Genetic variability of Akhal-Teke horses bred in Italy
Background. The Akhal-Teke horse (AKH) is native of the modern Turkmenistan area. It was introduced in Italy from 1991 to 2000 mainly as an endurance horse. This paper characterizes the genetic variability of the whole Italian AKH horse population and evaluates their inbreeding level by analyzing microsatellite markers and mitochondrial
D-Loop sequences. Methods. Seventeen microsatellite marker loci were genotyped on 95 DNA samples from almost all the AKH horses bred in Italy in the last 20 years. Standard genetic variability measures (Ho, He, FIS) were compared against the same variables published on other eight AKH populations. In addition, 397 bp of mtDNA D-loop region were sequenced on a sub-group of 22 unrelated AKH out of the 95 sampled ones, and on 11 unrelated Arab horses. The haplotypes identified in the Italian population were aligned to sequences ofAKH(56), Arab (five), Caspian Pony (13), Przewalskii (two) and Barb (15) horses available in GenBank. total 126 sequences. Results. Nucleic markers showed a high degree of polymorphism (Ho D 0.642; He D
0.649) and a low inbreeding level (FIS D 0.016) in Italian horses, compared to other AKH populations (ranged from 0.103 AKH from Estonia to 0.114 AKH from Czech Republic). High variability was also recorded in the D-Loop region. 11 haplotypes were identified with haplotype diversity (hd), nucleotide diversity () and average number
of nucleotide differences (k) of 0.938, 0.021 and 6.448, respectively. When all the 126 D-Loop sequences were compared, 51 haplotypes were found, and four were here found only in the Italian AKH horses. The 51 haplotypes were conformed to eight recognized mtDNA haplogroups (A, C, F, G, L, M, P and Q) and confirmed by MJN analysis, Italian horses being assigned to five haplogroups (A, C, G, L and M). Using a PCA approach to the same data, the total haplotypes were grouped into two clusters including A+C+M+P and G+F haplogroups, while L and Q haplogroups remained ungrouped. Identification of Solute carrier family 5 member 9 (SLC5A9) as biomarker of in vivo and in vitro oocyte developmental competence
The transcriptome associated with the ability of oocytes to support embryogenesis and development remains unclear. The aim of this study was to compare the mRNA expression pattern between oocytes collected from fertile heifers (H) and repeat breeders (RB) using RNAseq. Oocytes were collected by ovum pick-up from Italian Simmental (IS) and Holstein Friesian (HF) heifers that became pregnant at the following oestrus, and from cows of the same breeds classified as RB after they failed to become pregnant for a minimum of 3 consecutive AI. Oocytes from each animal (5 for HF_RB, 3 each for all other groups) were pooled and sequenced using Illumina HiSEqn 2000 (Illumina Inc., San Diego, CA, USA). Principal component analysis showed that HF oocytes clustered away from those of IS, irrespective of the fertility status. However, within breeds, H and RB always clustered apart from each other. This suggests that differences in the transcriptional blueprint between breeds was more prominent than differences related to fertility. However, these were still strong when considering breeds separately, therefore we treated breeds as independent groups and performed differential expression analysis on H v. RB in IS and HF. DESEqn 2 identified 135 differentially expressed genes when we compared H and RB in IS and 2124 in HF. This difference in number of DE genes between the 2 breeds reflects the fact that the variance between H and RB transcriptional patterns in HF is larger than in IS. We looked for genes that were differentially and consistently expressed between H and RB in both breeds and identified 10 genes: histone H2B, exo/endonuclease G, solute carrier family 5 member 9, BH3 interacting domain death agonist, TBK1 binding protein 1, purine-rich element binding protein A, tRNA-yW synthesising protein 1 homologue, Jun oncogene, FRA10AC1 homologue, and SKI oncogene. To validate their potential use as biomarkers, we performed a preliminary in vitro validation by RT-qPCR. As described in the literature, oocytes collected from ovaries with more than 10 follicles of 2 to 6 mm were classified as high quality, while those collected from ovaries with fewer than 10 follicles were classified as low quality (Gandolfi et al. 1997 Theriogenology 48, 1153-1160). Gene expression analysis confirmed SLC5A9, a membrane-bound protein involved in sodium-dependent transport of glucose, as a potential biomarker to discriminate between a competent and incompetent oocyte. Altogether, our data provide a detailed overview of the mRNA pattern defining the differences between a competent and incompetent oocyte in vivo, and identified a new potential marker able to differentiate fertile and infertile oocyte both in vivo and in vitro

Está constituído a partir de uma visão crítica da História, o que permite que ?outras imagens?, fragmentárias e não monumentais, também tenham espaço. Em diálogo com o princípio da montagem, este esboço apresenta-se em duas partes. Na primeira, Imagens possíveis, estão citadas as imagens elaboradas em vida e post mortem acerca do austríaco-brasileiro que nasceu em Viena em 1900, se exilou no Brasil em 1939 e morreu no Rio de Janeiro, em 1978. Na segunda, Montagens possíveis, apresentam-se duas possibilidades de exercício biográfico: pela leitura alegórica do documentário O velho e o Novo (Otto Maria Carpeaux), entendido como instrumento de intervenção no contexto ditatorial brasileiro e de uma reelaboração biográfica concernentes às suas experiências europeias; e pelo Caderno de imagens críticas, registro dos encontros em Carpeaux pelo meio de imagens críticas produzidas a partir da cesura do presente.Abstract : This biographical sketch attempts to quote some images of Otto Maria Carpeaux: various types of biographical constructions, carried out in different contexts by disparate authors under conditions just as distinct. It stems from a critical view of history, allowing for ?other images? fragmented and non-monumental ? to share the space.In dialogue with the montage principle, this sketch has two parts. The first, Possible Images, quotes the images produced during and after the life of the Austrian-Brazilian, who was born in Vienna in 1900, went to Brazil in exile in 1939 and died in Rio de Janeiro in 1978. Genome-wide association study between CNVs and milk production traits in Valle del Belice sheep
Copy number variation (CNV) is a major source of genomic structural variation. The aim of this study was to detect genomic CNV regions (CNVR) in Valle del Belice dairy sheep population and to identify those affecting milk production traits. The GO analysis identified possible candidate genes and pathways related to the selected traits. We identified CNVs in 416 individuals genotyped using the Illumina OvineSNP50 BeadChip array. The CNV association using a correlation-trend test model was examined with the Golden Helix SVS 8.7.0 tool. Significant CNVs were detected when their adjusted p-value was <0.01 after false discovery rate (FDR) correction. We identified 7,208 CNVs, which gave 365 CNVRs after aggregating overlapping CNVs. Thirty-one CNVRs were significantly associated with one or more traits included in the analysis. All CNVRs, except those on OAR19, overlapped with quantitative trait loci (QTL), even if they were not directly related to the traits of interest. A total of 222 genes were annotated within the significantly associated CNVRs, most of which played important roles in biological processes related to milk production and health-related traits. Identification of the genes in the CNVRs associated with the studied traits will provide the basis for further investigation of their role in the metabolic pathways related to milk production and health traits
