    Studi Potensi Ekstrak Brucea Javanica Sebagai Bioaktif Antikanker Payudara Terhadap Sel T47d

    Research. Buah makasar (Brucea javanica) known has anticancer activity.This research would bioassay the anticancer activity ofBrucea javanica extract toward T47D breast cancercell by in-vitro method, using SRB dyed. In order to study the mechanism, a dot blotting method was carried out to observe the interaction between the Brucea javanica extract with the chromosomal DNA molecule of T47D Cell Line. The interaction results were analyzed with High Performance Liquid Chromatography (HPLC) methods.The result of anticancer activity assay toward T47D cell showed that Brucea javanica extract has activity with IC50 2.69 g/mL, whereas, the interactionresult was analyzed with High Performance Liquid Chromatography (HPLC) methods, exhibited the reduction peaks with the retention time 2.26; 3.63 and 3.94 minutes.     Molecular Docking of Robustaflavone Derivatives on Pi3k Protein Target and The Correlation With Its Cytotoxic Activity on Cancer Cells

    Robustaflavone derivatives have cytotoxic effect on cancer cell. Inhibition of PI3K on cancer cell has correlation with apoptotic induction. The aim of this research is to observe interaction from robustaflavone derivatives on PI3K receptor that may give contribution to the cytotoxic effect of the compounds on cancer cell line. Geometric optimization of robustaflavone derivatives structured was done used Hyperchem 7.5 software. Arguslab 4.01 software with GAdock method applied for the docking step. The docking result showed that robustaflavone 7,4',7''- trimethyl ether (RTE) has affinity on PI3K target receptor which gave the lowest Gibbs free energy (∆G -10,855 kcal/mol).Keywords: robustaflavone, PI3K, cancer, dockin

    Tumbuhan Erythrina variegata (Leguminosae) secara tradisional dikenal oleh masyarakat Indonesia sebagai obat antikanker dan antimalaria. Bagian tumbuhan ini yang biasa digunakan sebagai bahan pengobatan adalah daun dan kulit batang, tapi kandungan senyawa kimia aktif biologisnya belum banyak dilaporkan. Tujuan penelitian ini adalah untuk mengungkapkan senyawa aktif antikanker dan antimalaria secara in vitro yang terdapat di dalam daun dan kulit batang E. variegata. Penelitian dilakukan dengan cara ekstraksi metanol dan fraksionasi dari daun dan kulit batang E. variegata yang dipandu dengan lIji antikanker secara in vitro terhadap sel kanker payudara T47D menggunakan metode Sulforodamin B (SRB) dan uji antimalaria secara in vitro terhadap Plasmodiumfalciparum 307 (sensitif klorokuin) dan Kl (resisten klorokuin) menggunakan metode laktatdehidrogenase (LDH). S-elanjutnya dilakukan pemisahan fraksi etil asetat daun dan kulit batang E. variegata yang dipantau dengan uji antikanker danmtimalaria secara in vitro menggunakan kombinasi kolom kromatografi diperoleh tiga senyawa aktif (1, 2, dan 3). Struktur kimia senyawa aktif (1, 2, dan 3) ditetapkan berdasarkan data-data spektroskopi dan diidentifikasikan sebagai turunan triterpenoid pentasiklik glikosida (1)flavonoid, eristagallin A (2)dan steroid, (22E)-5a,8a-epidioksiergosta-6,22-dien-3&#946ol (3). Senyawa (1) mcnunjukkan aktivitas antimalaria secara in vitro terhadap P.falciparum strain 307 (sensitif klorokuin) dengan ICso 1,8 I1g/mL dan terhadap strain KI (resisten klorokllin) dengan lC&#8325&#83203,3 µg/mL.Senyawa (2) dan (3) menunjukkan aktivitas antikanker secara in vitro terhadap sel kanker payurada T47D dengan IC&#8325&#8320masing-masing 3,0 dan 3,2 µg/mL.Tumbuhan E. variegata mempunyai potensi sebagai bahan dasar obat herbal antikanker payudara dan antimalaria

ABSTRACT Erythrina variegata (Leguminosae) plant used traditionaly as plant of  anti-cancer and anti-malarial in Indonesia. The leaves and stem bark of E. variegata used as medicinal folk of anti-cancer and anti-malarial, however haven’t reported yet of bioactive compounds. The purpose of this research was assayed an anti-cancer and anti-malarial compounds toward breast cancer cell-lines T47D and toward Plasmodium falciparum 3D7 (chloroquine sensitive) and K1 (chloroquine resistance) in vitro from E. variegata. The research was extraction of methanol and fractionation from the leaves and stem bark of E. variegata by using guide-assay in vitro Sulphorhodamine B (SRB) method and lactate dehydrogenase (LDH). Furthermore, by using the anti-cancer and anti-malarial activity to follow separation, the active fraction was separated by combination of column chromatography to yield three active compounds (1-3). The chemical structure of active compounds (1-3) were determined on the basis of spectroscopic evidences and comparison with those previously reported and identified as terpenoid pentacyclic glycoside (1), flavonoid, erystagallin A (2) and steroid, (22E)-5α,8α-epidioxyergosta-6,22-diene-3β-ol (3). The compound (1) showed anti-malarial activity in vitro against P. falciparum strain 3D7 and K1 with IC50 1.8 and  3.3  µg/mL, respectively.  The compounds (2-3) showed anti-cancer activity against of breast cancer cell-lines T47D with IC50 of 3.03and 3.2 µg/ml, respectively. This results strongly suggested that E. variegata is a promising sources of anti-cancer and anti-malarial agents

    Rumput mutiara merupakan tanaman yang berkhasiat sebagai obat tradisional, tanaman ini termasuk kedalam famili tumbuhan Rubiaceae. digunakan sebagai obat alternatif penyebab penyakit infeksi yang disebabkan karena masuknya mikroorganisme kedalam tubuh. Salah satu mikroorganisme yang mengakibatkan penyakit ini adalah bakteri ( Amonim, 1993). Antibakteri adalah senyawa kimia yang dapat membunuh atau memghambat pertumbuhan mikroorganisme penyebab penyakit dengan kerusakan minimal jaringan induknya. Dilakukan penelitian aktivitas antibakteri dari ekstrak rumput mutiara (Hedyotis corimbosa (L.) Lamk) terhadap pertumbuhan beberapa bakteri, yang dlilakukan melalui 4 tahapan yaitu tahapan maserasi, tahapan ekstraksi fraksinasi, tahap pengujian fitokimia, dan diteruskan dengan tahap pengujian aktivitas antibakteri menggunakan metode difusi agar, parameter yang digunakan adalah dengan melihat zona terang yang dihasilkan (zona yang tidak terjadi pertumbuhan). Hasil penelitian menunjukkan bahwa dalam ekstrak rumput mutiara terdapat zat aktif yang berfungsi sebagai antibakteri yang terdapat dalam fraksi metilen klorida, pada fraksi ini aktivitas paling baik dihasilkan oleh bakteri S. Disentry dengan diameter daerah hambatan sebesar 27,5 mm

    Drug discovery for cancer medication is the most important effort that researcher do at this time. Indonesia bio diversities have possibility as a cancer medicine sources. Finding a herbal medicine for cancer treatment is a first step to find a right cancer medicine in the future. This research has already completed for the earlier another research. Some fractions of Hedyotis corymbosa extract has been analyzed using Sulforhodamine B method with UV wavelength 515 nm against T47D cell line, a human breast cancer. There are Hexane extract, Methylene chloride extract and Ethyl acetate extract, and give inhibitory concentration 50 (IC50)  of 33.45 µg/ml, 54.59 µg/ml and 52.58 µg/ml, respectively. Ethanolic extract, itself has IC50 of 61.57 µg/ml, whereas IC50 value of Cisplatinum is  9.63 µg/ml.     The calcium deficient rat is condition of rat with calcium deficient in its body, one reason of this condition is intake with calcium deficient diet. The aim of this study was to analyze the effect of calcium deficient diet in premenopausal age female rat model of strain Sprague dawley. Research methods consist of: formulation of purified diet and calcium deficient diet, preparation of calcium deficient rat model strain Sprague dawley of premenopausal age; parameters measured were: consumption of dry matter; consumption of calcium, calcium concentration in feces, liver, kidney, and bone; calcium absorption; serum calcium concentration; progesterone hormone concentration; and daily body weight gain. Treatment with calcium deficient diet for 12 weeks could reduce serum calcium concentration into 7.7

    P-glycoprotein (P-gp) resistance in cancer cells decreases intracellular accumulation of various anticancer drugs. This multidrug resistance (MDR) protein can be modulated by a number of non-cytotoxic drugs. We have screened 30 chincona alkaloids derivatives as a potent P-gp inhibitor agent in silico. Hereby, we report the highest potential inhibitions of P-gp is Cinchonidine isobutanoate through molecular docking approach. with affinity energy -8.6 kcal/mol and inhibition constant, Ki is 4.89 x 10-7 M. Cinchonidine isobutanoate is also known has molecular weight below 500, Log P value 3.5, which is indicated violation free of Lipinski`s rule of five. Thus, Cinchonidine isobutanoate is the most potent compound as anticancer compare to other Cinchona alkaloids. Ultimately, we design Cinchonidine isobutanoate for further lead synthesis by using DBSA, act as a combined Brønsted acid-surfactant-catalyst (BASC) to obtain high concentration of organic product by forming micellar aggregates which is very powerful catalytic application in water environment

    Anticancer Activity of Ethanolic Extract of Selaginella Plana Hieron. on T47d Cell Line in Vitro

    Selaginella sp belongs to the Selaginellaceae family. It has been used in China and Indonesia as a traditional medicine. It hasseveral medicinal properties including antibacterial, anticardiovascular, and anticancer agent. The aim of the present study was to access the anticancer property of the ethanolic extracts of Selaginella plana Hieron. on T47D breast cancer cell line. The proliferation of T47D cell line was detected by SRB (Sulforhodamine B) assay which was measured at a wavelength of 515 nM. The result showed that the IC50 of the ethanolic extract was determined at 7.03 pg/mL. This significant activity was assumed due to its high total flavonoid content. The totalflavonoid content of the ethanolic extract was 23.04%. Flow cytometry analysis indicated that the extract may undergo the cell death via apoptosis pathway. In conclusion, the ethanolic extract of Selaginella plana Hieron: has considerable activity in inhibiting T47D cell line proliferation
