Retinal differentiation of human bone marrow-derived stem cells by co-culture with retinal pigment epithelium in vitro.
The goal of this study was to assess the in vitro differentiation capacity of human bone marrow-derived stem cells (hBMSCs) along retinal lineages. Mononuclear cells (MNC) were isolated from bone marrow (BM) and mobilized peripheral blood (mPB) using Ficoll-Paque density gradient centrifugation, and were sorted by magnetic-activated cell sorting (MACS) for specific stem cell subsets (CD34(+)CD38(+)/CD34(+)CD38(-)). These cells were then co-cultured on human retinal pigment epithelial cells (hRPE) for 7 days. The expression of stem cell, neural and retina-specific markers was examined by immunostaining, and the gene expression profiles were assessed after FACS separation of the co-cultured hBMSCs by quantitative reverse transcription polymerase chain reaction (qRT-PCR). Furthermore, in vitro functionality of the differentiated cells was analyzed by quantifying phagocytosis of CY5-labeled photoreceptor outer segments (POS). After 7 days of co-culture, hBMSCs adopted an elongated epithelial-like morphology and expressed RPE-specific markers, such as RPE65 and bestrophin. In addition, these differentiated cells were able to phagocytose OS, one of the main characteristics of native RPE cells. Our data demonstrated that human CD34(+)CD38(-) hBMSC may differentiate towards an RPE-like cell type in vitro and could become a new type of autologous donor cell for regenerative therapy in retinal degenerative diseases

Cell resolved, multiparticle model of plastic tissue deformations and morphogenesis
We propose a three dimensional mechanical model of embryonic tissue dynamics. Mechanically coupled adherent cells are represented as particles interconnected with elastic beams which can exert non-central forces and torques. Tissue plasticity is modeled by a stochastic process consisting of a connectivity change (addition or removal of a single link) followed by a complete relaxation to mechanical equilibrium. In particular, we assume that (i) two non-connected, but adjacent particles can form a new link; and (ii) the lifetime of links is reduced by tensile forces. We demonstrate that the proposed model yields a realistic macroscopic elasto-plastic behavior and we establish how microscopic model parameters affect the material properties at the macroscopic scale. Based on these results, microscopic parameter values can be inferred from tissue thickness, macroscopic elastic modulus and the magnitude and dynamics of intercellular adhesion forces. In addition to their mechanical role, model particles can also act as active simulation agents and modulate their connectivity according to specific rules. As an example, anisotropic link insertion and removal probabilities can give rise to local cell intercalation and large scale convergent extension movements. Using Lesson Study to Help Teachers Design Lessons with Purposeful Planned Movement and Build Efficacy
abstract: Due to the push down of academics, today's elementary students are being asked to learn more concepts and sit for longer periods of time. Sitting slows thinking, whereas movement wakes up the brain. Using movement to learn is embodied cognition, or learning through both the body and the brain. Movement should be part of instruction for young students; however teachers are often not sure how to incorporate movement in their lesson plans. The Japanese practice of lesson study may help because it embeds teachers' new learning in their classrooms while intimately connecting it to the learning of their students, and it links with the cyclical, constructed theory of learning provided by Vygotsky Space. If teachers incorporate movement in their lessons, children have the potential to become more engaged and learn. This action research study was designed to understand if two first grade, two second grade, and one third grade teacher at a Title One elementary school in the Southwestern United States could learn how to use movement more during instruction through lesson study. This innovation took place for 14-weeks during which 12 lessons using movement were developed and taught. Data were collected prior to the study and during each portion of the cyclical process including, while teachers learned, during lessons using movement, and when lessons were discussed and changed. The data sources were pre and post teacher surveys, student surveys, observation protocols, lesson plans, transcripts of lesson study meetings, and researcher notes. To reduce bias a triangulated mixed methods design was used. Results indicate that through lesson study teachers were able to learn about movement, try it, observe the results, and adjust it to fit their teaching style and their students' needs. Data showed increased student engagement in lessons that incorporated movement as evidenced in the students' words, bodies, and learning. After participating in the study, the teachers realized they personally use movement to learn, and teachers' efficacy regarding their ability to plan movement in their lessons increased. Using Social Media and Professional Learning Communities as Tools for Novice Teacher Collegiality and Improved Self-Efficacy
abstract: Teacher attrition and the migration between schools and districts can have a negative impact on quality of education and teacher performance. Novice teachers leave the profession because they are overwhelmed by the workload and responsibilities of the job. In a previous action research cycle, I found that novice teachers' perceptions of isolation and lack of opportunities to share experiences had a negative effect on teacher perceptions of efficacy. This action research project examines the effect of leveraging social media and professional learning communities to provide opportunities for a group of novice teachers to share experiences and seek advice. By addressing the challenges that novice teachers face and providing solutions for common problems, it is the hope of this researcher that highly effective teachers will remain in the classroom. The results of the study indicate that the combined use of Twitter and YouTube in collaboration with professional learning communities will improve teacher perceptions of efficacy. Teachers who participated in the social media based professional learning communities are also more likely to remain in the classroom.Dissertation/ThesisEd.D. Examining the Effect of Club Aspire on Low Achieving Middle School Students
abstract: This action research, mixed methods, case study examined middle school student perceptions of the effectiveness of Club Aspire. Club Aspire is an after-school program created to support the lowest achieving seventh and eighth graders in an Arizona K-8 school. The framework of this study comes from the theory of self-regulation, social learning theory and co-regulation. The primary focus of Club Aspire is to teach low achieving middle school students, self-regulation skills and learning strategies through goal setting, self-regulation learning strategy lessons and co-regulation activities.
The study took place over 13 weeks and included 11 participants and answered the following research questions. How do middle school Elevate students perceive the impact of Club Aspire on their self-regulation and themselves as a learner? How does Club Aspire affect middle school Elevate students’ academic success? What do middle school Elevate students perceive as the most influential elements of Club Aspire? Data collection tools consisted of interviews, class work, referral data, pre- and post-questionnaire and benchmark assessment data.
The study revealed that students made gains in self-regulation learning strategy usage, however, their academic achievement was not influenced. Students identified goal setting, learning self-regulation strategies and co-regulation activities with their peer partner as the most beneficial elements of Club Aspire. Considerations regarding "Portrait caricature" in the Republic of Moldova. The period of 1963-2023
The respective work proposes to present aspects of the evolution of "Portrait caricature" on the territory of the Republic of Moldova. "Portrait caricature" is a variation of caricature that was and is still practiced by lots of visual artists from Moldova, as Glebus Sainciuc, Sergiu Puica, Isai Cirmu, Alex Dimitrov, Margareta Chitcatii and more others. Over time, pieces of art made by these artists saw the light of being published in various periodicals, such as satirical and humour magazine "Chiparușul", magazines "Femeia Moldovei" and "Nistrul", "Tinerimea Moldovei" and "Timpul" newspapers. This study establishes, structures and reflects visual options of "Portrait caricature" in different time periods. CHANGES IN SOME MORPHOLOGICAL CHARACTERISTICS OF YOUTH
Determining of two different youth groups in terms anthropometric variable measured twenty years ago and now is the goal of this research. The testing sample has been defined as a sample taken out of the researched middle school students such from Gymansium ‘Zenel Hajdini’ Middle School of Economy ‘Marin Barleti’ and Middle Technical School ‘Mehmet Isai’ from Gjilan. In this sample 263 female students have been included aged 18 (six months older or younger). Out of the same number of students of male gender aged 18 (six months older or younger) form the same middle schools in the same measuring conditions, an investigation has been done in 1991 by the same author. Twelve anthropometric variables have been used. The T-test has been applied in order to certify weather it does or does not exist difference between the groups’ measures done in 2011 and the group of students in 1991. Considering the arithmetic average results of each variable the results are quite obvious particularly with the first group measured in 2011. The biggest difference has bee noticed with the measures in terms of body length, extremities and measures for assessing body weight
Uclacyanin Proteins Are Required for Lignified Nanodomain Formation within Casparian Strips
© 2020 The Author(s) Casparian strips (CSs) are cell wall modifications of vascular plants restricting extracellular free diffusion into and out of the vascular system [1]. This barrier plays a critical role in controlling the acquisition of nutrients and water necessary for normal plant development [2–5]. CSs are formed by the precise deposition of a band of lignin approximately 2 μm wide and 150 nm thick spanning the apoplastic space between adjacent endodermal cells [6, 7]. Here, we identified a copper-containing protein, Uclacyanin1 (UCC1), that is sub-compartmentalized within the CS. UCC1 forms a central CS nanodomain in comparison with other CS-located proteins that are found to be mainly accumulated at the periphery of the CS. We found that loss-of-function of two uclacyanins (UCC1 and UCC2) reduces lignification specifically in this central CS nanodomain, revealing a nano-compartmentalized machinery for lignin polymerization. This loss of lignification leads to increased endodermal permeability and, consequently, to a loss of mineral nutrient homeostasis
DAMPAK PENYESUAIAN LAJUR KHUSUS KENDARAAN BERAT TERHADAP KINERJA LALU LINTAS
Traffic performance on the Narogong highway has problems with the desired speed of the highway, as a result of the large number of heavy vehicles that pass through the Narogong Highway, this study was conducted to see that moving heavy vehicles to the left lane can have a positive impact on road traffic performance. observed. Data processing uses microscopic simulations to get the desired road simulation, from this simulation calibration and validation are carried out first to get the existing simulation conditions similar to the observed conditions of the road section being reviewed. From the results of the existing conditions that have been validated, a simulation of the design road model is carried out to see if there are changes that affect the design made by the author. Based on the simulation results, it was found that moving heavy vehicles to the left lane had an impact on increasing the speed of other vehicles such as motorcycles and cars significantly. Therefore, it is necessary to add new policies about heavy vehicle.

Kinerja lalu lintas di jalan raya Narogong memiliki permasalahan dengan kecepatan yang diinginkan dari jalan raya tersebut, akibat dari banyaknya kendaraan berat yang melewati ruas jalan raya Narogong, Penelitian ini dilakukan untuk melihat pemindahan kendaraan berat ke lajur kiri dapat berdampak positif pada kinerja lalu lintas ruas jalan yang diamati. Pengolahan data menggunakan simulasi mikroskopis untuk mendapatkan simulasi jalan yang diinginkan, dari simulasi ini dilakukan kalibrasi dan validasi terlebih dahulu untuk mendapatkan kondisi simulasi eksisting yang mirip dengan kondisi observasi ruas jalan yang ditinjau. Dari hasil kondisi eksisting yang sudah divalidasi, dilakukan simulasi model jalan desain untuk melihat apakah ada perubahan yang berpengaruh dari desain yang dilakukan oleh penulis. Berdasarkan hasil simulasi ditemukan bahwa pemindahan kendaraan berat ke lajur sebelah kiri berdampak pada peningkatan kecepatan kendaraan lain seperti motor dan mobil secara signifikan, sehingga diperlukannya kebijakan yang baru untuk kendaraan berat
