Sim-xl: A Powerful And User-friendly Tool For Peptide Cross-linking Analysis.
Chemical cross-linking has emerged as a powerful approach for the structural characterization of proteins and protein complexes. However, the correct identification of covalently linked (cross-linked or XL) peptides analyzed by tandem mass spectrometry is still an open challenge. Here we present SIM-XL, a software tool that can analyze data generated through commonly used cross-linkers (e.g., BS3/DSS). Our software introduces a new paradigm for search-space reduction, which ultimately accounts for its increase in speed and sensitivity. Moreover, our search engine is the first to capitalize on reporter ions for selecting tandem mass spectra derived from cross-linked peptides. It also makes available a 2D interaction map and a spectrum-annotation tool unmatched by any of its kind. We show SIM-XL to be more sensitive and faster than a competing tool when analyzing a data set obtained from the human HSP90. The software is freely available for academic use at http://patternlabforproteomics.org/sim-xl. A video demonstrating the tool is available at http://patternlabforproteomics.org/sim-xl/video. ANALISIS PENGARUH KUALITAS PRODUK, KUALITAS PELAYANAN DAN HARGA TERHADAP KEPUASAN KONSUMEN (STUDI KASUS PADA KONSUMEN SIM CARD GSM PRABAYAR XL DI KOTA YOGYAKARTA)
AbstrakPenelitian ini bertujuan untuk mengetahui: pengaruh kualitas produk, kualitas pelayanan, dan harga secara parsial dan simultan terhadap kepuasan konsumen Sim Card GSM Prabayar XL Yogyakarta. Jenis penelitian yang digunakan adalah survei. Populasi pada penelitian ini adalah seluruh konsumen Sim Card GSM Prabayar XL di kota Yogyakarta. Teknik pengambilan sampel menggunakan metode purposive sampling dengan jumlah sampel sebanyak 200 orang. Teknik pengumpulan data menggunakan kuesioner yang telah diuji validitas dan reliabilitasnya. Teknik analisis data adalah regresi berganda. Hasil penelitian ini menunjukkan bahwa: (1) Terdapat pengaruh positif kualitas produk terhadap kepuasan konsumen Sim Card GSM Prabayar XL Yogyakarta, (2) Terdapat pengaruh positif kualitas pelayanan terhadap kepuasan konsumen Sim Card GSM Prabayar XL Yogyakarta, (3) Terdapat pengaruh positif harga terhadap kepuasan konsumen Sim Card GSM Prabayar XL Yogyakarta, dan (4) Terdapat pengaruh kualitas produk, kualitas pelayanan, dan harga secara simultan terhadap kepuasan konsumen Sim Card GSM Prabayar XL Yogyakarta. reliabilitasnya. Teknik analisis data yang digunakan untuk menjawab hipotesis
adalah regresi berganda.
Hasil penelitian ini menunjukkan bahwa: (1) Terdapat pengaruh positif
kualitas produk terhadap kepuasan konsumen Sim Card GSM Prabayar XL
Yogyakarta, dibuktikan dari nilai t hitung sebesar 4,404 dengan nilai signifikansi
0,000<0,05; dan koefisien regresi sebesar 0,201, (2) Terdapat pengaruh positif
kualitas pelayanan terhadap kepuasan konsumen Sim Card GSM Prabayar XL
Yogyakarta, dibuktikan dari nilai t hitung sebesar 5,494 dengan nilai signifikansi
sebesar 0,000<0,05 dan koefisien regresi sebesar 0,245, (3) Terdapat pengaruh
positif harga terhadap kepuasan konsumen Sim Card GSM Prabayar XL
Yogyakarta. Hal ini dibuktikan bahwa hasil penelitian variabel brand trust
diperoleh nilai t hitung sebesar 5,074 dengan nilai signifikansi sebesar 0,000
(0,000<0,05) dan koefisien regresi mempunyai nilai positif sebesar 0,280, dan (4)

ANALISIS PENGARUH HARGA, KUALITAS, DAN PROMOSI TERHADAP KEPUTUSAN PEMBELIAN SIM CARD XL (Studi Kasus Pada Siswa SMA Negeri 3 Ponorogo
RINGKASAN
Penelitian ini merupakan studi kasus pada siswa di SMA Negeri 3 Ponorogo, dengan judul penelitian “Analisis Pengaruh Harga, Kualitas, dan Promosi terhadap Keputusan Pembelian Sim Card XL (Studi Kasus Pada Siswa SMA Negeri 3 Ponorogo). Tujuan dari penelitian ini adalah untuk mengetahui pengaruh pengaruh harga, kualitas, dan promosi terhadap perilaku konsumen dalam membeli Sim Card XL, dan untuk mengetahui faktor yang memberikan pengaruh paling dominan terhadap perilaku konsumen dalam membeli Sim Card XL
Dalam membuat perhitungan metode analisa yang digunakan untuk membuktikan hipotesis adalah teknik pengolahan data yang berbentuk angka yang diperoleh dari angket siswa, maka penulis menggunakan analisa yang terdiri antara lain: Uji Validitas dan Reliabilitas data, Analisa Regresi Linear Berganda, Analisa Korelasi Determinasi, Uji t hitung,dan, Uji F-tes (Uji F).
Berdasarkan analisis hasil perhitungan didapat hasil pengujian dengan menggunakan F test diperoleh hasil sebesar 9,33 di mana dari data F tabel diketahui nilai F tabel sebesar 2,72 hipotesis nol (Ho) ditolak atau hipotesis alternatif diterima. Maka dapat dikatakan bahwa variabel independent (variabel harga, kualitas, promosi) secara bersama mempunyai pengaruh yang signifikan terhadap variabel dependent. Variabel Promosi (X3) diperoleh hasil 2,637. Diketahui besarnya nilai t tabel adalah 2,000 terbukti T test > T tabel yaitu 2,637 > 2,000 sehingga Ho ditolak dan hipotesis penelitian diterima, artinya promosi mempunyai pengaruh yang signifikan terhadap keputusan pembelian untuk menggunakan Sim Card XL
Berdasarkan kesimpulan yang diperoleh dalam penelitian ini, maka diajukan saran (1) Untuk lebih meningkatkan loyalitas konsumen terhadap produk Sim Card XL,(2) agar dipertahankan dan lebih ditingkatkan kulitas sinyal, (3) promosi tidak membodohi atau membohongi konsumen.
Insight into the apoptosis mediated by feline herpesvirus (FeHV-1) on permissive cells
1.Introduction:
Apoptosis, also known as programmed cell death, is a critical component of host defense mechanisms against viral infection, limiting viral replication and transmission. Multiple signaling pathways, both intrinsic and extrinsic, form a complex network to modulate apoptosis, even though viruses have distinct mechanisms for mediating or inhibiting apoptosis to avoid this process. In comparison to other herpesvirus members, little is known about the relationship between FeHV-1, the etiological agent of feline rhinotracheitis, and apoptosis. This study aimed to learn more about this connection by evaluating the impact of apoptosis on FeHV-1 infection of permissive cells.
2.Materials and methods:
Confluent monolayers of Crandell-Rees Feline Kidney Cell (CRFK) were infected using a MOI 1 at different time points. The MTT (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide) assay was used to determine cell viability. Moreover, the apoptotic pathway was investigated using two different approaches: a flow cytometry analysis using an Annexin V-FITC detection kit and a western blot assay targeting specific markers of apoptosis (Caspase 8, Caspase 9, Caspase 3, Bcl-XL, Bcl-2).
3.Results:
Our results suggested that FeHV-1 triggers apoptosis process starting with 12 hours post infection. A larger percentage of early apoptotic cells were detected by the flow cytometry analysis, starting at 12 hours after infection, while late apoptotic cells increased at 24 hours after infection. The western blot analysis showed the cleavage of caspase 9 after 24 hours and a time-dependent cleavage of caspase 3 after 48 and 72 hours. We also observed a reduction of the expression of the anti-apoptotic proteins Bcl-XL and Bcl-2, beginning from 24 hours, also in this case.
4.Discussion and conclusions:
In this work we investigated on apoptosis modulation made by FeHV-1. Similar to other herpesviruses, such as Herpes simplex virus (HSV-1), Bovine herpesvirus (BoHV-1), Pseudorabies virus (PRV), Anatid alphaherpesvirus-1 (DEV), and Caprine Herpesvirus 1, infection on permissive cells results in the activation of the apoptotic process, which reduces cell viability. The induction of the intrinsic pathway and the caspase 9 cleavage suggested a dysfunction of the mitochondrial membrane, and the downregulation of Bcl-2 and Bcl-XL expression supported this hypothesis.
Even though additional research is needed to fully understand the viral mechanism and the viral genes/proteins involved in this process, this study provides new knowledge on the host-FeHV-1 interaction. The potential proviral effect may be examined since it has been shown that some viruses manipulate the cellular apoptotic mechanism to enhance their own dissemination within apoptotic bodies
Metode Simple Additive Weighting (SAW) Dalam Pemilihan Sim Card Provider
With the development of technology at this time Sim Card Provider is needed to support community activities. In Indonesia alone many Sim Card Provider many services and also ease of accessing the internet, sonsumers are required to see which Sim Card Provider suits their needs. To overcome the problem of selecting the Sim Card Provider, a Decision Support System will be made using the Simple Additive Weighting (SAW) method using data taken from questionnaires distributed to Sim Card Provider users, which is expected to help in the selection of the appropriate Sim Card Provider with Consumer needs. The author uses six criteria, namely Signal, Price, Internet Package, Internet Connection, Phone Bonus, SMS Bonus. By using five alternatives namely Simpati, XL, Indosat, Smartfren, Three (3). The final result of this study indicated that alternative 5 or Three (3) has the biggest result of 3,245 and alternative
3 namely Indosat has the lowest result among the alternatives which is 2,625.

Abstrak
Dengan berkembangnya teknologi pada saatini Sim Card Provider sangat dibutuhkan untuk menunjang aktivitas masyarakat. Di Indonesia sendiri banyak Sim Card Provider banyak menyediakan layanan dan juga kemudahan dalam mengakses internet, konsumen dituntut untuk melihat manakah Sim Card Provider yang sesuai dengan kebutuhan. Untuk mengatasi masalah pemilihan Sim Card Provider tersebut maka akan dibuat Sistem Pendukung Keputusan dengan menggunakan Metode Simple Additive Weighting (SAW) dengan menggunakan data yang diambil dari kuesioner yang dibagikan kepada para pengguna Sim Card Provider, yang diharapkan dapat membantu dalam pemilihan Sim Card Provider yang sesuai dengan kebutuhan Konsumen. Penulis menggunakan enam kriteria yaitu Sinyal, Harga, Paket Internet, Koneksi Internet, Bonus Telpon, Bonus Sms. Dengan menggunakan lima alternatif yaitu Simpati, XL, Indosat, Smartfren, Three (3). Hasil akhir dari penelitian ini menunjukan bahwa alternatif 5 atau Three (3) memiliki hasil terbesar yaitu 3,245 dan alternatif 3 yaitu Indosat memiliki hasil terendah diantara alternatif yang ada yaitu 2,625.

PENINGKATAN LOYALITAS PELANGGAN MELALUI KUALITAS PRODUK, CITRA MEREK DAN KEPUASAN PELANGGAN (Studi pada Pelanggan Sim Card GSM Prabayar Merek XL di Kabupaten Semarang)
Penelitian ini bertujuan untuk menganalisis pengaruh kualitas produk, citra merek dan kepuasan pelanggan terhadap loyalitas pelanggan sim card GSM prabayar XL di Kabupaten Semarang. Populasi yang digunakan adalah pelanggan sim card GSM prabayar XL di Kabupaten Semarang yang jumlahnya tidak dapat diketahui. Teknik pengambilan sampel dalam penelitian ini dilakukan dengan metode purposive sampling sebanyak 100 responden. Analisis data menggunakan regresi linier berganda uji t. Hasil penelitian menunjukkan bahwa kualitas produk dan citra merek berpengaruh signifikan terhadap kepuasan pelanggan, citra merek dan kepuasan pelanggan berpengaruh signifikan terhadap loyalitas pelanggan, sedangkan kualitas produk tidak berpengaruh signifikan terhadap loyalitas pelanggan. Dengan demikian, sebaiknya perusahaan XL terus meningkatkan kualitas produk, citra merek dan kepuasaan pelanggan agar pelanggan tetap percaya pada produk XL Prabayar dan terus menjadi pelanggan setia.

XL-SIM: Extending Superresolution into Deeper Layers
Of all 3D-super resolution techniques, structured illumination microscopy (SIM) provides the best compromise with respect to resolution, signal-to-noise ratio (S/N), speed and cell viability. Its ability to achieve double resolution in all three dimensions enables resolving 3D-volumes almost 10x smaller than with a normal light microscope. Its major drawback is noise contained in the out-of-focus-signal, which-unlike the out-of-focus signal itself-cannot be removed mathematically. The resulting "noise-pollution" grows bigger the more light is removed, thus rendering thicker biological samples unsuitable for SIM. By using a slit confocal pattern, we employ optical means to suppress out-of-focus light before its noise can spoil SIM mathematics. This not only increases tissue penetration considerably, but also provides a better S/N performance and an improved confocality. The SIM pattern we employ is no line grid, but a two-dimensional hexagonal structure, which makes pattern rotation between image acquisitions obsolete and thus simplifies image acquisition and yields more robust fit parameters for SIM

The aim of this study is to determine, (1) The effect of the product purchase decision XL prepaid SIM Card in Tangerang, (2) Effect of price on purchase decisions XL prepaid SIM Card in Tangerang, (3) Effect of distribution of prepaid SIM Card purchase decisions XL in Tangerang,(4) Effect of promotion to the premier prepaid card purchasing decisions XL Tangerang, (5) Effect of employees' purchasing decisions XL prepaid SIM Card in Tangerang, (6) Effect of physical evidence against the decision of purchasing a prepaid SIM Card XL in Tangerang, (7) The effect of the decision process of purchasing a prepaid SIM Card XL in Tangerang. The population in this study were all customers who use prepaid cards XL in Tangerang during the past year or so. The number of samples in this study were 200 respondents. Analytical techniques used in this research is descriptive analysis and pathway analysis (path analysis) by using sampling techniques areaccidental sampling.
