) data. CIQCAG is further shown to provide linear time and space evaluation of tree-shaped queries for a larger class of graph-shaped data than any method previously proposed. This larger class of graph-shaped data, called continuous-image graphs, short CIGs, is introduced for the first time in this thesis. A (directed) graph is a CIG if its nodes can be totally ordered in such a manner that, for this order, the children of any node form a continuous interval. CIQCAG achieves these properties by employing a novel data structure, called sequence map, that allows an efficient evaluation of tree-shaped queries, or of tree-shaped cores of graph-shaped queries on any graph-shaped data. While being ideally suited to trees and CIGs, the data structure gracefully degrades to unrestricted graphs. It yields a remarkably efficient evaluation on graph-shaped data that only a few edges prevent from being trees or CIGs

The definitive version was published in Applied and Environmental Microbiology 82 (2016): 4505-4516, doi:10.1128/AEM.00772-16.A new clade of archaea has recently been proposed to constitute the seventh methanogenic order, the Methanomassiliicoccales, which is related to the Thermoplasmatales and the uncultivated archaeal clades deep-sea hydrothermal vent Euryarchaeota group 2 and marine group II Euryarchaeota but only distantly related to other methanogens. In this study, we investigated the membrane lipid composition of Methanomassiliicoccus luminyensis, the sole cultured representative of this seventh order. The lipid inventory of M. luminyensis comprises a unique assemblage of novel lipids as well as lipids otherwise typical for thermophilic, methanogenic, or halophilic archaea. For instance, glycerol sesterpanyl-phytanyl diether core lipids found mainly in halophilic archaea were detected, and so were compounds bearing either heptose or methoxylated glycosidic head groups, neither of which have been reported so far for other archaea. The absence of quinones or methanophenazines is consistent with a biochemistry of methanogenesis different from that of the methanophenazine-containing methylotrophic methanogens. The most distinctive characteristic of the membrane lipid composition of M. luminyensis, however, is the presence of tetraether lipids in which one glycerol backbone is replaced by either butane- or pentanetriol, i.e., lipids recently discovered in marine sediments. Butanetriol dibiphytanyl glycerol tetraether (BDGT) constitutes the most abundant core lipid type (>50% relative abundance) in M. luminyensis. We have thus identified a source for these unusual orphan lipids.     Experimental validation of analytical wake and downstream turbine performance modelling

    Wake effects in wind farms can cause significant power losses. In order to reduce these losses layout and control optimization can be applied. For this purpose, simple and fast prediction tools for the wake flow are needed. In the first part of this work, five analytical wind turbine wake models are compared to small-scale turbine wind tunnel measurements. The measurements are conducted at several downstream distances, varying the ambient turbulence intensity and upstream turbine blade pitch angle. Furthermore, an adjustment of a recently developed wake model is proposed. Subsequently, the adjusted model is found to perform best throughout all test cases. In the second part, the performance of an aligned downstream turbine is modelled based on the predicted wake flow using a Blade Element Momentum method with guaranteed convergence. In order to consider the non-uniform inflow velocity a mean-blade-element-velocity method is developed. Moreover, a blockage effect correction is applied. A comparison to wind tunnel measurement data shows that the wake velocity as well as the combined power of two aligned turbines are fairly well predicted.
