Opioide-hartzaileen deskribapena saguen gametoetan: morfinak obozitoen in vitro heltzean duen eragina
Gaur egun, ugarkotasun ezaren arazoa dela eta, geroz eta gehiago dira haurdunaldia lortzearren lagunduriko ugalketa tekniketara (ART, assisted reproductive technology) jo behar duten pertsonak; baina, oraindik, teknika horien arrakasta ez da oso handia eta batez beste, %50eko arrakasta duela esan ohi da(SEF erregistroa 2013). ART guztien artean, obozitoen in vitro heltzea (IVM, in vitro maturation) oso teknika berria da baina duen arrakasta txikiagatik, gutxien erabiliena da. Hala ere, in vitro ernalketaren (IVF, in vitro fertilization) hasieran eragingo litekeenez, ARTen arrakasta asko igoko litzateke

Identification of taxonomic changes in the fecal bacteriome associated with colorectal polyps and cancer: potential biomarkers for early diagnosis
Colorectal cancer (CRC) commonly arises in individuals with premalignant colon lesions known as polyps, with both conditions being influenced by gut microbiota. Host-related factors and inherent characteristics of polyps and tumors may contribute to microbiome variability, potentially acting as confounding factors in the discovery of taxonomic biomarkers for both conditions. In this study we employed shotgun metagenomics to analyze the taxonomic diversity of bacteria present in fecal samples of 90 clinical subjects (comprising 30 CRC patients, 30 with polyps and 30 controls). Our findings revealed a decrease in taxonomic richness among individuals with polyps and CRC, with significant dissimilarities observed among the study groups. We identified significant alterations in the abundance of specific taxa associated with polyps (Streptococcaceae,
Lachnoclostridium
, and
Ralstonia
) and CRC (Lactobacillales, Clostridiaceae,
Desulfovibrio
, SFB,
Ruminococcus
, and
Faecalibacterium
). Clostridiaceae exhibited significantly lower abundance in the early stages of CRC. Additionally, our study revealed a positive co-occurrence among underrepresented genera in CRC, while demonstrating a negative co-occurrence between
Faecalibacterium
and
Desulfovibrio
, suggesting potential antagonistic relationships. Moreover, we observed variations in taxonomic richness and/or abundance within the polyp and CRC bacteriome linked to polyp size, tumor stage, dyslipidemia, diabetes with metformin use, sex, age, and family history of CRC. These findings provide potential new biomarkers to enhance early CRC diagnosis while also demonstrating how intrinsic host factors contribute to establishing a heterogeneous microbiome in patients with CRC and polyps
La voz en los ensayos audiovisuales de Carolina Astudillo. Análisis de El Gran Vuelo (2014) y Ainhoa, yo no soy esa (2018)a partir del esquema comunicativo de Julia Kristeva
This article studies the voice in Carolina Astudillo’s cinema by analysing her two feature films El gran vuelo (2014) and Ainhoa, yo no soy esa (2018). Her works contain charactertistics of an essay film, such as using different techniques and materials, an asystematic discourse, and self-reflective speech. This form of filmmaking is particularly suitable for exploring themes that concern the author. We will apply the communicative scheme developed by Julia Kristeva in Semiotics (1969) to identify the different voices in these two films. Thus, we will demonstrate how the director’s voice is filtered through her characters, establishing a dialogue with them while at the same time turning it into a mirrorlike technique for self-exploration.
Liver GPBAR1 Associates With Immune Dysfunction in Primary Sclerosing Cholangitis and Its Activation Attenuates Cholestasis in Abcb4-/- Mice
Background and Aims: Primary sclerosing cholangitis (PSC) is a chronic cholestatic liver disease characterised by progressive biliary inflammation and fibrosis, leading to liver cirrhosis and cholangiocarcinoma. GPBAR1 (TGR5) is a G protein-coupled receptor for secondary bile acids. In this study, we have examined the therapeutic potential of BAR501, a selective GPBAR1 agonist in a PSC model. Methods: Single-cell analysis of healthy human liver samples and gene expression analysis of PSC liver samples were conducted. In vitro studies on a human cholangiocyte cell line (NHC), U937 and human hepatic stellate cells (hSteCs) were performed. Additionally, Abcb4−/− mice were treated with BAR501 for 12–24 weeks. Results: Single-cell analysis demonstrated that GPBAR1 is expressed by macrophages, NK cells, sinusoidal cells and to a lesser extent by cholangiocytes. Total liver expression of GPBAR1 increases in PSC patients compared to that in healthy controls and positively correlates with markers for monocytes and NK cells and cytokeratin 19. In vitro treatment of NHCs with BAR501 reversed the acquisition of a pro-inflammatory phenotype and the downregulation of GPBAR1 expression promoted by LPS in an NF-κB-dependent manner. Treating Abcb4−/− mice reduced bile duct inflammation and liver fibrosis and prevented the downregulation of GPBAR1 expression. Treating mice with BAR501 also modulated the bile acid pool composition and reduced the dysbiosis-associated gut permeability, and intestinal and systemic inflammation. Ex vivo experiments using conditioned media from BAR501-treated cholangiocytes mitigated the activation of macrophages. Conclusions: Our study provides evidence for the therapeutic potential of selective GPBAR1 agonists in intestinal inflammation–associated cholestasis, warranting the evaluation of BAR501 in PSC patients

Defence signalling marker gene responses to hormonal elicitation differ between roots and shoots
15 p., 5 tablas, 3 figurasPhytohormones such as jasmonic acid (JA), salicylic acid (SA), ethylene (ET) and abscisic acid (ABA) play a key role in regulation of plant immune responses to different attackers. Extensive research over recent years has led to the identification of molecular markers for specific hormonal-regulated defence pathways. However, most of our current knowledge on the regulation of plant immunity derives from studies focused on above-ground organs, mainly on the model plant Arabidopsis thaliana. Therefore, it is unclear whether the paradigms based on experiments on above-ground organs are entirely transferable to roots. Here, we used the non-model plant Brassica rapa to study the regulation dynamics of hormonal-related marker genes in both roots and shoots. These markers were identified in Arabidopsis shoots after elicitation of the JA-, SA-, ET- or ABA-signalling pathways, and are commonly used to study induced responses. We assessed whether the regulation of those genes by hormonal elicitation differs between roots and shoots. To discern whether the differences in marker gene expression between roots and shoots are related to differences in hormone production or to differential responsiveness, we also measured actual hormone content in the treated tissue after elicitation. Our results show that some of the widely used markers did not show specific responsiveness to single hormone applications in B. rapa. We further found that hormonal elicitation led to different response patterns of the molecular markers in shoots and roots. Our results suggest that the regulation of some hormonal-related marker genes in B. rapa is organ specific and differs from the Arabidopsis-derived paradigms. This research was supported by the German Centre

[EN] Mesopelagic fish populations from the orders Myctophiformes and Stomiiformes were investigated to determine the potential effects of physical and biological factors on their trophic ecology, and characterize the ecological role that have in an oligotrophic system such as the western Mediterranean. The relevance of the mesopelagic fish community is connected to the great biomass that they represent worldwide, constituting common dietary resources for large pelagic fishes of commercial interest, marine birds and marine mammals. Myctophids are also key components of the trophic webs since they perform extensive diel vertical migrations throughout the water column, acting as vectors of matter and energy fluxes. This study was developed in a region off the Balearic Islands during two seasonal periods, late autumn (water mixing period) and summer (stratification of the water column). The trophic behaviour and vertical distribution of mesopelagic fishes were analysed, and ontogenetic dietary shifts from young individuals to adulthood were described. These aspects have been hardly reported in juveniles and adults of Mediterranean mesopelagic fishes. The present study comprises the most abundant and frequent species of myctophids (Ceratoscopelus maderensis, Notoscopelus elongatus, Benthosema glaciale, Hygophum hygomii, H. benoiti, Lampanyctus crocodilus, L. pusillus, Lobianchia dofleini, and Myctophum punctatum) and stomiiforms (mainly, the gonostomatid Cyclothone braueri and the sternoptychid Argyropelecus hemigymnus) of the Mediterranean Sea. The first objective of this thesis was to confirm whether the classical larval identification based on morphological and pigmentation characters, for both myctophids and stomiiforms, was supported by using barcoding genetic techniques.
