    Tato práce pojednává o životě, díle a osobnosti amerického skladatele, saxofonisty a flétnisty Henryho Threadgilla a společensko-kulturním kontextu, z něhož vychází. Threadgill a specifická generace amerických hudebníků, s kterou v šedesátých a sedmdesátých letech vyrůstal, představují důležitou a opomíjenou součást jazzové a hudební historie. V porovnání s USA, kde je dnes Henry Threadgill považován za stěžejního skladatele, je v českém a evropském hudebním prostředí známý pouze okrajově. Cílem práce je přiblížit Threadgillovo umělecké myšlení, hudební systém, vybrané stěžejní dílo a nejznámější ansámbly české veřejnosti. Práce obsahuje transkripce kompozic Henryho Threadgilla a odkazuje na hudební nahrávky.This thesis looks into the life, work and personality of the American composer, saxophonist and flutist Henry Threadgill and the sociocultural context which it is based on. Threadgill and especially the specific generation of musicians that he is part of, represents a significant and neglected part of jazz and music history. Compared to the USA, where Henry Threadgill is today considered a key composer, he is only marginally known in the Czech and European musical enviroment. The aim of the work is to bring his artistic personality, music system, selected work and the best known ensembles closer to the Czech audience. The work includes transcriptions of Henry Threadgill’s compositions and refers to music recordings

    Evidence for the decay B0→J/ψω and measurement of the relative branching fractions of meson decays to J/ψη and J/ψη′

    First evidence of the B 0 → J / ψ ω decay is found and the B s 0 → J / ψ η and B s 0 → J / ψ η ′ decays are studied using a dataset corresponding to an integrated luminosity of 1.0 fb -1 collected by the LHCb experiment in proton-proton collisions at a centre-of-mass energy of sqrt(s) = 7 TeV. The branching fractions of these decays are measured relative to that of the B 0 → J / ψ ρ 0 decay:frac(B (B 0 → J / ψ ω), B (B 0 → J / ψ ρ 0)) = 0.89 ± 0.19 (stat) - 0.13 + 0.07 (syst),frac(B (B s 0 → J / ψ η), B (B 0 → J / ψ ρ 0)) = 14.0 ± 1.2 (stat) - 1.5 + 1.1 (syst) - 1.0 + 1.1 (frac(f d, f s)),frac(B (B s 0 → J / ψ η ′), B (B 0 → J / ψ ρ 0)) = 12.7 ± 1.1 (stat) - 1.3 + 0.5 (syst) - 0.9 + 1.0 (frac(f d, f s)), where the last uncertainty is due to the knowledge of f d / f s, the ratio of b-quark hadronization factors that accounts for the different production rate of B 0 and B s 0 mesons. The ratio of the branching fractions of B s 0 → J / ψ η ′ and B s 0 → J / ψ η decays is measured to befrac(B (B s 0 → J / ψ η ′), B (B s 0 → J / ψ η)) = 0.90 ± 0.09 (stat) - 0.02 + 0.06 (syst)

    CLN3 is a transmembrane protein with a predominant localization in lysosomes in non-neuronal cells but is also found in endosomes and the synaptic region in neuronal cells. Mutations in the CLN3 gene result in juvenile neuronal ceroid lipofuscinosis or Batten disease, which currently is the most common cause of childhood dementia. We have recently reported that the lysosomal targeting of CLN3 is facilitated by two targeting motifs: a dileucine-type motif in a cytoplasmic loop domain and an unusual motif in the carboxyl-terminal cytoplasmic tail comprising a methionine and a glycine separated by nine amino acids (Kyttala, A., Ihrke, G., Vesa, J., Schell, M. J., and Luzio, J. P. (2004) Mol. Biol. Cell 15, 1313-1323). In the present study, we investigated the pathways and mechanisms of CLN3 sorting using biochemical binding assays and immunofluorescence methods. The dileucine motif of CLN3 bound both AP-1 and AP-3 in vitro, and expression of mutated CLN3 in AP-1- or AP-3-deficient mouse fibroblasts showed that both adaptor complexes are required for sequential sorting of CLN3 via this motif. Our data indicate the involvement of complex sorting machinery in the trafficking of CLN3 and emphasize the diversity of parallel and sequential sorting pathways in the trafficking of membrane proteins

    Measurement of the time-dependent CP asymmetry in B0 -> J/ψ KS0 decays

    This Letter reports a measurement of the CP violation observables SJ/ψK0S and CJ/ψK0S in the decay channel B0→J/ψK0S performed with 1.0 fb−1 of pp collisions at s√=7 TeV collected by the LHCb experiment. The fit to the data yields SJ/ψK0S=0.73±0.07(stat)±0.04(syst) and CJ/ψK0S=0.03±0.09(stat)±0.01(syst). Both values are consistent with the current world averages and within expectations from the Standard Model

    Measurement of the ratio of prompt χ c to J / ψ production in pp collisions at √s = 7 TeV

    The prompt production of charmonium χ c and J / ψ states is studied in proton-proton collisions at a centre-of-mass energy of √s = 7 TeV at the Large Hadron Collider. The χ c and J / ψ mesons are identified through their decays χ c → J / ψ γ and J / ψ → μ + μ - using 36 pb - 1 of data collected by the LHCb detector in 2010. The ratio of the prompt production cross-sections for χ c and J / ψ, σ (χ c → J / ψ γ) / σ (J / ψ), is determined as a function of the J / ψ transverse momentum in the range 2 < p T J / ψ < 15 GeV / c. The results are in excellent agreement with next-to-leading order non-relativistic expectations and show a significant discrepancy compared with the colour singlet model prediction at leading order, especially in the low p T J / ψ region

    Prevention Of Spontaneous Tumors By Pharmacological Inhibition Of The Epidermal Growth Factor Receptor

    The epidermal growth factor receptor (EGFR) is a ligand-activated tyrosine kinase receptor that is often overexpressed in cancers and plays a significant role in cell proliferation and inhibition of apoptosis. Mouse model studies have demonstrated the ability of EGFR inhibition to hinder spontaneous colorectal cancer occurrence by 50-80% when EGFR inhibition is induced before the establishment of tumors (Rinella, ES and Threadgill, DW doi 10.1371/journal.pone.0039552). The success rates reported in human clinical trials differ from mouse model studies, with a 15% success rate among colorectal cancer patients receiving anti-EGFR treatment (Pritchart CC, and Grady WM doi 10.1136/gut.2009.206250). Timing of treatment is the major difference between animal/mouse trials and human therapeutic treatment of end-stage cancers. In our study, it was hypothesized that earlier preventative doses of the AG1478 chemotherapy inhibitor would reduce the frequency of spontaneous tumor occurrence in a preventative, rather than therapeutic, manner in a murine population. This current study provides additional specificity and histopathological analysis of the spontaneous tumor occurrence in a large murine sample of four genetically distinct wild-type strain mice to better simulate a variable human population. Such specificity includes, but is not limited to, the number, type and severity of the tumors that did occur and whether the concentration of the drug is correlated to a reduction in tumor occurrence. Such information will broaden the discussion of EGFR’s role in tumor treatment
