Isolation and HPLC quantitative analysis of new flavonoid tetraglycosides from Brazilian beverages (Maytenus ilicifolia and M. aquifolium).
Supercritical Fluid Extraction Of A High-ash Brazilian Mineral Coal
The effects of extraction temperature, solvent density and addition of cosolvents on the conversion, liquid yield and also on the characteristics of the extracts and residues from SCFE with toluene of a high-ash Brazilian mineral coal were experimentally investigated (Rocha [1]). The experiments were performed in a semi-batch laboratory-scale unit in the coal pyrolysis region - 593 to 673 K and with pressures up to 11.5 MPa. It was observed a reduction in the total sulfur content up to 44%, whereas providing residues with maximum losses of 16% of the original heat-content. The extracts were characterized by a reliable, non-destructive Preparative Liquid Chromatography method - PLC-8 (Karam et al. [2]), providing eight discrete fractions with well defined chemical functionality. With the addition of 5 mol% tetralin 21.7% conversion was obtained, that means an increase of 15% when compared to pure toluene, while with n-methyl-2-pyrrolidone (NMP) and ethanol the conversion decreased. © 1996 Elsevier B.V. Supercritical Technology In Brazil: System Investigated (1994-2003)
In this work, a review of the Brazilian scientific production in the past 10 years in the field of supercritical fluid is presented. The historical facts associated with the previous edition of the Brazilian Meeting on Supercritical Fluids are discussed. Because of the importance of the other South America countries contribution to the filed, a summary of the research published in the past 5 years by all South America countries is also presented. In the past 5 years (1999-2003) 82 papers from South American countries were published in journals indexed in the Web of Science data base. Of these, 26 papers were related to the use of supercritical fluids as an analytical tool. Supercritical extraction from a variety of vegetable raw material contributed with 38 papers and the petroleum industry added 2 papers to the field. Reactions contributed with 3 publications while thermodynamics and fundamental studies were responsible for 13 publications. The Brazilian contribution represented 53-84% of the publications in the above areas. © 2004 Elsevier B.V. All rights reserved.342 SPEC. Indigofera Suffruticosa Mill As New Source Of Healing Agent: Involvement Of Prostaglandin And Mucus And Heat Shock Proteins
Ethnopharmacological relevance: Indigofera suffruticosa is specie typical of the "Cerrado" or Brazilian savannah; it is a member of the Fabaceae family - in folkmedicine is used for gastric disorders, infection and inflammation. Aim of the study: Ethyl acetate fraction (AcF) and aqueous fraction (AqF) of the methanolic extract of I. suffruticosa leaves were evaluated against acute gastric ulcer. The AcF fraction was selected to assess its activity in ulcer healing and its gastroprotective effects via mucus and gastric secretion. Materials and methods: The gastroprotective action of AcF and AqF fractions were evaluated in a rodent experimental model. The action mechanisms, involvements of the antisecretory action, mucus and prostaglandin production, toxicological and healing activity of the AcF (100 mg/kg, p.o.) were evaluated. We also used histological analysis (HE and PAS) and immunohistochemical (PCNA and HSP-70) assays to evaluate the effects of I. suffruticosa. Results: AcF significantly inhibited the gastric mucosal damage caused by ethanol. This effect was statistically significant in 100 mg/kg group compared vehicle. AcF did not interfered with gastric secretion, significantly increased the PGE 2 and mucus production (validated in PAS technique). The gastroprotection was attenuated by pretreatment with N-ethylmaleimide, but not L-NAME. In acid-acetic-induced ulcer model AcF accelerated ulcer healing. Immunohistochemistry analysis showed induction of proliferating cell (PCNA) and heat shock protein (HSP 70). Programa de Pós-Graduação em Farmácia.Sorocea bonplandii (Baillon) Burger, Lanjow e Boer, devido à sua similaridade morfológica com Maytenus spp., é utilizada pela população com os mesmos fins medicinais desta. Ao contrário das espécies de Maytenus, que possuem inúmeros estudos químicos farmacológicos, as informações a respeito da espécie Sorocea bonplandii ainda são incipientes. Neste trabalho foram analisadas as frações éter de petróleo, clorofórmio e acetato de etila - obtidas de um extrato clorofórmico das folhas de S. bonplandii - quanto às suas atividades no bioensaio de letalidade para larvas de Artemia salina Leach. A fração com maior bioatividade neste ensaio foi a clorofórmica, da qual foram isolados e identificados, por ressonância magnética nuclear de 1H e 13C, um álcool primário saturado, o diterpeno fitol, um álcool vinílico de cadeia isoprenílica, uma mistura dos esteróides sitosterol e estigmasterol (sendo o primeiro a substância majoritária). Em outra etapa, após deslipidificação das folhas com éter de petróleo, foram preparados os extratos acetônico e metanólico. Deste foi isolado e identificado por espectrometria de massas e espectroscopia de infravermelho, o dissacarídeo sacarose. Os extratos metanólico e acetônico foram avaliados quanto a sua atividade anticoagulante in vitro. O extrato acetônico apresentou atividade anticoagulante nos testes de tempo de protrombina (TP) e tempo de tromboplastina parcial ativada (TTPa). O mesmo extrato não apresentou atividade sobre a agregação plaquetária induzida por ADP assim como, no teste de tempo de trombina (TT). O extrato metanólico não demonstrou atividade em nenhum dos ensaios anteriores. O efeito anticoagulante evidenciado neste trabalho, nunca descrito para esta espécie, abre novas perspectivas para futuras investigações químico-biológicas, visando o isolamento e identificação da(s) substância(s) responsável(eis) por esta bioatividade
Obtaining phenolic compounds from jatoba (hymenaea courbaril L.) bark by supercritical fluid extraction
The extraction of polyphenol compounds from jatoba (Hymenaea courbaril L. var stilbocarpa) bark using supercritical fluid extraction (SFE) with CO 2 and cosolvents has been investigated. Among the solvent systems studied, SFE using CO2 and water (9:1, v/v), at 323 K and 35 MPa, presented the best results, with extract yield of 24%, and with high antioxidant activity (IC50 of 0.2 mg/cm3). This solvent system was used to determine global yield isotherms, which were built at 323 and 333 K, and 15, 25, and 35 MPa, using a second lot of jatoba. The highest yield was 11.5% at 15 MPa and 323 K, with maximum total phenolic compounds (TPC) of 335.00 mg TAE/g extract (d.b.) and total tannins content of 1.8 g/100 g raw material. A kinetic experiment was performed using optimized conditions, yielding 18% extract, and the kinetic parameters were used to scale-up the process from laboratory to pilot scale. Galloylquinic acid derivatives from Byrsonima fagifolia leaf extract and potential antifungal activity.
Ethnopharmacological relevance: Byrsonima fagifolia Niedenzu (Malpighiaceae) and other Byrsonima species are popularly employed in Brazilian traditional medicine in the form of preparations as cicatrizing, antiinflammatory, and antimicrobial. Aim of the study: To characterize the phytochemical profile of the hydromethanolic extract obtained from B. fagifolia leaves (BF extract) and to evaluate the toxicity and the antifungal activity. based on extensive analyses of 1D and 2D NMR spectra (HMQC, HMBC and COSY) data. The antifungal effect was determined by the broth microdilution method and the toxicity was evaluated on erythrocytes from sheep?s blood and Galleria mellonella larvae. Results: Phytochemical investigation of the BF extract led to the isolation and characterization of pyrogallol, nbutyl gallate, 3,4-di-O-galloylquinic acid, 3,5-di-O-galloylquinic acid, 3,4,5-tri-O-galloylquinic acid, and 1,3,4,5-tetra-O-galloylquinic acid. The BF extract showed high content of galloylquinic acid derivatives reaching more than twenty-times the quercetin derivatives content, according to the quantification by HPLC. These galloylquinic acid derivatives, obtained during this study, and quercetin derivatives, previously isolated, were submitted to the antifungal assays. The BF extract inhibited yeast growth mainly against Cryptococcus spp., at concentrations of 1?16 ?g/mL, comparable to isolated compounds galloylquinic acid derivatives. However, the quercetin derivatives as well as quinic acid, gallic acid, and methyl gallate showed lower antifungal effect compared with galloylquinic derivatives. In addition, the BF extract had no hemolytic effect and no toxicity on G. mellonella
Supercritical Extraction Of Coumarin From Guaco (mikania Laevigata And Mikania Glomerata) For Pharmaceutical Applications
Guaco has several applications in the chemical and pharmaceutical industries. This plant is a source of coumarin, a diterpene compound with expectorants properties. In Brazil, guaco extracts are used as medicinal products authorized by the National Health Surveillance Agency (ANVISA). Studies proved that guaco extracts are also effective against cancer, ulcers and infectious diseases. Supercritical extractions have been widely used to obtain highly pure natural extracts that do not contain organic solvents and have not lost their thermosensitive compounds and therefore are suitable for pharmaceutical applications. In this study, coumarin was obtained from guaco leaves extracts by supercritical extraction under different experimental conditions according to a central composite rotational design (CCRD) with two independent variables, temperature (T) and pressure (P). Lower values of T were used to maintain the stability of thermosensitive compounds and because coumarin is highly soluble in supercritical CO2 even at low T. The responses surface analyzes (RSA) acquired by fitting of linear and quadratic models to experimental extraction yields and coumarin concentrations showed similar conditions for improving yields from both plant species. Although the extract yield values were similar for both plant species, coumarin concentrations were higher in Mikania laevigata than Mikania glomerata. The greater extract yield of 4.20% coumarin, related to the higher coumarin concentration (0.43 g × 100 g-1 in guaco leaves), was obtained under extraction conditions of P above 175 bar and T above 45 C. © 2013 Elsevier B.V.
