Cryopreservation and oxidative stress in porcine oocytes

    No full text
Several vitrification protocols have been established for porcine oocytes so as to facilitate gene banking of female germplasm. Although live piglets have been successfully produced from pig oocytes vitrified at the germinal vesicle (GV) stage, the competence of vitrified oocytes to develop into the blastocyst stage is greatly compromised following cryopreservation. The focus of this review is to elucidate the impact of cryopreservation on the redox balance of pig oocytes, making special reference to the relevance of non-enzymatic and enzymatic antioxidant defences. Besides, the regulation of gene expression in response to oxidative stress is also considered. Finally, we discuss the effects of supplementing maturation and vitrification media with the exogenous nonenzymatic antioxidants that have hitherto yielded the most relevant results

Seminal extracellular vesicles subsets modulate gene expression in cumulus cells of porcine in vitro matured oocytes

Seminal plasma (SP), a fluid composed mainly by secretions from accessory sex glands, contains a heterogenous population of extracellular vesicles (EVs), involved in several reproductive physiological processes. Seminal plasma has been found to modulate ovary function, in terms of hormone secretion and immune regulation. This study evaluated the potential effect of SP-EV-subsets on the modulation of cumulus-oocyte-complex (COCs) physiology during in vitro maturation (IVM). Two SP-EV-subsets, small-EVs (S-EVs) and large-EVs (L-EVs), were isolated from pig SP by size-exclusion-chromatography. Next, COCs were IVM in the absence (control) or presence of each SP-EV-subset to evaluate their uptake by COCs (PKH67-EVs labelling) and their effect on oocyte and cumulus cells (CCs) (gene expression, and progesterone and estradiol-17β levels). S-EVs and L-EVs were able to bind CCs but not oocytes. Supplementation with L-EVs induced changes (P ≤ 0.05) in the transcript levels of oocyte maturation- (HAS2) and steroidogenesis-related genes (CYP11A1 and HSD3B1) in CCs. No effect on nuclear oocyte maturation and progesterone and estradiol-17β levels was observed when COCs were IVM with any of the two SP-EV-subsets. In conclusion, while SP-EV-subsets can be integrated by CCs during IVM, they do not affect oocyte maturation and only L-EVs are able to modulate CCs function, mainly modifying the expression of steroidogenesis-related genes

UKMARC AMC: Draft Rev 4.0: UK MARC format for archives and manuscripts control (UK MARC AMC)

    No full text
This draft is the first attempt to establish a UK MARC specifically for Archives and Manuscripts Control since the British Library indicated that it would countenance such extensions to the national UK MARC format. In order to keep consistency with the general UK MARC format, standard UK MARC subject fields are not included in this document, since they should be taken from the latest version of the UK MARC manual. {A note of them should perhaps be included in UK MARC AMC.} {NB Text in braces is intended to be explanatory material for readers of this draft}. Certain other fields have not been included that might occasionally be used in the cataloguing of archival materials but would generally only be used for such materials in organizations which were combining archive databases with library databases. This MARC version is intended for use with descriptions of archive or anuscript material that follow, or fit, the traditional style of cataloguing: we assume that these will normally relate to paper or parchment originals. It is not intended for use with descriptions of other kinds of material. For these, fields may be drawn from the appropriate UK MARC document. MARC versions for use with archives in special formats should be developed, in order to complete the full range of facilities available to archivists and curators

In the context of the Educational Turn in the Arts, this project brings together a group of 4th grade pupils from the Drossiness Primary School to inhabit the Centre d’Arts Santa Mònica for a week and within the framework of a temporary exhibition. Using a narrative methodology of a reflexive and dialogical approach, I focus on four significant scenes that took place in the Sojornar 2024. In which, I analyse the different ways of doing of the artists, to locate their relationships with the rest of the agents involved. Finally, I conclude with a series of contributions of the research
