    Through interviews with settlement workers and newcomers, three overarching themes were extracted, and divided into several sub-themes. Beginning with the diversity of workers and the positive work environment in the Multicultural and Immigration Integration Services division (MIIS), students in LINC classes show they have thorough knowledge about not only Canada and important, relevant topics to share with students, they also have multicultural exposure and knowledge. They’re also familiar with common struggles that newcomers face when settling into a new country, culture, and institutional system.This study was developed and written in two directed studies courses, supervised first by Dr. Linda Pardy and followed by Dr. Jeffrey Orr. Direction and brainstorming for the project was a collaboration between the student researcher (Darien Johnsen) and Archway Community Service's Multicultural and Immigration Integration Services division

    Abstract Introduction: Therapy of metastasized prostate cancer (PCA) largely depends on androgen deprivation therapy (ADT). Despite initial response, tumors inevitably reach castration resistance (CRPC), a stage at which chemotherapy and second-generation antiandrogens (e.g. Enzalutamide) represent the main therapeutic options. Acquired resistance to these drugs is an emerging problem, raising a need for new approaches. Deletion or mutation of the Chromodomain Helicase DNA binding protein 1 (CHD1) gene, encoding an ATP-dependent chromatin remodeler, is more prevalent in CRPC compared to earlier tumor stages. Therefore, we investigated the effects of CHD1 depletion in androgen-sensitive LNCaP cells. Experimental procedures: LNCaP cells were grown in media with either 10% fetal bovine serum (FBS), 10% charcoal-stripped serum (CSS) or 10% CSS plus 1nM testosterone. Growth was visualized by image-based cell cytometry. After 10 days of growth, RNA sequencing (RNA-seq) was performed to investigate changes in gene expression (between the different growth conditions with and without CHD1 depletion). Results were verified by qRT-PCR and Western blot. The identified treatment options were tested with small-molecule inhibitors in different concentrations. Finally, we performed chromatin immunoprecipitation followed by next-generation sequencing (ChIP-seq) in order to identify the epigenetic mechanisms underlying the differential regulation. Results: We identified a correlation between androgen-independent growth and CHD1 depletion which could be compensated by addition of 1 nM testosterone. Moreover, we identified CHD1-dependent regulation of Enhancer Of Zeste Homolog-2 (EZH2), which could be confirmed at the protein level. Treatment with 1 µM JQ-EZ (an EZH2 inhibitor) showed a stronger effect on the proliferation of CHD1-knockdown cells compared to controls. ChIP-seq and RNA-seq analysis helped to identify a subset of EZH2-bound genes depending on CHD1 knockdown and androgen deprivation. Among those genes, we identified the I-SMAD genes SMAD6 and SMAD7 which are regulators of TGFβ and BMP signaling to be upregulated after androgen-deprivation and downregulated with CHD1 knockdown. We also showed differential regulation of PMEPA1, a TGFβ-dependent inhibitor of PCA bone metastasis. Coherently, we found upregulation of mesenchymal markers such as CDH2, pointing towards a role in EMT. Conclusion: We identified a new regulatory mechanism for CRPC harboring a CHD1-deletion leading to a more mesenchymal state in an EZH2-dependent manner. First experiments with an EZH2 inhibitor in our system indicate a possible therapeutic use.     A global picture of the first abrupt climatic event occurring during the last glacial inception

    The orbital-scale transition from the last interglacial to glacial climate corresponds to the progressive organization of global millennial-scale climate variability. Here, we investigate the structure and the global fingerprint of the first warming event occurring during the last glacial inception, the Greenland InterStadial 25 (GIS 25). Using centennial to decadal-resolution measurements of d18O and dD in the ice together with d15N, d18O2 and CH4 in the trapped air, we show that GIS 25 does not coincide with large environmental changes at lower latitudes. Such an equivocal fingerprint questions whether GIS 25 is simply a smaller amplitude version of later rapid events or whether it reflects a more regional northern hemisphere origin for the initiation of the millennialscale climatic variability. After this ambiguous first rapid event, the onset of the global millennial-scale variability - characteristic of the last glacial period- occurs as a short (300 years) event ending GIS 25.     Mulige årsaker til fiskedød høsten 1993 ved Rygja Smolt A/S som ligger innerst i Lovrafjorden i Sandsfjordsystemet i Ryfylke, er vurdert. Settefiskanlegget tar sitt råvann fra Lovravatnet som har forbindelse med Lovrafjorden og dermed brakkvann i bunnen. Vannet har H2S i den nedre delen av vannsøylen, og fiskedøden settes i sammenheng med opp-pressing av H2s-holdig vann under kraftig nordavind. For å redusere sannsynligheten for tilsvarende episoder anbefales det å flytte råvannsinntaket lenger inn i vannet. H2S-holdig bunnvann representerer en potensiell fare for fisken i settefiskanlegget. For å redusere stabiliteten i vannsøylen og tilføre vannet oksygen foreslås det å føre ferskvann ned i bunnvannet. For å gjøre Lovravatnet om til en stabil råvannskilde, anbefales det å avstenge forbindelsen mellom Lovravatnet og Lovrafjorden og dermed gjøre Lovravatnet om til rent ferskvannssystem. For å fjerne det brakke bunnvannet må avstengningen kombineres med nedføring av ferskvann til dypvannet. En slik restaurering av Lovravatnet anses også å være positiv for den lokale fiskestammens utviklingsmuligheter. Rapporten inneholder også et overvåkingsprogram for Lovravatnet

    På grunnlag av modellkjøringer, befaring og generell kunnskap om effekter av vassdragsreguleringer på det marine miljø er det foretatt en konsekvensvurdering av etablering av Pollen kraftverk i Nordalsfjorden. I rapporten blir eventuelle effekter av redusert ferskvannstilførsel om sommeren og økt tilførsel om vinteren diskutert. Det konkluderes med at etablering av Pollen kraftverk ikke vil gi vesentlige endringer i brakkvannslaget i indre del av fjorden, og det marine miljø i selve Nordalsfjorden vil endres lite. I området utenfor munningen av Nordalselva og spesielt i Pollen vil det skje endringer i vekstvilkårene for både alger og dyr

    I rapporten er det gjort målinger og beregninger for å finne en strategi for utslipp av avløpsvann til Kobbavika slik at overslag til overflaten forhindres. For å forhindre opptrenging om vinteren når tetthetssjiktningen er liten, er det nødvendig å benytte diffusor. Utslippene til Hellandsfjorden vurderes totalt sett å gi liten endring av den totale primærproduksjonen i fjorden, men under ugunstige forhold kan økt algeproduksjon og sedimentasjon av partikler føre til økt belastning på oksygenet i bunnvannet. En ytterligere senkning av terskeldypet i Hellandsstraumen vil gi liten miljømessig gevinst i forhold til dagens tilstand.Fitjar Laks A/
