    In dairy herd management, somatic cell counting is largely used as an indicator of mastitis. Differential cell counts been indicated as a more accurate method for mastitis identification. Little is known, though, about the expected profile in healthy milk samples. This cross-sectional study explored the inflammatory profiles in milk from 2 dairy herds known to be affected by mammary gland infections from different Staphylococcus aureus strains, and 1 herd known to be free of contagious milk pathogens. Milk sampling was performed on 50 animals. Somatic cell counts and bacteriological examination were performed as described on NMC guidelines. Relative percents of lymphocytes (L), macrophages (M), polymorphonuclear cells (PMN); and their ratios were assessed through milk cytology. Results confirmed that herd A was free from contagious pathogens, herd B was positive for a coagulase negative strain of S. aureus, and herd C was positive for S. aureus. In all herds, healthy quarters as well as subclinically mastitic quarters were found. When cytology results were grouped by herd, the inflammatory profiles resulted markedly different. Our results suggest that milk immune response can vary from herd to herd. We do not know, though, if the differences are given to different genetic background, or from different stimulus from the different pathogen strains. Further studies must be done to determinate if a sole factor could be implicated, or if this differences are given to a combination of those factors

    In the process of the manufacture of Ultra Hard.Steel with the Powder Metallurgy Method,it is unavoidable to produuce the reluctant and abundant scraps,from which the expensive metals are not easily recovered. The author recovered tungsten and tantalum as oxides at the average rate of 97% (tungsten oxide)and 9O% (tantalum oxide)from the scrap pieces which consisted of tungesten carbide(8%),titanium carbide(6%),tantalum carbide(4%)and cobalt(l0%). The most important chemical treatments are to be experienced.in the next processes,the pieces are heated 800℃ for l.5−2 hours in the air,then they become powders of oxides not to be attainable with physical methods. Potassium hydroxide,five times as much,is added to the weighed sample,then fused−at 800℃ for about one time,and allowed to cool in the air. Next tme,potassium carbonate, five times as much,is added to the cooled one, and then fused atll00℃ thorougly.On this o㏄asion,the iron crucible is less corroded.For the chemical separation of tungsten and tantalum,the greatest nervous attention is necessary so as not to fail to catch either of them completely

This role is best served by metaphors with a simple syntactic structure, highly conventionalised and em- ploying stereotypical content in their vehicle. In her description of metaphor’s me- aning-creating mechanism, the author emphasises the particular status of metaphoric pre- dicate that is suggested by the utterance’s sender and independently produced by the rece- iver using the metaphor vehicle’s connotation. The valuing so expressed is not subject to direct negation and so is quite powerful in terms of persuasiveness. Sedangan pada tahun 2023 terdapat 17,4 juta ton timbulan sampah tahunan yang dihasilkan. Dari data tersebut, sebanyak 11,6 juta ton atau 66,47% sampah telah terkelola. Yang artinya Volume timbulan sampah turun sebanyak 51,41% dibanding 2022. Keberlanjutan lingkungan menghadapi tantangan serius akibat peningkatan jumlah sampah yang dapat terjadi kapan pun disetiap tahunnya. Penting untuk memulai edukasi mengenai pengenalan sampah dan pengelolahannya sejak masih anak- anak agar dapat memahami dampak lingkungan dari tindakan mereka. Kurangnya media pembelajaran yang spesifik dan menarik untuk anak�anak di umur 6-12 tahun tentang sampah saat ini membuat kurangnya pengetahuan anak tentang sampah disekitar mereka. Oleh karena itu, perancangan board game dapat dianggap sebagai solusi yang kreatif. Board game dirancang menggunakan metode design thinking yang memuat lima tahapan yaitu emphatize, define, prototype, dan test. Pengumpulan data dilakukan dengan cara observasi, wawancara, studi literatur, dan studi komparasi. Hasil dari perancangan board game diuji dengan bertahap hingga menghasilkan board game yang tepat guna untuk anak-anak dengan usia 7-12 tahun. Board game “Trash Pack” dapat menjadi solusi untuk media pembelajaran anak 7-12 tahun tentang pengelolaan sampah, diharapkan untuk peneliti selanjutnya dapat mengembangkan board game versi digital yang dapat dimainkan kapan saja dan di mana saja

    CD44 Modulates Cell Migration and Invasion in Ewing Sarcoma Cells

    The chimeric EWSR1::FLI1 transcription factor is the main oncogenic event in Ewing sarcoma. Recently, it has been proposed that EWSR1::FLI1 levels can fluctuate in Ewing sarcoma cells, giving rise to two cell populations. EWSR1::FLI1low cells present a migratory and invasive phenotype, while EWSR1::FLI1high cells are more proliferative. In this work, we described how the CD44 standard isoform (CD44s), a transmembrane protein involved in cell adhesion and migration, is overexpressed in the EWSR1::FLI1low phenotype. The functional characterization of CD44s (proliferation, clonogenicity, migration, and invasion ability) was performed in three doxycycline-inducible Ewing sarcoma cell models (A673, MHH-ES1, and CADO-ES1). As a result, CD44s expression reduced cell proliferation in all the cell lines tested without affecting clonogenicity. Additionally, CD44s increased cell migration in A673 and MHH-ES1, without effects in CADO-ES1. As hyaluronan is the main ligand of CD44s, its effect on migration ability was also assessed, showing that high molecular weight hyaluronic acid (HMW-HA) blocked cell migration while low molecular weight hyaluronic acid (LMW-HA) increased it. Invasion ability was correlated with CD44 expression in A673 and MHH-ES1 cell lines.     The Transcription Factor FEZF1, a Direct Target of EWSR1-FLI1 in Ewing Sarcoma Cells, Regulates the Expression of Neural-Specific Genes

    Ewing sarcoma is a rare pediatric tumor characterized by chromosomal translocations that give rise to aberrant chimeric transcription factors (e.g., EWSR1-FLI1). EWSR1-FLI1 promotes a specific cellular transcriptional program. Therefore, the study of EWSR1-FLI1 target genes is important to identify critical pathways involved in Ewing sarcoma tumorigenesis. In this work, we focused on the transcription factors regulated by EWSR1-FLI1 in Ewing sarcoma. Transcriptomic analysis of the Ewing sarcoma cell line A673 indicated that one of the genes more strongly upregulated by EWSR1-FLI1 was FEZF1 (FEZ family zinc finger protein 1), a transcriptional repressor involved in neural cell identity. The functional characterization of FEZF1 was performed in three Ewing sarcoma cell lines (A673, SK-N-MC, SK-ES-1) through an shRNA-directed silencing approach. FEZF1 knockdown inhibited clonogenicity and cell proliferation. Finally, the analysis of the FEZF1-dependent expression profile in A673 cells showed several neural genes regulated by FEZF1 and concomitantly regulated by EWSR1-FLI1.     EWS-FLI1 confers exquisite sensitivity to NAMPT inhibition in Ewing sarcoma cells

    Ewing sarcoma (EwS) is the second most common bone cancer in children and adolescents with a high metastatic potential. EwS development is driven by a specific chromosomal translocation resulting in the generation of a chimeric EWS-ETS transcription factor, most frequently EWS-FLI1.Nicotinamide adenine dinucleotide (NAD) is a key metabolite of energy metabolism involved in cellular redox reactions, DNA repair, and in the maintenance of genomic stability. This study describes targeting nicotinamide phosphoribosyltransferase (NAMPT), the rate-limiting enzyme of NAD synthesis, by FK866 in EwS cells. Here we report that blocking NAMPT leads to exhaustive NAD depletion in EwS cells, followed by a metabolic collapse and cell death. Using conditional EWS-FLI1 knockdown by doxycycline-inducible shRNA revealed that EWS-FLI1 depletion significantly reduces the sensitivity of EwS cells to NAMPT inhibition. Consistent with this finding, a comparison of 7 EwS cell lines of different genotypes with 5 Non-EwS cell lines and mesenchymal stem cells revealed significantly higher FK866 sensitivity of EWS-ETS positive EwS cells, with IC50 values mostly below 1nM.Taken together, our data reveal evidence of an important role of the NAMPT-mediated NAD salvage pathway in the energy homeostasis of EwS cells and suggest NAMPT inhibition as a potential new treatment approach for Ewing sarcoma.Austrian Science Fund (FWF), grant I1225-B19; European Union's FP7 project ASSET (grant agreement No. 259348); Research Manitoba and CancerCare Manitoba Foundation.
