Abstract 1472: The role of the BAF complex in Wnt signaling-mediated transcriptional regulation in colorectal cancer
Abstract Recent genome- and exome-wide sequencing studies have revealed a close association between the epigenome and the pathogenesis of cancer. Not only were chromatin regulators found to be commonly mutated in a wide range of cancers, but a class of these regulators, the ATP-dependent chromatin remodelers, especially subunits of the mammalian BAF complex, were among the most frequently mutated group of genes across cancer types, mutated in over 20% of cancers. Among these subunits, loss of function mutations in ARID1A (AT-rich interactive domain-containing protein 1A) are most frequent across cancer types including 10% of colorectal cancer (CRC). Very interestingly, a recent publication described the pivotal role of the loss of ARID1A in driving CRC wherein its inactivation alone led to the formation of invasive adenocarcinomas in mice. However, surprisingly, in contrast to the expected tumor suppressive role of ARID1A in CRC, we observe that the knockout (KO) of ARID1A in CRC cell lines leads to impaired proliferation. Moreover, subcutaneous xenografts in SCID/SHO mice using ARID1A KO CRC cells did not form more aggressive tumors than their wild type counterparts. One of the most commonly occurring mutations in CRC is in the APC gene, which leads to hyperactive Wnt signaling. Thus CRC progression is typically highly dependent on this pathway. Notably, it is reported that loss of ARID1A in the context of APC mutations results in diminished tumor formation in mice and increased differentiation. This led us to hypothesize that ARID1A is required for Wnt signaling-mediated transcriptional (de)regulation in CRC. To uncover this further, we utilized several publically available ChIP-seq, RNA-seq and ATAC-seq datasets as well as our own ChIP-seq data for ARID1A in the HCT116 cell line. We observe a substantial co-localization of the BAF complex with TCF7L2, a downstream effector of the Wnt signaling pathway, suggesting cooperation between these factors. Further, these sites are also co-occupied by AP1 transcription factors. A crosstalk between the TCF7L2 and AP1 factors has been reported in intestinal tumors. From data available in the ARID1A KO system, we observe a downregulation of potential target genes which are co-occupied at neighboring sites by these factors. Therefore, we suggest that ARID1A plays an important role in the regulation of colorectal cancer relevant genes that are targets of TCF7L2/AP1, thus facilitating a pathway that is most commonly hijacked in colorectal cancer. In this presentation, Dr. Rapp provides an overview of the principles of universal design for learning (UDL) and the application of each principle to the college setting. Dr. Rapp outlines the responsibility of higher education institutions for providing supports in coursework and field-based experiences while maintaining high expectations for knowledge and skill acquisition. She shares several specific strategies for universally-designed learning environments that help prepare a more diverse group of people for success in their chosen fields.
Biography
Dr. Rapp is Associate Professor of Inclusive Education at St. John Fisher College, where she teaches courses on inclusive education pedagogy, assessment, and classroom management. Dr. Rapp holds a bachelor’s degree in elementary education and psychology from the State University of New York at Potsdam and master’s and doctoral degrees in special education from Michigan State University. Dr. Rapp’s current research interests include universal design for learning (UDL) throughout the school years and college, as well as strategies for executive functioning. She is the author of the textbook Universal Design for Learning: 100 Ways to Teach All Learners (2014) and coauthor of the textbooks Teaching Everyone: An Introduction to Inclusive Education (2012, with K. L. Arndt) and Picture Inclusion! Snapshots of Successful Diverse Classrooms (Feb. 2019, with K. L. Arndt and S. M. Hildenbrand). Dr. Rapp presents at local, state, national, and international conferences on UDL and inclusive education pedagogy

A 23-year old woman with narrow nose, maxillary hypoplasia, cleft lip and palate, hypodontia, dystrophic nails and hypohidrosis is described. Her scalp hair was coarse, dry and wiry. Microscopic examination of hair showed longitudinal twisting at irregular intervals, typical of pili torti. Her mother presented only dystrophic nails and hypohidrosis. Rapp-Hodgkin syndrome is an uncommon, autosomal dominant condition characterized by distinctive craniofacial anomalies and cleft lip and palate. In the patients observed, the phenotypical manifestations were different. In fact, the proband showed all the major features of the syndrome, while her mother only onychodysplasia and hypohidrosis. The differential diagnosis between Rapp- Hodgkin syndrome and other forms of ectodermal dysplasia are discussed.

An employment agreement is a relationship between someone who acts as a laborer or worker and someone who acts as an employer. Work plays a vital role in human life, so it qualifies as a means of generating income to meet needs. One of the inalienable human rights that needs to be upheld is the freedom to work and receive rights in the form of wages to meet daily needs. Based on the author's background, here are several problem formulations raised by the author, first, how can an employee be fired for committing serious violations in accordance with applicable rules and regulations. Second, what is the legal protection for employees whose employment relationship is terminated due to making serious mistakes at work. The research used by the author is empirical research, namely conducting research directly at the research location to obtain accurate data which is carried out by means of interviews and documentation. The nature of the research used by the author is sociological law, by consistently expressing facts in the field. The location of the author's research is PT RAPP, Pangkalan Kerinci District, Pelalawan Regency. The method of drawing conclusions used by the author uses a deductive method, which involves conclusions from general to specific matters. From the results of the research and discussion, the author found that employees can be fired if they make serious mistakes in the form of making double gate passes without PT RAPP permission and taking action to ask PT RAPP employees for additional money. Legal protection for employees whose employment relationship is terminated is by taking legal action by conducting bipartite negotiations and the final effort by workers is by registering an industrial relations dispute lawsuit in the industrial relations court
