    Franchising is a business cooperation venture in which there are Intellectual Property Rights that are held and owned by the franchisor and can be used by other parties or the franchisee. In carrying out a franchise, an agreement is first made between the two parties. In this study, the authors examine the form and content of the Parikesit Cake franchise agreement and how the legal relationship between the franchisor and the franchisee is in the Parikesit Cake franchise agreement. This study aims to determine the form and content of the Parikesit Balok Cake franchise agreement and to find out the legal relationship of the parties in the Parikesit Baked Cake agreement. The research method used by the author to sell the Parikesit Balok Cake franchise agreement is to use a normative juridical research method, namely testing whether the form and content and legal relations in the Parikesit Balok Cake franchise agreement comply with the provisions of the Agreement Law and Government Regulation No. 42 of 2007 concerning Franchising. The results of this study indicate that the form and content of the Parikesit Cake Franchise agreement between the franchisor and the franchisee has been set forth in the form of sound articles from Article 1 to Article 15 of the Parikesit Cake Franchise Agreement and made in writing, this has fulfilled the requirements in article 5 of Government Regulation No. 42 of 2007 concerning Franchise and legal relations which both parties have stated in article 5 of the Parikesit Cake Bake franchise agreement using the principle of freedom of contract

    Bioinformatics Approach towards Transcriptomics of Filaggrin

    Filaggrin, or filaments which combines protein, is one of the important structural protein that works for the development, maintenance, and the formation of the skin as an intact barrier. Filaggrin breakdown products regulate the hydration of the skin; contribute to the acidic pH of the skin, which in turn is essential for the activity of various proteases in the stratum corneum desquamation and lipid synthesis. Filaggrin produced by keratinocytes granular as a major precursor called profilaggrin, encoded by the FLG gene, located in the epidermal differentiation complex on chromosome 1 (1q21 locus). The locus contains a group of genes involved in epidermal differentiation. Filaggrin deficiency has some consequences on the organization and function of epidermal with important implications such as increased risk for atopic disease or a microbial infection. FLG mutation, a gene that encodes filaggrin, has been shown to cause ichthyosis vulgaris, increasing the risk of atopic dermatitis and other atopic diseases. This research examined the FLG gene based bioinformatics approach to search for conserved region of representative mammals that encode coding (m) and non-coding (nc) RNAs. Expected mRNA expression can be used as a diagnostic and therapeutic agent against deficiencies and filaggrin mutations.Key words: filaggrin, FLG, profilaggrin, filaggrin deficiency, bioinformatics

    Molecular Docking and Dynamics of SARS-CoV-2 Programmed Ribosomal Frameshifting RNA and Ligands for RNA-Targeting Alkaloids Prospecting

    RNA-ligand docking is a part of computational biology, which is currently lowly recognized compared to the protein-ligand docking procedure commonly applied for drug discovery. This in silico study aims to create a simplified protocol for RNA-ligand docking, which is applicable to RNA-targeting small molecular drug screening. Four alkaloids (berberine, colchicine, nicotine, and tomatine) were subjected to this study and contended against the SARS-CoV-2 genomic RNA -1 PRF component targeting control drug, merafloxacin, including two known intercalator berberine and colchicine, a small alkaloid nicotine and a large alkaloid tomatine. The alkaloids were screened for drug-likeness properties (Lipinski’s Rules of 5 or LRo5), bioavailability indexes, and synthetic accessibility values using SwissADME before docking. The docking used PyRx – Autodock Vina and re-scored for RNA-ligand scoring using AnnapuRNA. The docking results have the interactions mapped using fingeRNAt and visualized using Discovery Studio. Molecular dynamics using CHARMM36 and AMBER forcefields were simulated in NAMD. The molecular dynamics 1 ns simulation results showed that the ligand interaction over time did not cause much interference with the RNA, indicated by the low number of RMSD changes between RNA itself and the RNA-ligand complex. Additionally, CHARMM36 forcefield provided more stable fluctuation compared to AMBER. The results indicated that tomatine disobeyed LRo5 and had a low bioavailability index and bad synthetic accessibility value, while the rest alkaloids passed. In the end, berberine has an even higher docking score than the control drug. The study also shows that this protocol can be useful for future RNA-ligand computational studies

    PREDIKSI STRUKTUR 2-DIMENSI NON-CODING RNA DARI BIOMARKER KANKER PAYUDARA TRIPLE-NEGATIVE DENGAN VIENNA RNA PACKAGE

    Berdasarkan data WHO, kanker adalah penyakit yang paling berbahaya. Dewasa ini para peneliti sedang berusaha memahami mekanisme molekular kanker. Berdasarkan dogma sentral, hanya protein coding gene yang diketahui fungsinya, sementara non-coding gene belum dapat dijelaskan. Kemudian, diketahui bahwa non-coding RNA (ncRNA) berperan dominan dalam regulasi molekular sel, sehingga berpengaruh secara langsung kepada proliferasi kanker. Dalam hal ini, instrumen RNA-seq maupun Tiling Array sudah mengumpulkan banyak data biologis dan mendeposisikannya kepada database genom. Diketahui bahwa, ncRNA tidak hanya dapat berperan sebagai biomarker untuk diagnostik kanker, namun juga akan dapat dikembangkan sebagai agen terapeutik. Kanker payudara memiliki empat subtipe molekular, yaitu luminal A, luminal B, Her-2 dan triple negative/basal-like. Kanker Payudara Triple-negative (TNBC) merupakan penyakit yang sangat berbahaya dan belum ditemukan pengobatan yang efektif. Memahami mekanisme dan struktur ncRNA pada Biomarker TNBC merupakan langkah awal untuk menentukan agen terapeutik dan propilaksis terbaik. Diketahui bahwa jalur ekspresi lincRNA-RoR/miR-145/ARF6 berperan dalam proliferasi TNBC. Berdasarkan pencarian di GenBank, ditemukan  lema-lema ncRNA untuk jalur tersebut. Hasil pencarian diolah dengan software Vienna RNA Package, untuk ditentukan struktur 2 dimensi (2-D) yang solid. Kedepannya, diharapkan dengan mencegah terbentuknya struktur 2-D tersebut, maka semua gen tersebut akan menjadi tidak aktif dan menghentikan proliferasi kanker
