Development STEM E-Booklet Curug Winong Fern Plants as a Student Learning Resource
This study aims to determine the types of ferns in the Curug Winong and the results of the E-booklet product feasibility test from the research results. This research uses the Research and Development method with a size trap of 4 x 4 m. The results of the identification of ferns recorded 39 species of ferns which are divided into 17 families, 6 ordo, and 3 classes. The dominant percentage can be seen from the highest INP calculation, namely P. Biaurita and S. Ornate, while the exposed INP values are A. Evecta and G. Verrucosum. The index of plant diversity in Curug Winong is of moderate criteria. The results of the validation of material experts (77.78%), media (96.13%), teacher responses (91.67%), overlap test (83.66%), and student responses (83.90%). These results indicate that the E-booklet that was developed is suitable to be used as a learning resource
Transformasi genetik Nicotiana benthamiana L. dan kedelai dengan Gen MaMt2 penyandi Metallothionein Tipe II dari Melastoma malabathricum L.
The low pH and high concentration of aluminum (Al) are major limiting factors for crops production. The capability of crops to cope those stresses can be improved by expressing genes controlling adaptation to low pH and high concentration of Al. One of these genes was MaMt2 encoding for metallothionein type II which isolated from Melastoma malabathricum. The objectives of the research were 1) to construct overexpression vector of MaMt2 gene with pIG6 plasmid, 2) to transform Nicotiana benthamiana and soybean with MaMt2 gene, and to analyze the transgenic plants. The MaMt2 gene were successfully fused with the strong promoter pUbiquitin in the pIG6 plasmid, and the recombinant plasmid was introduced into Escherichia coli DH5α by electroporation method. The pIG6-MaMt2 recombinant vector was successfully introduced into A. tumefaciens LBA4404 by triparental mating method (TPM). Genetic transformation was performed by co-cultivating A. tumefaciens LBA4404-pIG6-MaMt2 with N. benthamiana leaf explants and half seed explants of soybean cv. Lumut. The T0 and T1 generations of transgenic plants were already obtained. The integration of MaMt2 transgene into the genom of T0 transgenic plants was confirmed by PCR. Segregation analysis in the T1 generation of N. benthamiana showed that hpt gene was inherited to the offspring in Mendelian pattern and all samples of transgenic plants were heterozygote containing one functional hpt gene. Transformation of cv. Lumut half seed explants with MaMt2 gene was succesful, based on PCR by UbiF and NosTR primer combination. PCR analysis of T1 generation of transgenic soybean showed that MaMt2 gene was transmitted into the offspring indicating that this transgene is integrated in the genome of transgenic soybean plants.Cekaman pH rendah dan kandungan aluminium yang tinggi pada tanah masam, khususnya tanah Ultisol, merupakan faktor-faktor pembatas produksi dan kualitas tanaman pangan. Kemampuan beberapa tanaman pangan penting dalam mengatasi toksisitas logam dapat ditingkatkan dengan cara mengekspresikan gen-gen yang terkait dengan toleransi pH rendah dan aluminium (Al) tinggi. Melastoma malabathricum merupakan salah satu tanaman yang mampu hidup di tanah ber- pH rendah dan kelarutan Al tinggi. Sejumlah gen yang ekspresinya diinduksi oleh cekaman pH rendah dan aluminium tinggi pada M. malabathricum sebagian sudah berhasil diisolasi dan dikarakterisasi. Salah satu gen tersebut adalah gen MaMt2 yang menyandi metallothionein tipe II. Protein ini banyak mengandung sistein yang mampu mengikat berbagai macam logam. Feasibility of The E-booklet Teaching Material Supplement on Fruit Fly (Bactrocera spp.) for High School Students on Biodiversity
This study aims to analyze the feasibility of fruit fly e-booklets (Bactrocera spp.) for grade X SMA students on biodiversity material. This study analyzes the validity, readability test, and response of e-booklets. The method used is Research & Development (R&D) with the development model used is ADDIE with three stages, namely analysis, design, and development. The feasibility of the e-booklet media used is determined based on validation from material experts and media experts. The response to the e-booklet is determined based on teacher and student responses. The readability test using the gap test was carried out in grade X. The results of the material validation got a percentage of 91.2% with a very valid category, media validation got a percentage of 91.6% with a very valid category, the readability test got a percentage of 98.7% with an independent level category, teacher responses got a percentage of 90%, and student responses got a percentage of 82% with a very good category. Based on the results of the study, it was concluded that the e-booklet media was very valid to use, positive responses were very good from teachers and students, and readability included an independent level or free which means easy to read
MAJALAH IT-FLY VA: ALTERNATIF PILIHAN SUMBER BELAJAR BIOLOGI
Penelitian ini bertujuan untuk menguji kelayakan majalah It-Fly va yang sengaja ditulis berdasarkan hasil penelitian tentang keanekaragaman lalat buah di Agro Cepoko sebagai alternatif pilihan sumber belajar biologi yang memuat pengetahuan mengenai keanekaragaman hayati, serangga, pengelolaan limbah, serta upaya dan teknologi pengendalian lalat buah. Metode penelitian yang digunakan adalah penelitian Research and Development (R&D) modifikasi dari Borg and Gall dan Allesi & Trollip melalui dua tahapan uji produk yaitu uji alfa dan uji beta. Uji alfa dalam penelitian ini dilakukan menggunakan lembar validasi penilaian kelayakan buku yang diadaptasi dari BSNP 2014 meliputi empat komponen kelayakan yaitu teknik penyajian, isi, kebahasaan, dan kegrafikan yang diujikan kepada ahli materi dan media. Uji beta dalam penelitian ini dilakukan untuk menguji produk dalam skala kecil. Uji beta dilakukan dengan menggunakan lembar validasi dan lembar tanggapan yang diujikan kepada pengguna yaitu guru dan siswa. Analisis hasil penelitian menggunakan uji alfa yang dilakukan kepada ahli materi dan media terhadap majalah mendapat kategori sangat layak pada aspek teknik penyajian, isi, dan kegrafikan. Pada aspek kebahasaan memperoleh kategori penilaian yang layak. Analisis hasil uji beta yang dilakukan kepada guru dan siswa menunjukkan tanggapan yang positif terhadap majalah melalui lembar tanggapan siswa. Dengan demikian dapat disimpulkan bahwa majalah It-Fly va yang ditulis berdasarkan hasil penelitian tentang lalat buah di Agro Cepoko memperoleh kriteria penilaian buku berdasarkan standar BSNP dengan kategori sangat layak digunakan sebagai alternatif pilihan sumber belajar biologi untuk siswa.Kata kunci: majalah, sumber belaja
The Readiness of Biology Laboratories to Support Practical Work in State High Schools throughout Tegal Regency
This study aimed to analyze the readiness of biology laboratories to support practical work in State High Schools throughout Tegal Regency. The readiness refers to the availability of all things that can support practical activities including the readiness of infrastructure facilities and laboratory management. This study used quantitative descriptive research design. The population in this study is all independent biology laboratories in State High Schools throughout Tegal Regency that used for practical works. The sampling technique used is saturation sampling. This study has been done at biology laboratory of SMA Negeri 1 Bojong, SMA Negeri 1 Kramat, SMA Negeri 1 Warureja, SMA Negeri 1 Balapulang, SMA Negeri 1 Pangkah, SMA Negeri 1 Slawi, SMA Negeri 2 Slawi, SMA Negeri 3 Slawi, dan SMA Negeri 1 Pagerbarang. The data were obtained using the observation method, questionnaire, and interview with reference to Permendiknas Number 24 Year 2007, Permendiknas Number 26 Year 2008, and Permendikbud Number 24 Year 2016. Further, the data were analyzed using descriptive statistics. The result of the study shows that the readiness of infrastructure facilities and management successively scored 58.5% and 62.8% which is belonged to sufficient criteria. The conclusion of this study is the biology laboratories in State High Schools throughout Tegal Regency is ready enough to support biological practical work with an average readiness of 60.52%
PERUBAHAN KARAKTER KUANTITATIF MUCUNA PRURIENS GENERASI M1 PASCA IRRADIASI SINAR GAMMA CO-60
Koro benguk (Mucuna pruriens) berpotensi sebagai bahan substitusi kedelai. Keragaman genetik koro benguk termasuk kategori rendah, sehingga perlu ditingkatkan keragaman genetiknya terutama pada faktor-faktor produksi. Tujuan penelitian adalah mendapatkan informasi mutasi mikro pada koro benguk generasi M1 setelah diradiasi dengan sinar gamma dari Cobalt-60. Benih koro benguk diperoleh dari Balai Pengembangan Perbenihan Tanaman Pangan dan Hortikultura (BP2TPH) Ngipiksari, Sleman, Yogyakarta. Radiasi sinar gamma dilakukan di PAIR BATAN Pasar Jumat, Jakarta. Dosis radiasi gamma yang digunakan adalah 0; 25 Gy, 50 Gy; 100 Gy; 150 Gy, 200 Gy, dan 250 Gy. Eksperimen dilakukan menggunakan Rancangan Acak Kelompok (RAK). Parameter yang diamati adalah umur berbunga, jumlah polong, jumlah biji, berat 100 biji tanaman generasi M1. Data dianalisis menggunakan Anava, dilanjutkan dengan Uji Beda Nyata Duncan (DMRT). Hasil penelitian menunjukkan bahwa peningkatan dosis radiasi sinar gamma akan memperpanjang hari munculnya bunga dan berat biji, namun menurunkan jumlah polong dan jumlah biji pada tanaman generasi M1
Pendekatan Inovatif dalam Manajemen Ternak: Analisis Perbandingan Good Farming Practices (GFP) dan Inseminasi Buatan (IB) Berbasis Biologi Molekuler: Innovative Approaches in Livestock Management: A Comparative Analysis of Good FarmingPractices and Molecular Biology Based Artificial Insemination
Livestock farming is an important sector in meeting the food needs of the community, especially for animal protein from meat and dairy products. Although livestock farming has great potential, livestock productivity still faces significant challenges, including inadequate husbandry systems, technological limitations, and a lack of understanding of reproductive management. In an effort to improve productivity, two approaches that can be applied are Good Farming Practices (GFP) and Artificial Insemination (AI) based on molecular biology. GFP includes principles of feed management, sanitation, livestock health, and a good environment, which are expected to improve livestock welfare and the quality of production results. On the other hand, IB uses advanced technology to improve the genetic quality of livestock and enhance reproductive efficiency, particularly with the integration of molecular biology techniques. This study aims to compare the effectiveness of both approaches in improving livestock productivity. By applying GFP and IB synergistically, it is hoped that the increasing market demand can be met without neglecting animal welfare aspects
TINJAUAN PUSTAKA SISTEMATIS: KARAKTERISASI KEANEKARAGAMAN GENETIK SPESIES JERUK (Citrus sp.) MENGGUNAKAN PENANDA SIMPLE SEQUENCE REPEAT (SSR)
Background: The biodiversity of citrus plants is very abundant and many are cultivated, creating various types. However, the high demand makes orange farmers feel overwhelmed. This is what causes some orange cultivars to become rare. Characterization of the genetic diversity of citrus germplasm at the molecular level is an important and crucial step that can assist in the breeding and conservation process. The aim of this literature review is to provide information regarding molecular markers regarding the application of SSR molecular markers to analyze the genetic diversity of citrus.
Method: The method used is a systematic literature review with the literature sources used coming from articles taken from national journals and international journals indexed by Scopus.
Results: The application of molecular markers using the SSR method is useful for identifying and characterizing the genetic diversity of citrus germplasm so that it can assist in breeding and conservation actions. SSR has many advantages both in terms of its use and the results obtained.
Conclusion: One application of molecular markers is to analyze plant genetic diversity. Simple Sequence Repeat (SSR) molecular markers can help to analyze the characteristics and genetic diversity of oranges so that they can assist in plant breeding and conservation purposes
The Influence of Learning Cycle Model Against the Motivation of Learning and Critical Thinking Skills of Students
This research aims to know the influence of motivation towards Learning Cycle of learning and critical thinking skills of students on the material of the human digestive system in SMP Negeri 03 Jekulo. The sample used is grade VIII VIII C and D taken with the technique of convenience sampling and research design a non-randomized control group pretest-posttes design. Data obtained in the form of motivation and critical thinking skills as well as the response of teachers and students towards learning Learning Cycle model. Data analysis using SPSS 16.0. The research results obtained studying motivations of students after learning Learning Cycle has increased from 76.87 with higher category be the category with very tinngi 84.67. Results the average critical thinking skills has increased from 76.34 with critical categories into classes with 82.11 wants a category of highly critical. The t-test based on posttest showed thitung > 11.628 from ttabel 2.073 which means there is a significant difference. The response of the students towards the Learning Cycle shows interest because of the interesting activities and serves the appropriate issues with everyday life. The teacher gave a positive response, namely students became active in learning, so that students acquire the knowledge of the concept of direct experience of the menjadiikan pengetahuaanya longer remember. The conclusions of this research are the Learning Cycle model provides a positive influence motivation towards learning and critical thinking skills on the material of the human digestive system in SMP Negeri 3 jekulo
