    Automating data management in science and engineering

    In this thesis we investigate a profile of data management problems, from the initial stages of data capture and storage, through management of evolving data structures, to retrieval and analysis of distributed data. We concentrate on automating the data management process for scientific and engineering users, who are skilled in their field of work, but less familiar with database technologies. We first look at data capture, and automatic database generation and data storage, using metadata about the process of engineering design as an example. We use an abstraction layer that is independent of platform, database and data domain. A persistent archive should be flexible enough to cope with data structures that change over time. We show that our architecture naturally facilitates database schema evolution, providing such flexibility automatically. Finally, we demonstrate that scientific simulation results can be retrieved through an automatically generated, customisable web based interface and analysed with a secure post-processing facility for archived and user uploaded code.</p

    Characterization of a New Potyvirus Isolated from Jasmin (Jasminum sambac (L.)Ait) in Taiwan

    A previously undescribed virus disease that caused severe yellow mosaic on leaves of jasmin plants was found in 1995 at Kaohsiung and Pingtung, Taiwan. The first discernible symptom observed is irregular yellow spot. Later, the spot advanced and making the whole leaf yellowing. Three major symptom types, i.e. symptomless virus infection, irregular yellow spot and severe yellow mosaic were evidenced in the field. The virus disease is a complex disease, more than three filamentous viruses were observed in crude saps of field jasmin. A flexuous filamentous virus of 720-750 nm in particle size, was successfully isolated by mechanical inoculation from diseased jasmin showing yellow mosaic symptom. It is the first report of jasmin virus disease in Taiwan. The thermal inactivation point of the virus was 50-55 °C, dilution end point was 10-3 to 10-4 and the longevity in vitro was 2-3 days at 24°C and more than 10 months at -70 °C. Pinwheel-shaped and laminated inclusion bodies typical of potyvirus infection were found in local lesion of Chenopodium amaranticolor. Cellular organelles alterations of nuclei, chloroplast, mitochondria in virusinfected leaf of jasmin showing yellow mosaic symptom were also observed by electron microscopy. It had a very narrow host range restricted to three species in chenopodiaceous plants among 56 tested plant species belonging to 12 families. Attempts to transmit the virus by aphids using C. quinoa and C. amaranticolor as source plants failed. Protein with a molecular weight of 32 kDa was detected where purified virus was denatured with sodium dodecyl sulfate (SDS) and analyzed by electrophoresis in polyacrylamide gels (SDS-PAGE) and by western blotting. An antiserum with a titre of 512 was obtained by immunizing a white rabbit with the purified virions. In Western blotting, indirect ELISA, SDS-agar gel double diffusion test, the antiserum against the jasmin virus isolate reacted strongly with its homologous antigens, but not to antigens of fourteen other potyviruses. In order to further characterize the virus at the molecular level, CP genes of jasmin virus isolate was cloned, sequenced and analyzed. A sequence of 1289 nucleotides (nts) was found and the percent of nucleotide identities of the CP gene and the NCR were less than 69% and 54%, comparing to twenty potyviruses, respectively. Similarly, percent identities of CP amino acid sequence to those of twenty other known potyviruses were all below 69%. The virus was identified as a new potyvirus species on the basis of its particle morphology and size, physical properties, cytopatholoy, serological characteristics and percent of nucleotide and amino acid sequences identity, and a name, Jasmin virus T, JaVT, was previsionally proposed. It is the first report about successful virus isolation from jasmin in the world

    Jasmin Güven is a PhD student in the School of Chemistry at the University of Edinburgh. She took part in the Fortissat Science Alliance podcast recordings in August 2023.What is the Fortissat Science Alliance?The Fortissat Science Alliance was a Wellcome Trust & Children In Need "Curiosity" project. This scheme provided informal STEM learning opportunities for young people who attended the community centre Getting Better Together Shotts (GBT Shotts) between 2019 and 2023. Due to the COVID-19 pandemic, deliveries had to pivot online so the podcast was founded. These recordings were made via Zoom with warm-up STEM activities sent to every young person in advance, along with a profile page for each researcher, so that they were relaxed and able to ask excellent questions.Link to episode on Spotify.Depending on the broadcast date, podcast deliveries were co-sponsored by Glasgow Science Festival, EXPLORATHON 2021, or EXPLORATHON 2022/23.For the duration of the project, it was supported jointly by Children in Need and the Wellcome Trust. Student ovim projektom završava studiranje na Akademiji za umjetnost i kulturu u Osijeku i Odsjeku za kazališnu umjetnost. Ovaj pisani rad je njegov subjektivni osvrt na proces rada te finalni rezultat te objektivno iznošenje činjenica i podataka o autoru, kao i svima koji su sudjelovali u ovom projektu. Cilj ovog pisanog rada je da student postane magistar glume.Ivo Brešan's comedy The sighting of Jesus Christ in the barracks of M.P. 2507 is a graduation play by the student of acting Ivan Simon which was mentored by doc. art. Jasmin Novljaković. With this project the student completes his studies at the Academy of arts and culture in Osijek and the Department of Theater Arts. This masters thesis is his subjective review on the working proces as well as the final result and unbiased extraction of facts and informations about the author and everybody that participated in this project.
