    No full text
AbstractPalm oil of Elaeis guineensis Jacq. contains approximately 50% saturated and 50% unsaturated fatty acids. In the composition of saturated fatty acid, palmitic acid is the most abundant which is predominantly around 44% of the total fatty acids. The high level of accumulation of palmitic acid should be reduced in order to get a better dietary fatty acid and a more nutritional value oil in the market. The purpose in reducing palmitic acid content could be carried out by silencing palmitoyl-ACP thioesterase (PATE) gene activities. Primer pairs to amplify partial PATE fragments, intron and 3Untranslated Region (3UTR) were designed from PATE of Elaeis guineensis var tenera (GenBank DQ422858). PATE fragments, intron and 3UTR were constructed in two expression cassettes with inverted repeat (IR) and intron-spliced inverted repeat (ISIR) orientations to generate hair-pin RNAs (hpRNA). Combinations of restriction enzymes (RE) were used to confirm, the size, orientation and location of the inserts

    No full text
Kata Kunci: Pengolahan hasil kelautan, Muara Angke, waterfront./ Indonesia is the largest maritime country in the world with two-thirds of its territory consisting of waters. Indonesia with ± 18,000 islands, about 5.8 million km2 of sea, and 81,000 km of beaches. The sea in Indonesia is quite wide and the marine products are quite diverse. Samudera Indonesia produces many types of shrimp, large pelagic fish (Tuna, Shark, etc.), with a production of 1,076,890.00 tons/year. Indonesia must have a good maritime strategy. To visit the World Maritime Axis, several things must be considered, namely, marine spatial planning as well as infrastructure development and maritime connectivity. To support this government plan, the author wants to develop the Muara Angke Fish Market which is a Marine Product Processing Center in Muara Angke with a Waterfront Theme Approach. Muara Angke is a fairly large supplier of seafood in Jakarta. The sea products do not only come from the catch of local fishermen, but also from fishermen from other areas. Muara Angke is a center for trading marine products, therefore the development of marine product processing is able to encourage the growth of investment and marine business, various marine products and various processing are able to make Muara Angke a Center for Seafood Processing. Its location by the sea is also a potential for the implementation of the Waterfront theme. Keywords: Marine product processing, Muara Angke, waterfront

AbstractOil palm ((Elaeis guineensis Jacq) is one of the most productive oil crops in the world. It can produce oil up to 90% of its dry weight and has the highest oil content reported among other plant tissues. WRI1 gene is one of many contributing genes that control oil content in oil palm fruit. Despite its obvious scientific and economic interest, literature and molecular resources about WRI1 gene in oil palm are still limited. The objective of this research was to isolate sequence and characterize the WRI1 gene from oil palm genome. WRI1 gene was successfully amplified from oil palm cDNA library. The amplification produced a single DNA fragment of 1 000bp and which was cloned into pJET cloning vector for sequencing. The sequencing result showed two clones that were identified as WRI1 transcription factor

AbstractJatropha curcas oil seed is used as an alternative energy to replace fossil fuel. J. curcas genome can be modified by adding desired gene using Agrobacterium transformation to develop J. curcas plant quality. The explants regeneration was carried out in tissue culture solid medium. However, established tissue culture method may not give the same result due to genetic variation from the explants. The media optimization was done by combining IBA and BAP for callus induction and combination of IBA, BAP, and GA3 for shoot induction. The most optimum media for Jatromas callus induction was BIL and SIG for shoot induction

    No full text
AbstractThe predominant fatty acids of jatropha oil are oleic acid (42.02%), linoleic acid (35.38%), palmitic acid (14.54%), and stearic acid (6.3%). Ratio of fatty acids content of jatropha oil, especially for oleic acid (42.02%) compare to linoleic acid (35.38%) is approximately equal. The oxidation stability of jatropha oil showed that methyl oleate is around 2.79h and methyl linoleate is 0.94h. Therefore, the presence of methyl linoleate will decrease the value oxidation stability of jatropha oil. The objective of the research was to modify fatty acid composition, especially to increase oleic acid and to reduce linoleic acid content of jatropha oil. To achieve the objective construction of transformation vectors for Δ 12 fatty acid desaturase FAD2 gene silencing was designed. In two transformation vectors, FAD2 gene fragment was constructed in inverted repeat (IR) and intron-spliced inverted repeat (ISIR) orientations to generate hair-pin RNAs (hpRNA)

AbstractTissue culture technique has been used to produce elite oil palm clonal materials in commercial scale. Proliferation of embryogenic callus in oil palm commonly utilizes solid and liquid medium, but the result are still not optimum. Recently, researchers have developed an improved technique called Temporary Immersion System (TIS) and this technique has shown great potential to increase proliferation rate of the embryogenic callus. This research can be divided into two groups i.e. selection of liquid medium using shaker to increase the proliferation rate and proliferation of embryogenic callus using TIS. The objectives of this research are to obtain the best medium for increasing embryogenic proliferation rate using shaker and to obtain the best medium and time interval of immersion to increase the proliferation rate using TIS. Approximately 0.3g of embryogenic callus from embryoid-line A1 were weighted and cultured in twenty two types of liquid medium for the first research and two types of medium with the composition of MSD and MSA for the second research. The proliferation rate based on fresh weight obtained from week 8 observations. The data was analysed with Anova of F (α = 5%) and then with DMRT in α = 5%. TIS observation was done every 1h, 3h, 6h of immersion time with time interval of 3min. The results showed that the best medium composition to increase proliferation of embryogenic callus using shaker was MSA9, the best medium composition to increase proliferation of embryogenic callus using TIS was MSD, and the best time interval of immersion was every 3h for 3min
