Insights into the role of metal ions
No full textDie vorliegende Arbeit untersucht die Frage: Welche Wirkung haben Metall-Ionen auf die Struktur, Dynamik und Wechselwirkungen des Proteins ShhN, der N-terminalen Signal- Domäne von Sonic Hedgehog? Zur Beantwortung wurden molekulare Strukturen von ShhN mit verschiedenen Computer-gestützten Methoden analysiert. Die Analysen sagen einige überraschende Eigenschaften von ShhN vorher.
Zuerst legen die Analysen nahe, dass ShhN eine Zink-Protease ist, deren katalytisches Zentrum durch die Bindung von Calcium-Ionen reguliert wird. Speziell ändert die Bindung des zweiten Calcium-Ions die Konformation des Zink-Zentrums und beeinflusst so vermut- lich die Bindung des Substrats. Diese Konformationsänderung kaskadiert vom Calcium- Ion zum katalytisch Zink-Zentrum über mehrere Residuen und destabilisiert die Substrat- Bindestelle und das Zink-gebundene katalytische Wasser-Molekül. Änderungen des elek- trostatischen Potenzials um das Zink-Zentrum in Abhängigkeit von der Calcium-Bindung weisen hin auf eher unpolare Substrate, z.B. den C-Terminus von ShhN. ShhN könnte also ShhN-Moleküle abbauen, und auf diese Weise seinen Konzentrationsgradienten än- dern. Die Implikationen des beschriebenen neuartigen Mechanismus der Schaltung von ShhN über Calcium-Ionen für die biologische Funktion des Proteins sind noch weitgehend unverstanden, und erfordern weitere Forschung.
Die Flexibilität von ShhN-Monomeren und -Dimeren hängt stark ab von der Zahl der gebun- denen Calcium-Ionen. Im Besonderen sind die Calcium-bindenden Schleifen von ShhN davon betroffen. ShhN-Dimere werden stabilisiert durch das Cardin-Weintraub-Motiv am N-Terminus und durch Wechselwirkungen mit hydrophoben Residuen an der Schnittstelle zwischen den Monomeren. Die schwächere Konservierung von I48 in Wirbeltieren kön- nte ein Hinweis sein auf eine wichtige Rolle dieses Residuums in der Oligomerisierung von ShhN. Die Nähe von Calcium-Ionen zur Dimerisierungs-Region sorgt für ein positives elektrostatisches Potenzial und könnte so die Bindung von Proteoglykanen fördern. Ins- gesamt wird ein Modell der Multimerisierung von ShhN vorgeschlagen, das über mehrere Kontrollmechanismen verfügt, die experimentell getestet werden sollten.The present work studies the effect of metal ions on the structure, dynamics and inter- actions of the protein ShhN, the N-terminal signaling domain of Sonic Hedgehog. To accomplish this task, molecular structures of ShhN proteins were analyzed with a set of computational methods, revealing new features of ShhN proteins.
The results suggest that, ShhN is an enzyme with a zinc catalytic center that is regulated by the binding of the calcium ions. Explicitly, the binding of the second calcium ion involves a conformational change that is accompanied by a significant perturbation of the putative catalytic center, possibly affecting substrate stabilization. The dragging of E127 towards the calcium center implies the pulling of H135 with it and the disruption of the hydrogen bond between G128 and H141. Besides, the distance between residues E177 and H135 increases and therefore, the well-defined position of the catalytic water molecule is lost destabilizing the zinc environment. Electrostatic potential differences among calcium states suggest the possible binding of nonpolar substrates. One of the predictions is that ShhN autodegradates tuning its own concentration gradient. This possibility does not rule out, of course, the existence of other mechanisms that govern ShhN concentration gradient. The novel switching mechanism proposed could have many implications in the biological function of HhN proteins, but these are not well understood and require further research.
Both ShhN monomers and dimers show a flexibility pattern that strongly depends on the number of calcium ions. Specially, calcium binding loops reflect this behavior. The Cardin- Weintraub motif located within the N-terminal of ShhN proteins together with buried hydrophobic residues at the interface lead to an stable complex that enhances ShhN dimerization. The lower degree of conservation of I48 in vertebrate homologs might indicate that this is a hot spot residue with an important role in ShhN oligomerization. The presence of the calcium ions at the dimeric interface can promote ShhN-proteoglycan interactions providing a large positively charged region which is the ideal scenario for the binding of these molecules.
Taken all together, a multimerization model where different levels of interaction can control the way that ShhN multimers form is proposed. However, this model has yet to be tested

Para alcançar tal propósito, buscou-se compreender a relação dos moradores, turistas, empresários e representantes de instituições com o turismo na localidade. Outro objetivo diz respeito à identificação dos principais produtos turísticos da colônia. No que se refere à metodologia, este estudo caracteriza-se como qualitativo. Como ferramenta de coleta de dados foram utilizadas entrevistas semiestruturadas. Para a verificação dos dados, foi adotada a análise de conteúdo. Os resultados demonstram que a colônia possui ligação com a imigração alemã e que estes elementos também são reconhecidos na atividade turística. Os dados evidenciam que os principais segmentos trabalhados na localidade são o turismo de negócios, eventos, turismo cultural e de aventura. Os resultados sugerem que os moradores possuem uma percepção positiva da atividade turística. Como proposta para pesquisas futuras, sugere-se abordar a temática a partir do associativismo ou ampliar o número de entrevistados. Palavras-chave: Turismo Cultural. Imigração alemã. In order to achieve this purpose, it was sought to understand the relationship of residents, tourists, entrepreneurs and representatives of institutions with tourism in the area. Another objective is to identify the main tourist products of the colony. In terms of methodology, this study is characterized as qualitative. Semi structured interviews were used as a data collection tool. For data verification, content analysis was adopted. The results show that the colony has a link with German immigration and that these elements are also recognized in the tourist activity. The data show that the main segments worked in the location are business tourism, events, cultural and adventure tourism. The results suggest that the residents have a positive perception of tourism activity. As a proposal for future research, it is suggested to approach the subject from the associativism point of view or to expand the number of interviewees. Keywords: Cultural Tourism. Calidad de servicio y satisfacción de pacientes en un puesto de salud del distrito de Yanacancha, Pasco - 2023

calidad de servicio y satisfacción de los pacientes en un puesto de salud del distrito de
Yanacancha, Pasco - 2023?, y tuvo como objetivo general establecer la relación que existe entre
la calidad del servicio y la satisfacción de los pacientes en un puesto de salud del distrito de
Yanacancha, durante el año 2023. Metodología: Adopta un enfoque cuantitativo, se emplea el
método científico como enfoque general, junto con el método hipotético deductivo, en el diseño
de nivel es correlacional y básica; asimismo, se eligió un diseño transversal noexperimental;
además, la población estuvo conformado por 350 pacientes del puesto de salud y la muestra por
183 sujetos que fueron evaluados mediante las encuestas. Resultados:alrededor del 49.18%de
los pacientes perciben que, casi siempre el servicio proporcionado porel puesto de salud de
calidad, mientras que, un 38.25% de los pacientes casi siempre se sientensatisfechos con los
servicios de atención médico brindado. Conclusión: Existe relación significativa y positiva
(rs=0.602; p < 0.05) entre calidad de servicio y satisfacción del pacienteen el puesto de salud de
Yanacancha; lo cual significa que, mientras la percepción de la calidad del servicio
proporcionado por el puesto de salud mejora, también aumenta el nivel desatisfacción de
los pacientes. Recomendación: Implementar un Sistema de Gestión de Calidadbasado en la
norma ISO 9001, que permitirá mejorar la eficiencia, efectividad y seguridad en la prestación
de servicios de atención médica
Lesbianity, censorship and transgression: Cassandra Rios' autobiographical writing
No full textCassandra Rios (1932-2002), pseudônimo de Odete Rios, foi uma autora brasileira de literatura erótica que representava personagens homossexuais, e, sobretudo, relações amorosas entre mulheres. O principal período de sua produção marcou os anos da Ditadura Civil-Militar no Brasil (1964-1985), rendendo à autora e seus livros inúmeros processos de proibição, censura e perseguição, por serem estigmatizados e vistos como pornográficos. Além disso, Rios publicou duas obras autobiográficas, Mezzamaro, Flores e Cassis: o pecado de Cassandra (2000) e Censura: minha luta meu amor (1977), em que aborda momentos íntimos de sua trajetória e memória, tratando também da questão de sua perseguição durante, especialmente, as décadas de 1960 e 1970. Visando a possibilidade do estudo do campo das biografias e, mais
especificamente, das escritas autobiográficas como forma de compreender aspectos do contexto da autora em questão, bem como as dinâmicas de poder inseridas no processo de produção de uma escrita “de si” de autoria feminina, essa pesquisa tem como objetivo dar continuidade a um trabalho de iniciação científica e analisar as narrativas autobiográficas de Rios a partir de um viés historiográfico. that represented homossexual characters and above all, amorous relationships between women. The main period of her production marks the Brazilian Civil-Military Dictatorship's Years (1964-1985), yelding to the author her books countless processes, prohibitions, censorship and persecution for being stigmatized and seen as pornographic. In addition, Rios published two autobiographical writings, Mezzamaro, Flores e Cassis: o pecado de Cassandra (2000) e Censura: minha luta meu amor (1977) when she mentions intimate moments of her trajectory and memory, also dealing with the question about her persecution during especially the 1960s and 1970s. Aiming at the possibility of the biographies' studies and more specifically the autobiographical writings as a way of understanding aspects of the author's contexto in question, as well a the dynamics of power inserted in the process of producting a feminine "self-writing" this Project aims to continue scientific initiation work and analyze the autobiographical narratives of Rios from a historiographic perspective. In this sense, the construction of the author's memorialistic writing will be interprete and mapped, establishing approximations and distances with the female autobiographical theory relating such genre to the context of Cassandra Rios' production and her autobiographical performance.Não recebi financiament
Tito's Say: A Cantata by James DeMars with Texts by Alberto Rios
Get PDFabstract: Composer James DeMars has found a unique voice in the poetry of Alberto "Tito" Rios. The imagery combines memories and experiences of life on the Mexican/American border with scenes of love at various stages of life. The purpose of this study is to examine the structure, artistry and contemporary significance of Tito's Say, a cantata for soprano and baritone soloists, chorus, and orchestra by James DeMars with text by Alberto Rios, and to gain understanding of its inception, creation, composition, and subsequent performances. Description will be given in detail to increase familiarity with the work among choral musicians. Within this document, the reader will learn from personal interviews about James DeMars and Alberto Rios, focusing on their influences and experiences, including the circumstances leading up to the creation of Tito's Say. Furthermore, the reader will discover why the work was composed and how the collaboration came to be, details about its premiere, subsequent performances, and varying versions of this composition. A descriptive exploration and analysis of the text, its musical treatment, as well as an analysis of each of the five movements of the work are included herein, focusing on harmony, form, and style. Last, the author seeks to address stylistic and formal traits that unify the work, and performance considerations and challenges for the choir and for the orchestra. Negativity and failure in Cassandra Rios: on rethinking queer affections
No full textPartindo da fortuna crítica sobre a escritora Cassandra Rios, analiso, com base na fenomenologia queer de Sara Ahmed (2006) e a crítica de Heather Love (2007) sobre as genealogias da história LGBTQIA+, possibilidades de reelaborar percepções passadas, bem como o passado literário e pessoal da escritora lésbica Cassandra Rios. O intuito é encarar as faltas e elementos problemáticos presentes nos romances da autora, muitas vezes deixados de lado pelos estudos cassandrianos, de modo a enfocar as faltas e a negatividade queer presente na escrita de Cassandra Rios. Com isso, espera-se abrir caminho para pensar novas possibilidades e pistas para enxergar um elemento chave da história lésbica e LGBTQIA+ brasileira.Departing from the critical fortune on the writer Cassandra Rios, I analyze, based on the queer phenomenology by Sara Ahmed (2006) and Heather Love’s criticism on the genealogies of LGBTQIA+ history (2007), possibilities of re-elaborating past perceptions, as well as the literary and personal past of lesbian writer Cassandra Rios. The aim is to face the lacks and problematic elements present in the author\u27s novels, often left aside by Cassandrian studies, in order to focus on the failures and queer negativity present in Cassandra Rios\u27 writing. No full textThis paper aims to investigate the origins and developments of Cassandra Rios\u27 publishing and writing career, a writer known for the high number of works censored during the Brazilian military dictatorship. To that end, a brief overview of the book censorship activity during the military period was presented, as well as a closer look at the writer\u27s biography and cultural context. The main sources used to assess the reception of her work were articles published by journalists and newscasters in the newspapers O Pasquim, Correio da Manhã, Correio Braziliense and Diário de Pernambuco, O Dia and Correio Paulistano, among others. Lastly, the article proposes a reading of her 1980 novel Eu sou uma lésbica (I am a lesbian). Structured around the themes of child sexuality, podolatry, fetishism, pedophilia, incest and, of course, homoaffectivity, this novel was the last of her works to gain market circulation and offers important keys to reading the author\u27s work and reception throughout her career.Este trabalho se dedica a investigar as origens e os rumos da trajetória editorial de Cassandra Rios, pseudônimo de Odette Pérez Rios, escritora conhecida pelo alto número de obras censuradas durante o período da ditadura militar brasileira. Para tal, fez-se necessário apresentar um breve panorama da atividade censória de livros no período militar, e lançar um olhar mais detido sobre a biografia e o contexto cultural da escritora. Para avaliar a recepção de suas obras, consultou-se sobretudo a crítica expressa por periodistas e noticiaristas dos jornais O Pasquim, Correio da Manhã, Correio Braziliense e Diário de Pernambuco, O Dia e Correio Paulistano. It builds upon the Working Group I contribution to the IPCC's Fourth Assessment Report (AR4), and incorporates subsequent new findings of research. As a component of the fifth assessment cycle, the IPCC Special Report on Managing the Risks of Extreme Events and Disasters to Advance Climate Change Adaptation (SREX) is an important basis for information on changing weather and climate extremes.
This Summary for Policymakers (SPM) follows the structure of the Working Group I report. The narrative is supported by a series of overarching highlighted conclusions which, taken together, provide a concise summary. Main sections are introduced with a brief paragraph in italics which outlines the methodological basis of the assessment.

La literatura erótica de Cassandra Rios: O bruxo espanhol (1959) y uma mulher diferente (1968)

Through the generality of the work of the brazilian writer Cassandra Rios, aspects of personal life and her literary career are pointed out in order to understand the contradictions of the context surrounding the author as the constructed narratives responsible for opening the debate on preconceptions in the face of sexuality.El presente trabajo propone el estudio de dos novelas de Cassandra Rios, O Bruxo Espanhol (1959) y Uma Mulher Diferente (1968), en los cuales se pretende analizar aspectos fundamentales de la censura sobre sexualidad, para fundamentar la prohibición de los libros de Cassandra Rios desde la época de los 50´s hasta los 70´s.
