DEVELOPMENT OF A REFERENCE METHOD FOR THE ABSOLUTE MEASURING OF FLUORESCENCE IN BIOLOGICAL ANALYSIS

    No full text
The research presented here was aimed to develop a method traceable to SI to compare the absolute fluorescence intensity measured by different instruments. This is due to the fact that the most common instruments use arbitrary measurement units. Each measure strictly depends on the technical intrinsic properties of the single instrument, hence the use of suitable reference materials is necessary to define its calibration scale. To overcome this problem, it is necessary to define a reliable and universal measurement unit allowing to measure the absolute fluorescence intensity, whichever system is used, independently from the reference materials which are used. In the present work, a protocol that permits the comparison of data taken with different fluorescence instruments has been developed The relationship between the arbitrary unit of these instruments and the molecule numbers of the standard reference material used has been studied. This allows the transformation from arbitrary unit to MESF, i.e. MOLECULAR EQUIVALENTS OF SOLUBLE FLUOROCHROME [1]. The standard reference fluorescence material used was Fluorescein. Standard reference materials of Fluorescein were used to ensure the traceability of measurements results. The work goals were to obtain: a method to prepare fluorescein solutions of calibration; characterization of fluorescence instruments ad their calibration; study of a measurement method of the calibration samples and calculation of their uncertainty budget ,expressed in number of molecules of fluorescein, (NF). The final work goal was to obtain an uncertainty budget for measurements, expressed in equivalent number of molecules of fluorescein (MESF) for each used instrument.. By applying this method It has been possible to compare different measurements, using different fluorescence instruments

Large transformations with moderate strains of tensile membrane structures

Beatini, Valentina/0000-0002-6916-9897; Royer Carfagni, Gianni/0000-0003-4879-9846Using a classical non-linear theory, we analytically investigate possible ways for transforming the shape of a curved elastic membrane while keeping it tensioned and moderately strained. This is a critical issue because, as a rule, membranes must be considerably stretched in order to avoid wrinkling and slackening. If the final configuration is fixed, the membrane can be cut and formed according to the final shape, but this cannot be done if more configurations, considerably distant from one another, have to be achieved. Nevertheless, we propose large transformation movements that can be obtained starting from flat membranes while maintaining their strain as limited. We discuss in detail the paradigmatic example of the hyperbolic-paraboloid-shaped membrane. These opportunities are suitable for applications of transformable architecture because they do not require excessive tensioning, compatible with the strength of materials used for this kind of structures.Italian "Dipartimento della Protezione Civile" under project ReLUIS-DPCThe author(s) disclosed receipt of the following financial support for the research, authorship, and/or publication of this article: G.R.C. acknowledges the partial support of the Italian "Dipartimento della Protezione Civile" under project ReLUIS-DPC 2014-2018

The role of educative thought in the life and work of Antonio Gramsci

    No full text
Many philosophers have propounded a vision of an improved society, what distinguishes Antonio Gramsci is his continuous effort to make it happen by understanding the process in order to put into practice. Gramsci's conviction about the importance of educative development came from both theory and experience. While there has been considerable examination of Gramsci's work in relation to the Prison Notebooks, this study will seek to address a lacuna in Gramsci scholarship. Using Gramsci's philological method, I analyse Gramsci's pre-prison activity; his pre-prison articles and letters, which, together with his letters from prison, formed part of his educative mission. This educative process was necessary, in order to construct a new party which would develop a collective will, collaboratively, with the masses. In this study therefore, I explore the contexts and formative experiences of the first part of his life together with the intellectual sources from which Gramsci developed his later theories, making central hitherto underemphasised connections between them which informed his writing and ideas. I intend to illustrate that Gramsci's underlying purpose in his writing, and political activity, was not only practical, on how to create a new socialist ruling class, but also educative in forming the mindset and values of his comrades. So that in addition to outlining his vision of a new order, he implicitly guided or explicitly explained the processes by which the necessary changes in social relations and moral climate could be made in order to achieve it. Each person had to engage with the values of the new order so that each could contribute to the construction of a new robust state. It was essential to build a hegemony at the most profound level, one which was dependent on collective understandings and a collective will

'Beni comuni'. Note minime di ordine metodologico

Commons (in Italian “beni comuni”) occupy the centre of legal debate from different perspectives. The author deals with the possible Roman roots of this topic with some criticism from the point of wiew of the method. The author stresses the opportunity to use a correct and historical approach, avoiding short circuits and superimpositions (“autoproiezioni” according to Riccardo Orestano) between the present and the past

Gianni Vattimo: a questão do pensamento e a racionalidade hermenêutica

This article presents the thesis the weak thinking and nihilistic vocation of Gianni Vattimo's hermeneutics, unlike the criticisms and misunderstandings underwent, does not necessarily imply the assumption, defense and proposal of a philosophical and hermeneutical irrationality. Based on a hermeneutics of the Italian philosopher texts, it is evident the criticisms of irrationalism are contested by the author, however, like all philosophy, it is necessary to deepen and reflect on the implications this philosophy has for modern philosophy and hodiernal hermeneutics. The concepts of thought and hermeneutic rationality are the axes upon which Gianni Vattimo's philosophy is analyzed and philosophically reflected in this articleO presente artigo apresenta a tese segundo a qual o pensamento fraco e a vocação niilista da hermenêutica de Gianni Vattimo, ao contrário das críticas e incompreensões sofridas, não implica necessariamente a assunção, defesa e proposta de uma irracionalidade filosófica e hermenêutica.

Children’s literature, in its multidisciplinary nature, offers a wealth of research options that span from psychology, to education, to translation. In this book, Alborghetti looks into the potential of children’s literature to promote empathy in young readers, and especially delves into the narrative dialogue among author, translator, and target reader. The author explores the literary and translation contexts in the United States and the United Kingdom, before presenting the case study of Gianni Rodari’s Italian works translated into English, and their relevance in the debate around the power of children’s literature in these Anglophone countries. This book is useful for researchers, educators and professional translators who wish to explore the world of children’s literature in translation

Un flâneur al pascolo. Gianni Celati e l’esperienza dell’ambiente

The essay aims to analyze the writings of Gianni Celati and his way of ‘re-narrating’ and ‘re-mapping’ the environment in contemporary times, focusing on two trilogies, namely Narratori delle pianure (1985), Quattro novelle sulle apparenze (1987) and Verso la foce (1989), as well as on the collections of short stories Un eroe moderno (2008), Il benessere arriva in casa Pucci (2008), and Selve d’amore (2013). The paper discusses how the themes addressed by the author, who assumes the role of a flâneur of postmod- ernism, are a way to express his continuous nostalgia for the naturalness, simplicity, and originality of living and narrating. The early dynastic texts from Lagash present numerous philological and historical problems. In this work, the author considers some of the philological and historical problems raised by the onomastic element LUM-ma, which is variously attested as a component of canal and place-names; as a theophoric element in personal names; as personal name in its own right; and, according to many scholars, as one of the names of the third king of the Lagash dynasty, E'annatum—no doubt the most prominent of the pre-Sargonic rulers of Lagash and one of the most important figures in the history of Babylonia in the third millenium B.C.E

    An introduction to Γ-convergence

The last twentyfive years have seen an increasing interest for variational convergences and for their applications to different fields, like homogenization theory, phase transitions, singular perturbations, boundary value problems in wildly perturbed domains, approximation of variatonal problems, and non- smooth analysis. Among variational convergences, De Giorgi's r-convergence plays a cen- tral role for its compactness properties and for the large number of results concerning r -limits of integral functionals. Moreover, almost all other varia- tional convergences can be easily expressed in the language of r -convergence. This text originates from the notes of the courses on r -convergence held by the author in Trieste at the International School for Advanced Studies (S. I. S. S. A. ) during the academic years 1983-84,1986-87, 1990-91, and in Rome at the Istituto Nazionale di Alta Matematica (I. N. D. A. M. ) during the spring of 1987. This text is far from being a treatise on r -convergence and its appli- cations
