    The course of hepatitis E virus infection in pigs after contact-infection and intravenous inoculation

    Abstract Background Worldwide, hepatitis E virus (HEV) genotype 3 is observed in pigs and transmission to humans is implied. To be able to estimate public health risks from e.g. contact with pigs or consumption of pork products, the transmission routes and dynamics of infection should be identified. Hence, the course of HEV-infection in naturally infected pigs should be studied. Results To resemble natural transmission, 24 HEV-susceptible pigs were infected either by one-to-one exposure to intravenously inoculated pigs (C1-pigs; n = 10), by one-to-one exposure to contact-infected pigs (C2-pigs: n = 7; C3-pigs: n = 5) or due to an unknown non-intravenous infection route (one C2-pig and one C3-pig). The course of HEV-infection for contact-infected pigs was characterized by: faecal HEV RNA excretion that started at day 7 (95% confidence interval: 5–10) postexposure and lasted 23 (19–28) days; viremia that started after 13 (8–17) days of faecal HEV RNA excretion and lasted 11 (8–13) days; antibody development that was detected after 13 (10–16) days of faecal HEV RNA excretion. The time until onset of faecal HEV RNA excretion and onset of viremia was significantly shorter for iv-pigs compared to contact-infected pigs, whereas the duration of faecal HEV RNA excretion was significantly longer. At 28 days postinfection HEV RNA was detected less frequently in organs of contact-infected pigs compared to iv-pigs. For contact-infected pigs, HEV RNA was detected in 20 of 39 muscle samples that were proxies for pork at retail and in 4 of 7 urine samples. Conclusion The course of infection differed between infection routes, suggesting that contact-infection could be a better model for natural transmission than iv inoculation. Urine and meat were identified as possible HEV-sources for pig-to-pig and pig-to-human HEV transmission.</p

    New synthetic bile acid analogue agonists of FXR and TGR5 receptors: Analytical methodologies for the study of their physico-chemical properties, pharmacokinetic activity and metabolism.

    This thesis reports an integrated analytical approach for the study of physicochemical and biological properties of new synthetic bile acid (BA) analogues agonists of FXR and TGR5 receptors. Structure-activity data were compared with those previous obtained using the same experimental protocols on synthetic and natural occurring BA. The new synthetic BA analogues are classified in different groups according also to their potency as a FXR and TGR5 agonists: unconjugated and steroid modified BA and side chain modified BA including taurine or glycine conjugates and pseudo-conjugates (sulphonate and sulphate analogues). In order to investigate the relationship between structure and activity the synthetic analogues where admitted to a physicochemical characterization and to a preliminary screening for their pharmacokinetic and metabolism using a bile fistula rat model. Sensitive and accurate analytical methods have been developed for the quali-quantitative analysis of BA in biological fluids and sample used for physicochemical studies. Combined High Performance Liquid Chromatography Electrospray tandem mass spectrometry with efficient chromatographic separation of all studied BA and their metabolites have been optimized and validated. Analytical strategies for the identification of the BA and their minor metabolites have been developed. Taurine and glycine conjugates were identified in MS/MS by monitoring the specific ion transitions in multiple reaction monitoring (MRM) mode while all other metabolites (sulphate, glucuronic acid, dehydroxylated, decarboxylated or oxo) were monitored in a selected-ion reaction (SIR) mode with a negative ESI interface by the following ions. Accurate and precise data where achieved regarding the main physicochemical properties including solubility, detergency, lipophilicity and albumin binding . These studies have shown that minor structural modification greatly affect the pharmacokinetics and metabolism of the new analogues in respect to the natural BA and on turn their site of action, particularly where their receptor are located in the enterohepatic circulation

    The Late Ilerdian-Early Cuisian Roda Sequence spans approximately 3 Ma and formed in the South Pyrenean Foreland Basin (Spain). It is mapped in detail across the present-day Tremp-Graus, Ager and Ainsa sub-basins, and across the Boltaña Anticline into the Jaca sub-basin. The South Pyrenean Foreland Basin basin formed during the early stage of oblique collision of the Iberian Plate with the Eurasian Plate, and was bounded to the north by the Pyrenean chain and to the south by the Sierras Marginales. Within the basin, a time-sequenced series of curved thrusts developed (controlled by the location of inverted Mesozoic extensional faults) of which the Montsec thrust was decisive for the development of the Roda Sequence. This thrust sheet was displaced in a mostly southward direction and has an E-W orientation; it carried the Ilerdian-Cuisian sediments of the present-day Tremp-Graus and Ainsa sub-basins. It is well-expressed at the surface as well as partly buried by younger sediments. The Montsec thrust sheet is bounded east and west by the corresponding lateral ramps oriented SW-NW (Segre line), and SE-NW (La Fueba thrust system). Beyond the SE-NW oriented lateral ramp, the present-day Ainsa and Jaca sub-basins are located. The thrust had a significant influence on sediment dispersal patterns and the nature and location of depositional environments during the development of the Roda Sequence.Applied Geolog

    Estimation of hepatitis E virus transmission among pigs due to contact-exposure

    Locally acquired hepatitis E in humans from industrialized countries has been repeatedly suggested to originate from pigs. Pigs may serve as a reservoir of hepatitis E virus (HEV) for humans when a typical infected pig causes on average more than one newly infected pig, a property that is expressed by the basic reproduction ratio R(0). In this study, R(0) for HEV transmission among pigs was estimated from chains of one-to-one transmission experiments in two blocks of five chains each. Per chain, susceptible first-generation contact pigs were contact-exposed to intravenously inoculated pigs, subsequently susceptible second-generation contact pigs were contact-exposed to infected first-generation contact pigs, and lastly, susceptible third-generation contact pigs were contact-exposed to infected second-generation contact pigs. Thus, in the second and third link of the chain, HEV-transmission due to contact with a contact-infected pig was observed. Transmission of HEV was monitored by reverse transcriptase polymerase chain reaction (RT-PCR) on individual faecal samples taken every two/three days. For susceptible pigs, the average period between exposure to an infectious pig and HEV excretion was six days (standard deviation: 4). The length of HEV-excretion (i.e. infectious period) was estimated at 49 days (95% confidence interval (CI): 17-141) for block 1 and 13 days (95% CI: 11-17) for block 2. The R0 for contact-exposure was estimated to be 8.8 (95% CI: 4-19), showing the potential of HEV to cause epidemics in populations of pigs

    A B S T R A C TThe realization of cement procurement in Indonesia in the fourth quarter of 2020 decreased. This is in line with the declining domestic cement sales. However, three-wheel cement topped the first position in the top Brand index, this proves the strong position of the three-wheeled cement Brand in the community. This achievement certainly occurred because of the trust given by the community to Semen Tiga Roda in terms of quality dan service to the needs of the community. This study was conducted with the aim of identifying the effect of Product Quality (X) on Customer Loyalty (Y) for the three-wheel cement Brand. This research was conducted using descriptive quantitative research methods dan verification through validity test, reliability test, normality test,multicollinearity test, simple regression test dan partial test (T test). This research was conducted by collecting primary data in the form of questionnaires from 100 respondents dan secondary data in the form of three-wheel cement annual reports, previous research journals, books, dan data obtained from the official website. Based on the data from the results of the tests conducted by the author through the partial regression test (T test) it shows that Product Quality (X) has a positive dan significant effect on the Customer Loyalty variable (Y) with a significant value of 0.000 less than 0.05. Namun, semen tiga roda memuncaki posisi pertama pada top Brand index, hal ini membuktikan kuatnya posisi Brand Semen Tiga Roda di tengah masyarakat. Prestasi ini tentu terjadi karena adanya kepercayaan yang diberikan masyarakat terhadap Semen Tiga Roda dari segi kualitas maupun pelayanannya terhadap kebutuhan masyarakat. Penelitian ini dilakukan dengan tujuan untuk mengidentifikasi pengaruh Kualitas Produk (X) terhadap Loyalitas Pelanggan (Y) Brand semen tiga roda. Penelitian ini dilakukan dengan menggunakan metode penelitian kuantitatif deskriptif dan verifikatif melalui uji validitas, uji reliabilitas, uji normalitas, uji multikolinieritas, uji regresi sederhana dan uji parsial (uji T). Penelitian ini dilakukan dengan cara mengumpulkan data primer berupa hasil kuesioner dari 100 responden dan data sekunder berupa dari laporan tahunan semen tiga roda, jurnal penelitian terdahulu, buku, serta data yang didapatkan dari website resmi.     Slavonska knjizevnost na nehrvatskim jezicima još nije pronasla svoje mjesto u opusu nacionalne knjizevnosti iako je rijec o autorima i djelima koji svoje nadahnuce pronalaze u slavonskoj zemlji i čovjeku. Alexander Roda Roda (1872.–1945.) jedan je od rijetkih knjizevnika koji je cijeli knjizevni opus napisao na njemackome jeziku, a slaveci Slavoniju. Roman Vragolije tatina sina Marije, djelo je koje je samo na prvi pogled namijenjeno mladjem uzrastu. No bogatstvo obicaja, zanata, slavonske svakodnevice, saljive situacije koje proizlaze iz dodira slavonskoga ruralnoga covjeka s razvojem, mehanizacijom i inovacijom, ljepote slavonskoga krajolika – elementi su koji ce zainteresirati i starije citatelje i ovo djelo okarakterizirati kao solidan (dokumentaristicki) izvor za promisljanje o kulturi slavonskoga sela u posljednjim desetljecima 19. i u prvim desetljecima 20. stoljeca. Temeljni je cilj rada detektirati zanrovske osobitosti ovoga Roda Rodina djela. Pisac je, naime, sȃm svoje djelo zanrovski odredio kao roman: no otkrivajuci strategije i postupke koje je rabio u gradbi svijeta svoga djela (prije svega mislimo na kompoziciju teksta, polozaj pripovjedaca, njegov legitimitet, odnosno uvjerljivost, gradbu likova, njihovu zivotnost te funkciju opisa i pripovijedanja u djelu) pronalazimo diskrepanciju izmedju intencija što ih je autor najavio kako zanrovskim imenovanjem, tako i "nultom pripovijescu" u kojoj eksplicitno navodi razloge zbog kojih pise bas takvo djelo i ciljeve koje tim djelom zeli ostvariti.Slavonic literature in non-Croatian languages has still not found its place in the opus of national literature, although it is about authors and works who find their inspiration in Slavonian land and man. Alexander Roda Roda (1872–1945) is one of the few writers who wrote the entire literary work in German, but glorifying Slavonia. Novel Mischiefs of Dad's son Maria, a work that is only at first glance intended for younger audience. But the wealth of customs, crafts, Slavonian everyday life, humorous situations arising from the touch of Slavonian rural man with development, mechanization and innovation, the beauties of the Slavonian landscape – are elements that will interest older readers and mark this work as a solid (documentary) source for thinking about culture of Slavonian village at the end of the 19th and in the first decades of the 20th century. The basic aim of the work is to detect the genre features of this Roda Roda's work. The writer has himself, in fact, designated his work as a novel: but disclosing the strategies and procedures used in the construction of his work (above all, we think of the composition of the text, the narrator's position, his legitimacy, persuasiveness, character building, their vitality and the function of description and narration in the work) we find the discrepancy between the intents that the author announced both by the genre determination of the work and by the "null narrative" in which he explicitly states the reasons for which he writes such a work and the aims he wants it to accomplish

    MreC and MreD balance the interaction between the elongasome proteins PBP2 and RodA

This is an open access article distributed under the terms of the Creative Commons Attribution License, which permits unrestricted use, distribution, and reproduction in any medium, provided the original author and source are credited.Rod-shape of most bacteria is maintained by the elongasome, which mediates the synthesis and insertion of peptidoglycan into the cylindrical part of the cell wall. The elongasome contains several essential proteins, such as RodA, PBP2, and the MreBCD proteins, but how its activities are regulated remains poorly understood. Using E. coli as a model system, we investigated the interactions between core elongasome proteins in vivo. Our results show that PBP2 and RodA form a complex mediated by their transmembrane and periplasmic parts and independent of their catalytic activity. MreC and MreD also interact directly with PBP2. MreC elicits a change in the interaction between PBP2 and RodA, which is suppressed by MreD. The cytoplasmic domain of PBP2 is required for this suppression. We hypothesize that the in vivo measured PBP2-RodA interaction change induced by MreC corresponds to the conformational change in PBP2 as observed in the MreC-PBP2 crystal structure, which was suggested to be the “on state” of PBP2. Our results indicate that the balance between MreC and MreD determines the activity of PBP2, which could open new strategies for antibiotic drug development

The jamboree was held in the Asheville City Auditorium (now known as Thomas Wolfe Auditorium) from 1948 to 1973, and Hayes’ wife, Leona Trantham Hayes (1913-1989) continued to direct the program after his death in 1964.
