    Ganyong merupakan jenis umbi-umbian yang dapat dimanfaatkan sebagai bahan pangan. Olahan ganyong berupa pati digunakan sebagai bahan alternatif pengganti terigu. Penelitian ini bertujuan untuk mengetahui manfaat lain dari pati ganyong dalam pembuatan ragam hias pada kain. Metode penelitian yang digunakan adalah eksperimentatif yakni dengan melakukan eksperimen secara langsung pada olahan pati ganyong yang dibuat pasta untuk selanjutnya digunakan sebagai bahan perintang warna pada kain.  Hasil penelitian menujukan bahwa pati ganyong dapat menahan warna masuk ke dalam serat kain. </p

    Characterization of Salmonella isolates from beef cattle, broiler chickens and human sources on Prince Edward Island

    Non-typhoid Salmonella serovars remain a potential threat to human health, and beef cattle and broiler chickens are possible sources of these organisms on Prince Edward Island (PEI). In this study, the ceca of beef cattle belonging to fasted and non-fasted groups, and broiler chickens were examined for Salmonella at the time of slaughter. The characteristics of the isolates, including antimicrobial resistance patterns and virulence genes, were studied along with the isolates obtained from cases of human salmonellosis on PEI during the study period (1996-97). The prevalence of Salmonella in beef cattle was 4.6% (11/240). The rate was significantly higher in fasted cattle (7.46%), than in non-fasted cattle (0.94%). The prevalence rate in chickens was 32.5% (39/120). In beef cattle, Salmonella typhimurium phage type (PT) or definitive type (DT) 104 which was resistant to ampicillin, chloramphenicol, streptomycin, sulfisoxazole and tetracycline, was the most predominant type (64%). In chickens, S. heidelberg, with resistance to gentamicin, streptomycin and sulfisoxazole, predominated. Of 26 isolates from humans, the most common serovar was S. typhimurium, including a multidrug-resistant strain of DT104. Examination by PCR revealed presence of the virulence gene invA in all serovars, and the spvC gene in all S. typhimurium isolates, of both beef cattle and human origin. Among the other serovars the latter gene was found in 7 human isolates, but in none of the chicken or beef isolates. All but 3 of the spvC-positive isolates possessed a 90 kilobasepair (kbp) plasmid suggesting that the 3 isolates had the spvC gene on their chromosome.     Genetic diversity of food originated Salmonella isolates

    Currently, Salmonella enterica is the most common bacterial foodborne pathogen, causing serious extraintestinal disease. Typing methods play an important role on pathogens’ source tracking, knowing the source(s) of bacteria in pharmaceutical sciences, preventing and controlling the diarrhea and food-poisoning outbreaks. The purpose of this study is to use different moleculer typing methods to determine the genetic variability of 38 foodborne Salmonella isolates that were previously identified by biochemical tests. The methods were evaluated by four molecular techniques including 16S rRNA sequencing, PFGE, PCR-RFLP and invA-spvC PCR. 16S rRNA sequencing results showed that four of the 38 isolates were Escherichia coli, Proteus mirabilis and Citrobacter murliniae, and the others were Salmonella enterica. Thirty-eight strains were subtyped by XbaI-PFGE into 20 profiles with different clusters, while they were subtyped by 16S rRNA-RFLP into 9 profiles with a single cluster. Out of two Salmonella isolates, the invasion gene (invA) was detected in all other Salmonella isolates (94%) and the virulence gene (spvC) was detected in 11% of Salmonella isolates. Our results suggested that the PFGE subtyping is the prominent method for the evaluation and benchmarking of molecular subtyping. © 2018 The Author(s).     SYBR®Green qPCR Salmonella detection system allowing discrimination at the genus, species and subspecies levels

    In this work, a three-level Salmonella detection system based on a combination of seven SYBR®Green qPCR was developed. This detection system discriminates Salmonella at the genus, species and subspecies levels using a single 96-well plate. The SYBR®Green qPCR assays target the invA, rpoD, iroB and safC genes, as well as the STM0296 locus, putatively coding for a cytoplasmic protein. This study includes the design of primer pairs, in silico and in situ selectivity, sensitivity, repeatability and reproducibility evaluations of the seven SYBR®Green qPCR assays. Each detection level displayed a selectivity of 100 %. This combinatory SYBR®Green qPCR system was also compared with three commercially available Salmonella qPCR detection kits. This comparison highlighted the importance of using a multi-gene detection system to be able to detect every target strain, even those with deletion or mutation of important genes. © 2013 The Author(s)

    Stationary System for Monitoring Technical State of Power Transformer

    AbstractImplementation of stationary systems of continuous monitoring technical state of power transformers is considered to be an important and relevant task. The paper provides a summery on the systems implemented at transformers of OJSC Magnitogorsk Iron and Steel Works. It characterizes the transformer of the power generating set of the thermal power plant equipped with the system for monitoring technical state. It lists diagnostic methods and offers an integrated system structure. The monitoring system is based on TDM package developed by OAO Dimrus. As a device for oil condition control, the Hydran M2 analyzer identifying gases dissolved in oil was used. Functions of separate modules, their version and arrangements are under consideration. The system interface is characterized, too. The main screen of controlled parameter of the transformer is provided in Inva-format. There is a challenge to develop methods for fault localization and identification based on the total of diagnostic features. The paper highlights a practical need for investigation of discharge activity with portable devices for control of partial discharges
