    Beton is tijdens het verharden onderhevig aan thermische en autogene vervormingen. Wanneer deze worden verhinderd, resulteert dit in spanningen en het risico op scheurvorming, mede afhankelijk van relaxatie. Aan de TU Delft is onderzoek gedaan naar het effect van spanningsrelaxatie op scheurvorming in jong beton. In tegenstelling tot de verwachtingen, blijkt dat spanningsrelaxatie in de eerste twee dagen juist resulteert in een verhoogd risico op scheurvorming bij bepaalde constructies met verhinderde vervorming. Ook blijkt dat autogene zwelling daarbij een significante rol speelt, die tot op heden in de norm buiten beschouwing wordt gelaten.Materials and EnvironmentConcrete Structure

    Characterization of an endochitinase able to rescue the carrot somatic embryo variant ts11

    Cultured carrot cells secrete proteins, many of which are glycosylated, into the culture medium. A correlation has been found between somatic embryogenesis and the presence or absence of some of these secreted proteins, and evidence has been obtained that one or more secreted glycoproteins are actually essential for somatic embryo formation. The starting point of the experiments described in this thesis was the temperature-sensitive carrot cell line ts11, originally identified on the basis of the temperature- sensitive arrest in the transition of globular to heart stage somatic embryos. The arrest in ts11 embryo development at the nonpermissive temperature could be lifted by addition of medium proteins, secreted by wildtype cells, to the culture medium. The major goal of the study presented in this thesis was to identify the secreted proteins, that were able to rescue the arrested ts11 embryos.In chapter 1 a brief introduction in zygotic and somatic embryogenesis is presented, followed by an overview of what is currently known about the first essential steps of the development of the zygotic embryo and of the formation of embryogenic cells and somatic embryos in vitro. Based on these studies, it is discussed whether analogous cellular mechanisms control early zygotic embryogenesis and the formation of embryogenic cells in tissue culture.In chapter 2 the experiments are described that demonstrate that is 11 embryos can be rescued by a single secreted protein of 32 kD. The amino acid sequences of two tryptic peptides of this protein shared homology with several plant endochitinases. Biochemical analysis showed that the 32-kD protein is an acidic endochitinase.In chapter 3 the results of a search for putative products of endochitinase activity effective in ts l 1 rescue, are presented. A molecule produced by Rhizobium, the N-acetylglucosamine-containing lipo-oligosaccharide, NodR1v-V(Ac, C18:4), appeared to be effective in stimulating the formation of ts11 embryos with a similar efficiency as the 32-kD endochitinase.In chapter 4 evidence is presented that a decreased amount of an otherwise fully functional endochitinase is closely correlated with the window of sensitivity of ts1 1 cells to addition of the 32-kD endochitinase. Morphological observations suggest that the original ts l 1 mutation is quite pleiotropic and does not only affect embryogenesis in this line.In chapter 5 experiments are described to identify a 32-kD endochitinase cDNA. The deduced amino acid sequence of the isolated cDNAs was found to be nearly identical to the amino acid sequences of the 32-kD endochitinase-derived peptides. The EP3 cDNA sequences suggested that the 32-kD endochitinse is a class IV chitinase.Finally, in chapter 6 the significance of chitinases and lipo-oligosaccharides for plant development in general is discussed

In the present paper, certain resultant data of the laboratory experiments with the Kozaka’s broad-bean method on evaluation of the fungicidal action of certain commercial fungicides against Pellicularia sasakii are reported.     Implications of Social Practice Theory for Sustainable Design

    Sustainable design faces challenges at a scale and level of complexity that are ill at ease with design’s mainstream focus on products and users. Recently, social practice theory has been suggested as a promising theoretical framework to inform new ways of designing. In social practice theory, practices – socially shared entities such as cleaning, cooking and playing – are taken as the fundamental unit of analysis. So far, however, design research in this area has been scattered and varying strongly in its interpretations of the implications of a ‘practice-orientation’.     Sheath blight disease incited by Pellicularia sasakii on rice plants is of the second importance to blast in distribution, as well as damage for the rice culture in Taiwan (Hashioka 1951). Under the present situation in Taiwan, the japnica type of rice is more susceptible. Yield loss from this disease is around 14-17% (Lo 1961). According to Lo (Personal communication), the loss due to the blast disease might have been exceeded by the scierotial sheath blight which has become increasingly prevalent in recent years (Chiu 1962). Sawada (1912) stated that the fungus might be able to infect a diverse number of plants, 47 species belonging to 4 families in Taiwan; 60 species belonging to 16 families in Japan. Matsumoto (1934) reported that there were 16 strains of the fungus collected in Taiwan from various hosts, including rice, sugarcane, groundnut, french bean, cowpea and camphor. Nakada and Kawamura (1939) pointed out that there were 16, 9 and 3 biotypes observed according to the cultural characteristics of the fungus on Czapek’s, Potato sucrose and dried plum agar, respectively. They also stated that the suscepts attacked by this parasite were about 188 species of 32 families in Japan. In consideration of the information stated above, one can presume the exi-stence of many physiologic races of the fungus in nature. For this reason an attempt on studies of the feasible existence of different races in Taiwan have been made in Department of Plant Pathology at Taiwan Agricultural Research Institute since 1957. In the present paper, experimental evidence confirmed the existence of physio-logic races of P. sasakii in Taiwan. 1. 自宜蘭、臺北、東勢、嘉義、屏東、臺東等地區,採集稻紋枯病害標本。經人工純系分離後,在馬鈴薯培養基上鑒別其培養型,並於溫窒內舉行人工接種。所選用十六鑒別稻品種係本研究室供鑒定稻熱病菌生理小種所用者。 2. 305 個純系分離之稻紋枯病病原菌,在馬鈴薯培養基上,以其菌絲之生長,培養基之色澤,菌核之形成日期、數目、色澤、形狀及在培養皿內之位置等特性,可分為七種培養型。 3. 根據以上七種培養型中,各選出一菌號,接種於選定之鑒別稻品種上,得到六種病原性不同之生理小種,即可分類為 1 , 2 , 3 , 4 , 5 及 6 等小種。 4. 在十六鑒別稻品種上,在來稻之抗病性高於蓬萊稻。在來稻中,陸稻比水稻更顯著

    The ideological sources of North Korean regime resiliency during Kim Jong-Un's leadership

    In this thesis, the author observes and analyzes how ideological discourse contributes to the\ud resiliency of the North Korean regime during the leadership of Kim Jong-un. He utilizes an\ud integrated quantitative and qualitative method to perform a content analysis of editorials\ud published in Rodong Sinmun, the official news publication of the ruling Korean Workers???\ud Party, between 2011 and 2013. In particular, he observes predication, presupposition, and\ud subject-object placement in sentence structure. The author finds that the party uses\ud numerous forms of discourse ??? Confucian, corporatist, strong-nation, and resistance ??? to\ud frame relationships shared by entities internal to North Korea. The party constructs political\ud reality for subjects by binding all together ??? the Kim family leaders, the party, the army, the\ud people, and other internal entities. In so doing, the party also prevents realization of\ud alternative political reality. Surprisingly, resistance discourse is generally limited in usage and\ud development through the two years of observation; however, it is more effectively used and\ud developed during a certain period in 2013, the year in which Kim Jong-un becomes a greater\ud discursive presence than his father and former North Korean leader, Kim Jong-il.     Development of scaling methods for broadband turbulent noise in internal flow devices by means of experiments and CFD

    Broadband turbulent noise can lead to fluid-acoustic and fluid-structure interaction and ultimately to mechanical failure of pipe systems. In HVAC (Heating Ventilating and Air Conditioning) systems broadband turbulent noise can radiate outwards where it is a nuisance or even harmful to people. A fast and simple approach for investigating broadband noise is the scaling law approach originally derived by (Nelson & Morfey, 1981) based on pressure drop. Recently this approach is improved by (de Jong & Golliard, 2013) using steady Reynolds Averaged Navier Stokes (RANS) simulations but was only validated using benchmark geometries. In this work experiments are performed on several DIN standard bends as well as on an orifice. Their sound power spectra are successfully scaled.
