    Asiantuntijana kehittyminen rikosseuraamusalalla

    Tämän päiväkirjamuotoisen opinnäytetyön tavoitteena oli tutkia omaa työtäni asiantuntijana rikosseuraamusalalla ja selvittää millaista osaamista asiantuntijalla on hyvä olla. Opinnäytetyössä tutkin omaa osaamistani ja kehittymistarpeitani sekä pohdin työhyvinvointia ja sitä tukevia tekijöitä. Tutkimus toteutettiin pitämällä päiväkirjaa kymmenen viikon ajan. Jokainen viikko päättyi viikkokoosteeseen pohdintoineen. Viikkokoosteen tukena käytettiin lähdekirjallisuutta teemojen mukaisesti. Päiväkirjahavaintojen, vertaispalautteen ja lähdekirjallisuuden pohjalta laadin yhteenvedon asiantuntijan työtehtävän menestystekijöistä ja toin esiin omaa osaamistani sekä kehittymistarpeitani asiantuntijana. Opinnäytetyön tuloksena syntyi Asiantuntijatyön menestystekijät Talo. Talo-mallin avulla kuvaan millaista osaamista asiantuntijalla on hyvä olla, millaista osaamista minulla on ja sekä tuon esiin omia kehittymistarpeitani sekä työhyvinvointia tukevia asioita. Pohdinnoissa tuon esiin kehittämisajatuksia liittyen asiantuntijoiden perehdyttämiseen ja koulutuspolun rakentamiseen sekä oppimisen kehittämiseen näkemällä työpaikat luonnollisina oppimisympäristöinä. Asiatuntijatyön osaamisen johtamista tulisi myös kehittää. Tulevaisuuden työelämäntaitona nostan esiin vuorovaikutustaidot ja siihen liittyvän koulutuksen lisäämisen. Opinnäytetyö oli matka omaan ammatilliseen kasvuun ja tutkimuksellisen tavoitteen lisäksi prosessi vahvisti ammatillista identiteettiäni ja avasi uusi ammatillisia näkymiä. Opinnäytetyössä käytetty päiväkirjamuoto teki prosessista hyvin henkilökohtaisen. Omien ajatusten, tunteiden ja näkemysten reflektoiminen asettaa tutkijan myös alttiiksi arvostelulle. Toisaalta taas ajattelen, että päiväkirjamuotoisen opinnäytetyön tarinallisuus lisää mahdollisuutta samaistua kirjoittajaan.The objective of this bachelor’s thesis was to explore the author`s work as an expert in the prison and probation services and to map the skill set an expert should possess. The thesis explores the author´s competences, development needs and well-being as well as factors supporting it. The study was carried out by keeping a diary for ten weeks. Each week ended with a weekly summary with reflections. Source literature was used to support the weekly summary according to the themes. Based on the diary observations, peer feedback and source literature, a summary of the success factors of the expert work was drawn up and the author`s development needs were highlighted. As a result of the thesis, a model was created that presents the success factors of expert work. This model helps to describe what competence an expert should have, what competence the author has and highlight the author´s development needs and factors that support well-being at work. In the reflections, development ideas are presented concerning the training of experts and the construction of an educational path, as well as the development of learning by seeing workplaces as natural learning environments. Expert work competence management should also be developed. Interaction skills and the related increase in education are highlighted as future working life skills. The thesis was a journey to professional growth, and in addition to the research goal, the process expands the authors professional prospects. The diary format used in the thesis work made the process very personal. Reflecting on one's own thoughts, feelings and views also exposes the researcher to criticism. On the other hand, the story-like nature of a thesis in the form of a diary increases the possibility of identifying with the author

    Lessons from International Experience

    In the aftermath of the global financial crisis, policymakers around the world are focusing once again on government debt sustainability. In China, subnational government debt is an important part of total government debt, and therefore deserves the attention that policymakers have paid to the topic. Subnational debt has played an important role in financing China’s impressive infrastructure that is the envy of the world. And subnational debt was instrumental in the economic stimulus that China so effectively staged after the global financial crisis, through which China maintained high levels of economic activity. This e-book starts with a summary and presentation of the Chinese perspective on this subject of subnational debt in session one. Session two introduced some international cases which are highly relevant to the current situation in China. Session three brought these international case studies and experts’ perspectives together to arrive at syntheses that narrowed down key issues which China needs to consider going forward in subnational debt management and restructuring. The roundtable discussion among international and Chinese experts on the way forward for China in session four provided an illuminating discussion of this issue. The author hope that this will inform specific policies that will be formulated to help local governments in China manage their subnational debts

    Abstract LB-252: Single-cell mRNA profiling reveal transcriptional heterogeneity among pancreatic circulating tumor cells

    Abstract Background: Single-cell analysis of the transcriptional heterogeneity of circulating tumor cells have the prospect of better understanding the biology of these cells and their involvement in the metastatic process. In addition, such analyses may reveal new knowledge about the mechanisms leading to chemotherapy resistance and tumor progression in pancreatic cancer patients. Methods: Single circulating tumor cells were enriched from blood samples (n = 50) from patients with advanced pancreatic cancer using our previously established negative depletion strategy MINDEC, and detected by immuno-fluorescence microscopy. The single cells were isolated by micromanipulation, reverse transcribed, pre-amplified, and analyzed using single cell multiplex mRNA profiling to reveal transcriptional heterogeneity. Results: Circulating tumor cells were detected in 29 (58%) of the examined blood samples. A total of 48 potential circulating tumor cells were isolated, in which 17 cells had detectable mRNA levels and were identified as circulating tumor cells by expression of epithelial (KRT8, KRT19, EPCAM, E-Cadherin) or mesenchymal (Vimentin, N-Cadherin, ZEB1) markers. These pancreatic circulating tumor cells were revealed to heterogeneously express the examined transcripts, and also showed aberrant expression of cancer stem cell (CD24, CD44, ALDH1A1) markers and the extracellular matrix marker SPARC. Hierarchical clustering and principal component analyses revealed epithelial-like and mesenchymal-like circulating tumor cell subpopulations, which were distinct from white blood cells and cancer cell line cells. Further analyses of the transcript SPARC suggested a relation to epithelial-mesenchymal transition in pancreatic circulating tumor cells. Conclusion: The analysis of single pancreatic circulating tumor cells defines distinct subpopulations and reveals aberrant expression of transcripts relevant for the dissemination of circulating tumor cells to distant sites.     More-Than-Human Empathy in Arts-Based Material Research Processes

    This research paper is related to discussions around the human-environment relationship. It opens the discussion around the roles of humans and nature, or natural materials, as an author in an arts-based process. It attempts to ask questions about the role of the creative process in creating a more equal relationship and empathy building between these roles. Severe global environmental changes suggest that Earth may have entered a new human-dominated geological epoch called the Anthropocene.This chapter explores the role of the creative process in building a more empathic relationship between humans and the environment. Using a theoretical framework including more-than-human empathy, a materialbased approach and creative processes, the authors present two case studies. In ‘The Secret Life of a Material’, design students work with renewable gluten plastic, letting the material guide their creative process. ‘The Biggest Bread Soup in the World’ presents the artist’s immersive experience with Lake Tjörnin in Reykjavik, observing and interacting with its natural environment. These studies show how, when designers and artist–researchers engage in creative practices with nonhuman agents as coauthors, this can lead to unexpected outcomes and enhance empathic connections between humans and the natural world. This chapter highlights how an art-based research (ABR) process can deepen our understanding of and empathy towards nonhumans, encouraging collaborative practices that integrate human and natural agencies.</p

    Pengaruh Campuran Minyak Kelapa Sawit Dengan Bahan Bakar Dexlite Terhadap Kinerja Mesin Diesel Satu Silinder

    Meningkatnya jumlah kendaraan berakibat ke semakin tingginya konsumsi bahan bakar minyak di Indonesia. Oleh sebab itu, perlu dilakukanya upaya penghematan pemakaian bahan bakar. Salah satu upaya nya adalah dengan memanfaatkan minyak kelapa sawit sebagai bahan campuran dexlite. Tujuan dari penelitian ini untuk mempelajari kinerja mesin diesel satu silinder dengan menggunakan bahan bakar campuran minyak kelapa sawit dan dexlite yaitu : Daya, Torsi, Efesiensi Termal, Pengggunaan Bahan Bakar Spesifik dan Emisi Gas Buang yang dihasilkan. Penelitian dilakukan dengan uji eksperimental pada sebuah mesin diesel 1 silinder dengan pembebanan elektrik (lampu pijar) dari 500 Watt sampai 3000 Watt dengan interval 500 watt pada putaran mesin stasioner (1500 rpm) . Dapat disimpulkan bahwa Emisi Gas Buang yang terbaik terdapat pada campuran bahan bakar dexlite 80% + minyak kelapa sawit 20% dengan memiliki rata-rata paling rendah diantara bahan bakar lainnya. Konsumsi Bahan Bakar yang terbaik terdapat pada campuran bahan bakar D60M40 dengan memiliki rata-rata paling rendah diantara bahan bakar lainnya, dengan nilai rata rata 297,694 gr/kWh , dibandingkan dengan bahan bakar campuran D50M50 memiliki rata-rata yaitu 546,93 gr/kWh

    Institutional Dimensions in Open Government Data : A Deep Dive Into Indonesia’s Satu Data Initiative and Its Implications for Developing Countries

    Institutional factors such as data standardization, data interoperability, and the ability to include other stakeholders (e.g., civil society) in ecosystems are now understood as crucial considerations in Open Government Data (OGD) implementation. However, most of the current understanding of institutional factors linked to OGD growth has evolved around highly digitalized and developed countries. This study aims to address this gap by investigating open data implementation in Indonesia, a developing country that has been an early advocate of OGD in the "Asia Pacific". In 2014, Indonesia implemented Satu Data Indonesia (One Data Indonesia) as a national initiative to promote OGD objectives. We employ institutional dimensions as the central theoretical lens to unpack the successes and impediments to Satu Data Indonesia’s development. This research dataset is built on 16 expert interviews from government (n = 14) and private (n = 2) organizations that directly contribute to the growth of Satu Data Indonesia, qualitatively triangulated with documentary analysis of key policy and regulatory documents. These new data provide important insights into the complexity and challenges of nationwide OGD implementation and data-sharing. Our findings show that despite its being an early advocate, Indonesia’s OGD initiative is still in the early stages of development, impeded by several policy and administrative bottlenecks.© 2024 The Author(s).     Currently, technology in the telecommunications sector is developing rapidly.Telecommunication users need fast communication technology with wider bandwidth. Therefore, FBMC / OQAM was developed, which is a modulation candidate technology to be used in 5G.&nbsp;FBMC / OQAM is a development of OFDM which is modified by using filters to reduce noise. OFDM uses a multiplexing technique that divides bandwidth into several sub-carrier frequencies. But OFDM has a weakness, because it requires a Cyclic Prefix (CP) to overcome Intersymbol nterference (ISI) and Intercarrier Interference (ICI). With FMBC, the increased bandwidth due to the Cyclic prefix can be reduced.&nbsp;The filter used is based on the Physical layer for dynamic spectrum access and cognitive radio (PHYDYAS).The results achievedin this study &nbsp;were that FMBC was succesfully and the data sent was the same as the data receive

    Tujuan dari penelitian ini adalah untuk meningkatkan kemampuan operasi hitung perkalian satu angka bagi peserta didik cerebral palsy dengan menerapkan penggunaan media papan stick. Penelitian ini menggunakan metode Penelitian Tindakan Kelas (PTK) dengan desain rencana tindakan adaptasi dari model Kemmis dan Mc. Taggart. Penelitian ini merupakan penelitian kuantitatif-kualitatif dengan teknik pengumpulan data dilakukan melalui observasi, wawancara, dan dokumentasi, kemudian data keseluruhan diolah dengan teknik analisis data kualitatif melalui proses reduksi data, penyajian data kemudian dilakukan penarikan kesimpulan. Subjek dalam penelitian ini adalah peserta didik cerebral palsy tipe spastik berjumlah 3 orang dengan masing-masing peserta didik memiliki karakteristik kemampuan matematika yang berbeda. Hasil penelitian menunjukkan bahwa tiga peserta didik cerebral palsy kelas V mengalami peningkatan keterampilan operasi hitung perkalian satu angka secara signifikan setelah diterapkannya penggunaan media papan stick, hasil peningkatan tersebut berdasarkan analisis dari observasi kemampuan awal, pengamatan setiap siklus, dan refleksi serta data hasil evaluasi tes siklus peserta didik. Penggunaan media pembelajaran papan stick telah terbukti membantu peserta didik dalam meningkatkan kemampuan operasi hitung perkalian satu angka. Penelitian lebih lanjut diperlukan untuk terkait penggunaan media papan stick pada tingkatan kemampuan operasi hitung perkalian yang lebih tinggi dari kemampuan operasi hitung perkalian satu angka. ***** The purpose of this study was to improve the ability of one-digit multiplication calculation operation for cerebral palsy students by applying the use of stick board media. This research uses the Classroom Action Research (PTK) method with an adapted action plan design from the Kemmis and Mc. Taggart model. This research is quantitative-qualitative research with data collection techniques are carried out through observation, interviews, and documentation, then the overall data is processed with qualitative data analysis techniques through the process of data reduction, data presentation and then drawing conclusions. The subjects in this study were 3 spastic type cerebral palsy students with each student having different characteristics of mathematical ability. The results showed that three fifth grade cerebral palsy learners experienced a significant increase in one-digit multiplication calculation skills after the implementation of the use of stick board media, the results of the increase were based on analysis of initial ability observations, observations of each cycle, and reflections as well as data from the evaluation of students' cycle tests. The use of stick board learning media has been proven to help students in improving the ability of one-digit multiplication counting operations. Further research is needed on the use of stick board media at higher levels of multiplication calculation skills than one-digit multiplication calculation skills

    High-fidelity DNA polymerase enhances the sensitivity of a peptide nucleic acid clamp PCR assay for K-ras mutations.

    Sensitive detection of tumor-specific point mutations is of interest in both the early detection of cancer and the monitoring of treatment at a molecular level. Recently, peptide nucleic acid (PNA) clamp real-time PCR has provided a time-sparing and sensitive method for the detection of mutations in the presence of a large excess of wild-type DNA. We present the first report that the sensitivity of PNA clamp PCR is limited by the low fidelity of TaqDNA polymerase. Replication errors introduced by Taq polymerase in the PNA-binding site were amplified during PCR due to the resulting mismatches between PNA and DNA. To reduce the frequency of polymerase-induced errors, we developed a PNA clamp PCR assay for the detection of mutations in codons 12 and 13 of the K-ras gene based on a high-fidelity DNA polymerase. The sensitivity of our assay increased approximately 10-fold, significantly detecting mutant DNA diluted 20,000-fold in wild-type DNA (P = 0.025), compared with its detection at 2000-fold dilution (P = 0.039) when Taq polymerase was used. Our data suggest that the replication errors caused by Taq polymerase must be taken into consideration for PNA clamp PCR and for other methods based on selective PCR amplification, and that these assays can be enhanced by high-fidelity DNA polymerases
