    Daerah Bantul tercatat sebagai penyumbang terbesar ekspor Provinsi Yogyakarta (DIY) dari sektor kerajinan. Banyak klaster-klaster kerajinan yang tersebar sehingga untuk menggali keunggulan bersaing tiap klaster perlu dilakukan pengkajian kemampuan internal Usaha Kecil Menengah kerajinan (UKM) dalam klaster-klaster tersebut. Penelitian ini adalah penelitian dengan fokus pada observasi lapangan lingkungan internal UKM kerajinan. Penelitian ini meneliti kapabilitas inovasi dan kapabilitas pembelajaran UKM. Kedua kapabilitas tersebut diuji dalam kaitannya dengan kinerja. Penelitian dilakukan di Sentra kerajinan Bantul DIY. Dengan purposive sampling, sejumlah 75 kuesioner valid yang diisi pengusaha UKM dianalisis untuk mengetahui pengaruh kapabilitas inovasi dan pembelajaran terhadap kinerja usaha kerajinan mereka. Analisis regresi linear digunakan, dimana kapabilitas inovasi dan kapabilitas pembelajaran menunjukkan pengaruh positif signifikan terhadap kinerja UKM. Didalam lingkungan persaingan yang dinamis disektor kerajinan (ekonomi kreatif), kemampuan berinovasi dan menyerap hal-hal baru menjadi daya saing yang menentukan keberhasilan UKM sektor kerajinan

    Regenerasi Kultur Lengkeng Dataran Rendah cv. Diamond River melalui Embriogenesis Somatik

    ABSTRAK. Beberapa kultivar lengkeng toleran dataran rendah telah diintroduksi ke Indonesia termasuk lengkengcv. Diamond River. Kultivar tersebut telah dibudidayakan secara komersial di daerah Kalimantan Barat. Namun,pengembangannya menghadapi kendala dalam hal penyediaan bibit. Dalam rangka memperoleh bibit lengkengdalam jumlah yang berlimpah, perlu penerapan teknik kultur in vitro. Penelitian ini bertujuan untuk menginduksidan meregenerasikan kalus embriogenik lengkeng cv. Diamond River. Induksi kalus dilakukan menggunakan daunmuda sebagai eksplan. Regenerasi kalus embriogenik dilakukan dalam 4 tahap. Pada tahap pertama digunakan airkelapa pada konsentrasi 5 dan 10%. Pada tahap kedua, diuji pengaruh auksin (IBA dan NAA) serta sitokinin (BAdan kinetin) masing-masing pada taraf 0,5 ppm. Pada tahap ketiga, diuji pengaruh auksin IBA dan NAA pada taraf0,1; 0,5; dan 1 ppm. Pada tahap keempat diuji perlakuan sukrosa pada taraf 2 dan 3% dengan atau tanpa auksin(IBA dan NAA) masing-masing pada taraf 0,5 dan 1 ppm. Hasil penelitian menunjukkan bahwa regenerasi melaluiembriogenesis somatik berpeluang diterapkan pada tanaman lengkeng cv. Diamond River. Respons kalus embriogeniklebih dominan ke arah pembentukan akar daripada tunas. Penggunaan media yang mengandung NAA 1 ppm mampumeningkatkan pembentukan tunas hingga mencapai lebih dari 30%, sedangkan penggunaan sukrosa 3% tanpa auksinmampu meningkatkan pembentukan planlet hingga mencapai 12%. Several low-land longan cultivars have been introduced to Indonesia,including cultivar of Diamond River. This cultivar has been planted commercially and produced well in WestKalimantan. Unfortunately, the development of this cultivar was facing a problem on the availability of plantingmaterials. In order to provide large number of Diamond River seedlings, tissue culture technique was used. Theaim of the study was to induce and regenerate embryogenic calli of longan cv. Diamond River. A research on callusinduction was conducted using young leaves as explants source. Regeneration of embryogenic calli was conductedin 4 steps. The first, coconut water at the rate of 5 and 10% were used. The second, the auxin (IBA and NAA) andcytokinin (BA and kinetin) at the level of 0.5 ppm, respectively were tested. The third, the IBA and NAA at thelevel of 0.1, 0.5, and 1 ppm were used. The fourth, the sucrose at the level of 2 and 3% with or without addition ofIBA and NAA at the level of 0.5 and 1 ppm were used respectively. The results showed that somatic embryogenesisregeneration was potentially applied to longan cv. Diamond River. The root formation was more dominant than theshoot formation. The use of 1 ppm NAA could increase the shoot formation up to more than 30% whereas the useof 3% sucrose without auxin could increase the plantlet formation up to 12%. The 14% of plantlet produced by thistechnique grew well during acclimatization period

    Penyimpanan plasma nutfah pisang di lapang mempunyai risiko hilangnya genotipe tertentu karena cekaman biotik dan abiotik. Penyimpanan in vitro dengan kriopreservasi adalah salah satu cara alternatif untuk melestarikan plasma nutfah pisang. Dengan demikian, metode regenerasi perlu dioptimalkan sebelum menerapkan metode kriopreservasi. Penelitian dilaksanakan di Laboratorium Kultur Jaringan, Balai Besar Penelitian dan Pengembangan Bioteknologi dan Sumberdaya Genetik Pertanian Bogor (BB Biogen). Penelitian ini mencakup empat percobaan utama yaitu, (1) organogenesis aksis bunga pisang bergenom AA, BB, AAA, AAB dan ABB, (2) kultur embrio zigotik pisang Breviformis, Tomentosa dan Zebrina, (3) enkapsulasi embrio zigotik dengan penggunaan manitol, (4) penyimpanan secara kriopreservasi. Tujuan dari penelitian ini adalah untuk (1) mendapatkan formulasi terbaik untuk organogenesis aksis bunga pisang (AA, BB, AAB dan ABB), (2) mengetahui pengaruh durasi simpan terhadap perkecambahan pisang liar (AA) Breviformis, Tomentosa dan Zebrina, (3) untuk mengetahui efektivitas manitol untuk pertumbuhan minimal, (4) untuk mengetahui pengaruh durasi prakultur terhadap daya hidup dan regenerasi scalp dari pisang Kosta yang dikriopreservasi secara pembekuan sederhana, (5) untuk mengetahui pengaruh durasi loading dan dehidrasi terhadap daya hidup dan regenerasi scalp dari pisang Kosta yang dikriopreservasi dengan metode vitrifikasi. Organogenesis dilakukan dengan menggunakan kepingan aksis bunga pisang yang ditanam pada media perlakuan. Kultur embrio dilakukan dengan terlebih dahulu menyimpan buah pisang Breviformis, Tomentosa dan Zebrina selama 1, 3 dan 4 bulan. Pertumbuhan minimal dilakukan dengan teknik enkapsulasi dengan penambahan manitol. Kriopreservasi dilakukan dengan teknik pembekuan sederhana dan teknik vitrifikasi. Hasil penelitian menunjukkan bahwa tidak ada interaksi antara BA dan TDZ terhadap organogenesis aksis bunga pisang (AA, BB, AAA, AAB dan ABB). Pisang dengan genom AAA menunjukkan respon yang paling baik dalam pembentukan nodul dan tunas. Perlakuan yang terbaik untuk organogenesis pisang adalah media yang mengandung BA 3 mg L-1. Pada kultur embrio zigotik, pisang Tomentosa memiliki daya kecambah yang lebih tinggi dibandingkan dengan Breviformis dan Zebrina yaitu sebesar 70%. Perlakuan prakultur pada media yang mengandung sukrosa 0.4 M selama dua minggu merupakan perlakuan terbaik pada kriopreservasi secara pembekuan sederhana. Tidak ada interaksi yang nyata antara durasi loading dengan larutan LS dan dehidrasi dengan larutan PVS2 dalam pembentukan nodul baru dari scalp pisang Kosta. Perlakuan LS selama 40 menit dan PVS2 selama 1 jam merupakan perlakuan terbaik untuk kriopreservasi scalp pisang Kosta dengan teknik vitrifikasi

    Superior customer value is essential to win competition. The perception of customer value has grown in interest since it has been found to have stable impacts on satisfaction, behavioral intentions and ultimately business performance. Considering that the most common definition of perceived value refers to the trade-off between benefits and sacrifices, thus, this study adopts the second-order formative conceptualization of perceived value and examined its links to satisfaction and loyalty. Four main tourist shopping locations in Yogyakarta were used to collect the data. By applying Partial Least Squares to test the model in the tourism shopping context, the results indicate that perceived value has positive relationships with satisfaction and loyalty.     BRANDING CITY DEVELOPMENT PROGRAM TO SUPPORT TOURISM COMPETITIVENESS ( CASE STUDY: JAKARTA AND KUALA LUMPUR )

    Tourism has important roles in supporting the nation development. In the situation where competition in the tourism sector is getting higher, the interests in practicing city branding by the government is increasing. This study applies the combination of qualitative and descriptive approaches in analyzing the role of branding and how it is applied as a case study in Jakarta and Kuala Lumpur. The positioning of Indonesia and Malaysia tourism industry within the ASEAN scope is also analyzed. Jakarta and Kuala Lumpur are chosen as the objects for the case study since both cities have practiced city marketing as well as both have their own city brand. The results using secondary data show that the processes of establishing city branding in both cities have been done through appropriate processes involving many stakeholders. However, the “Enjoy Jakarta” brand is still considered unable in fulÞ lling the promises that it offered. On the other hand, apart from the success of tourism Malaysia in being the most visited country as compared to other ASEAN countries, the “Visit KL” should be consistently evaluated in order to maintain the brand equity that it should have and be able to position the city in the same level as other big cities in the world.     How students' perceive tourism impacts and their supports in tourism development

    The purpose of this study is to examine the residents’ perceptions of tourism impacts and how those perceptions influence their support for tourism development.Higher education students in the final year in Yogyakarta were assessed regarding their perceptions on tourism impacts and their willingness to support tourism development. 108 questioners were valid as primary data for Partial Least Squares (PLS) analysis.Among five proposed hypotheses, only two (economic impact and social benefits impact) were found to be positively significant to tourism development supports.Whereas, social and cultural costs as well as cultural benefits did not significantly influence tourism development supports

    In Vitro Conservation of Pomelo (Citrus maxima (Burm.)Merr.) cv Srinyonya Using Osmoticum and Retardant.Iswari S. Dewi, Gani Jawak, Ika Roostika, M. Sabda,Bambang S. Purwoko, and Widiati H. Adil. Pomelo is anunderutilized citrus fruit with a potential for commercialization.Only some cultivars have been conserved ex situ, suchas in home yards or in botanical gardens. Such collectionsare vulnerable to biotic and abiotic hazards. The goal of theexperiment was to study the effect of osmoticum (sorbitol)and retardant (ancymidol) on in vitro growth of pomelo.Four-leaf in vitro shoots of pomelo cultivar Srinyonya wereused as plant materials. Murashige-Skoog (MS) medium wasused as the basal medium for the culture. The trial wasarranged in a completely randomized design with threereplications. The treatments consisting of MS + sorbitol (0,20, 40, and 60 g/l) and MS + ancymidol (0, 1, 3, and 5 mg/l).The results indicated that based on plant height, number ofnew leaves, and visual plant architecture, sorbitol treatmentsfrom 20-60 g/l retard the growth of the pomelo plant significantly.On the other hand, ancymidol did not inhibit thepomelo growth significantly, but it was a suitable osmoticumfor improvement of in vitro plant vigor, increasing greencolor of leaf, and increasing root initiation. Leaf senescenceof in vitro plants cultured on media containing sorbitol 40and 60 g/l began 20 week after storage. The best medium forconservation of pomelo cv Srinyonya was MS + 20 gsorbitol/l

    &lt;p&gt;In the beginning, cryopreservation&lt;br /&gt;technique was primarily used for germplasms&lt;br /&gt;long-term storage as passive collection because cell division&lt;br /&gt;and metabolism process can be stopped at super low&lt;br /&gt;temperature, commonly in liquid nitrogen. The technique is&lt;br /&gt;suitable for vegetative propagated plants and recalcitrant&lt;br /&gt;seeds. Recently, its application is extending for storing many&lt;br /&gt;species and orthodox seeds. In this paper, the development&lt;br /&gt;of cryopreservation application is discussed. In Indonesia,&lt;br /&gt;cryopreservation is being studied but the application of&lt;br /&gt;cryopreservation has been significantly developed abroad.&lt;br /&gt;The application of cryopreservation technique is not only for&lt;br /&gt;preserving passive collections but also for storing active&lt;br /&gt;collections, including to provide plant materials for hybridization,&lt;br /&gt;cellular engineering, genetic transformation, as well&lt;br /&gt;as pathogens eradication or cryotherapy. It is concluded that&lt;br /&gt;cryopreservation plays an important role in conventional and&lt;br /&gt;modern plant breeding program.&lt;/p&gt;</jats:p

    Modeling Customer Loyalty: Empirical Study on Mobile Internet Users

    The Internet application has grown at a very impressive rate. In a high competitive market, gaining customer loyalty is not an easy job. Quality is no longer adequate to retain customers since today’s customers are faced with so many potential alternatives. Customer value and reputation is becoming more important in helping the companies to acquire customer’s loyalty. This study simultaneously integrates service quality, customer value and reputation to predict loyalty. The results from PLS analysis have identified the important role of service quality on loyalty as compared to customer value and reputation. It is also important that service quality should be highly emphasized since it strongly effects on how customer valuing the offerings and how reputation is built.
