    Sofi Oksasen romaanitaide

    No full text
Sofi Oksanen is the most visible and widely discussed Finnish author of the 21st century, yet her novels have gained less attention than her public performances. This study shifts the focus from the author’s persona to her literary art, proposing to read Oksanen’s fiction from the methodological viewpoint of the rhetorical theory of narrative. Accordingly, Oksanen’s six novels published to date – Stalinin lehmät, Baby Jane, Puhdistus, Kun kyyhkyset katosivat, Norma, and Koirapuisto – are considered as examples of authorial rhetoric and ethics, as narrative and textual constructions, and as affective readerly experiences. Instead of only following the rhetorical theory’s emphasis on character, plot, and progression, however, the study develops a new kind of narrative rhetoric, which also pays attention to language and politics. In the study, Sofi Oksanen emerges as a feminist narrative artist, who employs fiction as a serious rhetorical resource in order to say something worthwhile about the past history as well as the contemporary world

Smoothness correction for better SOFI imaging

    No full text
Sub-diffraction or super-resolution fluorescence imaging allows the visualization of the cellular morphology and interactions at the nanoscale. Statistical analysis methods such as super-resolution optical fluctuation imaging (SOFI) obtain an improved spatial resolution by analyzing fluorophore blinking but can be perturbed by the presence of non-stationary processes such as photodestruction or fluctuations in the illumination. In this work, we propose to use Whittaker smoothing to remove these smooth signal trends and retain only the information associated to independent blinking of the emitters, thus enhancing the SOFI signals. We find that our method works well to correct photodestruction, especially when it occurs quickly. The resulting images show a much higher contrast, strongly suppressed background and a more detailed visualization of cellular structures. Our method is parameter-free and computationally efficient, and can be readily applied on both two-dimensional and three-dimensional data.BN/Kristin Grussmayer La

Development of a Reversibly Switchable Fluorescent Protein for Super-Resolution Optical Fluctuation Imaging (SOFI)

    No full text
Reversibly switchable fluorescent proteins (RSFPs) can be effectively used for super-resolution optical fluctuation imaging (SOFI) based on the switching and fluctuation of single molecules. Several properties of RSFPs strongly influence the quality of SOFI images. These properties include (i) the averaged fluorescence intensity in the fluctuation state, (ii) the on/off contrast ratio, (iii) the photostability, and (iv) the oligomerization tendency. The first three properties determine the fluctuation range of the imaged pixels and the SOFI signal, which are of essential importance to the spatial resolution, and the last may lead to artificial aggregation of target proteins. The RSFPs that are currently used for SOFI are low in averaged fluorescence intensity in the fluctuation state, photostability, and on/off contrast ratio, thereby limiting the range of application of SOFI in biological super-resolution imaging. In this study, we developed a novel monomeric green RSFP termed Skylan-S, which features very high photostability, contrast ratio, and averaged fluorescence intensity in the fluctuation state. Taking advantage of the excellent optical properties of Skylan-S, a 4-fold improvement in the fluctuation range of the imaged pixels and higher SOFI resolution can be obtained compared with Dronpa. Furthermore, super-resolution imaging of the actin or tubulin structures and clathrin-coated pits (CCPs) in living U2OS cells labeled with Skylan-S was demonstrated using the SOFI technique. Overall, Skylan-S developed with outstanding photochemical properties is promising for long-time SOFI imaging with high spatial-temporal resolution

Everything is Worth Selling: Commercial Sex, Violence and Heterosexuality in Sofi Oksanen’s and Essi Henriksson’s Novels Maria Jotuni, a celebrated Finnish realist female author of the 19th century, depicted women who were forced to sell both their bodies and their souls in business-like, heterosexual marriage arrangements. The same can be said of the women depicted by two contemporary Finnish female authors, Sofi Oksanen (Baby Jane 2005) and Essi Henriksson (Ilmestys 2007, ‘Revelation’), although their female characters do not get married. Instead, they objectify their bodies and feelings in slightly different business arrangements, that is, in sex work. The novels do not represent only commercial sex and sex work, but they also depict heterosexual relationships. These relationships become, like in Jotuni’s novels, gendered and unequal power relations in which money plays a significant role. Economical power signifies for gendered and heterosexual power, and by oppressing and objectifying women’s bodies and feelings this economical, gendered and heterosexual power system is connected also to violence. This article discusses the representations of commercial sex, sex work, and gendered, sexual violence in the novels of Sofi Oksanen and Essi Henriksson, and their interrelations with heterosexuality and heteronormativity. In addition, this article examines the meanings of gender and sexuality in the late capitalistic consumption culture. The image that the novels create of gendered and heterosexual power relations encourages the reader to reconsider gendered power relations in general and the idea of heteronormative social order – which still, a hundred years after the publication of Jotuni’s works, supports the hierarchies and inequality between genders. This is how the novels of Oksanen and Henriksson participate in the continuum of Finnish female authors who criticize the cultural norms of gender and sexuality
