GAYA KOMUNIKASI DAKWAH NING UMI LAILA PADA LIVE COMAL PEMALANG
Communication style is a way for a person to convey messages verbally and non-verbally in the hope that it will be conveyed well to the recipient of the message. In receiving da'wah messages, the communication style used by a da'i is considered important to attract the attention of the mad'u so that the da'wah message is well received by the mad'u. The problem in this study is how the communication style used by Ning Umi Laila in the live Comal Pemalang.
The method used in this study is qualitative descriptive, using Norton's communication style which will produce descriptive data that is emphasized as deeply as possible through the theory of the communication style.
The results of this study provide an answer to the problem that the author found several communication styles used by Ning Umi Laila in delivering her lectures. This is in line with the 10 communication styles according to Norton that are applied in the parts of Ning Umi Laila's lectures, namely friendly communication style, animated style, dominant style, dramatic style, open style, argumentative style, relaxed style, attentive style, precise style, and memorable leaving style. Furthermore, for the intonation used by Ning Umi Laila in delivering her lecture using these communication styles, namely dynamic intonation, tempo intonation, and high-pitch intonation in the context of cheerfulness
Polyhydroxyalkanoate production by a novel bacterium Massilia sp UMI-21 isolated from seaweed, and molecular cloning of its polyhydroxyalkanoate synthase gene
We successfully isolated one microorganism (UMI-21) from Ulva, a green algae that contains starch. The strain UMI-21 can produce polyhydroxyalkanoate (PHA) from starch, maltotriose, or maltose as a sole carbon source. Taxonomic studies and 16S rDNA sequence analysis revealed that strain UMI-21 was phylogenetically related to species of the genus Massilia. The PHA content under the cultivation condition using a 10-L jar fermentor was 45.5% (w/w). This value was higher than that obtained after cultivation in a flask, suggesting the possibility of large-scale PHA production by UMI-21 from starch. A major issue for the industrial production of microbial PHAs is the very high production cost. Starch is a relatively inexpensive substrate that is also found in abundant seaweeds such as Ulva. Therefore, the strain isolated in this study may be very useful for producing PHA from seaweeds containing polysaccharides such as starch. In addition, a 3.7-kbp DNA fragment containing the whole PHA synthase gene (phaC) was obtained from the strain UMI-21. The results of open reading frame (ORF) analysis suggested that the DNA fragment contained two ORFs, which were composed of 1740 (phaC) and 564 bp (phaR). The deduced amino acid sequence of PhaC from strain UMI-21 shared high similarity with PhaC from Ralstonia eutropha, which is a representative PHA-producing bacterium with a class I PHA synthase. This is the first report for the cloning of the PHA synthase gene from Massilia species
Production of poly(3-hydroyxybutylate) by a novel alginolytic bacterium Hydrogenophaga sp. strain UMI-18 using alginate as a sole carbon source
A novel alginolytic bacterium Hydrogenophaga sp. strain UMI-18 that produces poly(3-hydroyxybutylate) (PHB) in the alginate-mineral salt (AMS) medium containing 1% (w/v) sodium alginate as a sole carbon source was isolated from a decayed brown seaweed litter. The yield of PHB produced by strain UMI-18 was 1.1 +/- 0.15 g/L of AMS and the PHB content in dried cell pellet was 58 +/- 4% (w/w). Glucose, fructose, galactose, mannose, mannitol, sucrose and lactose were also available for the production of PHB by strain UMI-18. The yield of PHB in 1% (w/v) carbohydrate media reached 2.03-2.24 g/L for glucose and fructose, 0.75-1.64 g/L for alginate, galactose, mannitol and sucrose, and similar to 0.15 g/L for lactose. The PHB produced by strain UMI-18 showed a glass-transition temperature (T-g) at 4 degrees C, a melting temperature at 175 degrees C, and an average molecular mass of 860 kDa. Draft genome analysis of the strain UMI-18 revealed that an alginate-assimilating gene cluster is located in contig 8 comprising 453,520 bp and the PHB-synthesis gene cluster is in contig 15 comprising 653,793 bp.
