An algorithm for compositional nonblocking verification of extended finite-state machines
No full textThis paper describes an approach for compositional nonblocking verification of discrete event systems modelled as extended finite-state machines (EFSM). Previous results about finite-state machines in lock-step synchronisation are generalised and applied to EFSMs communicating via shared variables. This gives rise to an EFSM-based conflict check algorithm that composes EFSMs gradually and partially unfolds variables as needed. At each step, components are simplified using conflict-equivalence preserving abstraction. The algorithm has been implemented in the discrete event systems tool Supremica. The paper presents experimental results for the verification of two scalable manufacturing system models, and shows that the EFSM-based algorithm verifies some large models faster than previously used methods

Factors that mediate and prevent degradation of the inactive and unstable GudB protein in Bacillus subtilis
No full textThe Gram-positive model bacterium Bacillus subtilis contains two glutamate dehydrogenase-encoding genes, rocG and gudB. While the rocG gene encodes the functional GDH, the gudB gene is cryptic (gudBCR) in the laboratory strain 168 due to a perfect 18 bp-long direct repeat that renders the GudB enzyme inactive and unstable. Although constitutively expressed the GudBCR protein can hardly be detected in B. subtilis as it is rapidly degraded within stationary growth phase. Its high instability qualifies GudBCR as a model substrate for studying protein turnover in B. subtilis. Recently, we have developed a visual screen to monitor the GudBCR stability in the cell using a GFP-GudBCR fusion. Using fluorescent microscopy we found that the GFP protein is simultaneously degraded together with GudBCR. This allows us to analyze the stability of GudBCR in living cells. By combining the visual screen with a transposon mutagenesis approach we looked for mutants that show an increased fluorescence signal compared to the wild type indicating a stabilized GFP-GudBCR fusion. We observed, that disruption of the arginine kinase encoding gene mcsB upon transposon insertion leads to increased amounts of the GFP-GudBCR fusion in this mutant. Deletion of the cognate arginine phosphatase YwlE in contrast results in reduced levels of the GFP-GudBCR fusion. Recently, it was shown that the kinase McsB is involved in phosphorylation of GudBCR on arginine residues. Here we show that selected arginine-lysine point mutations of GudBCR exhibit no influence on degradation. The activity of McsB and YwlE, however, are crucial for the activation and inhibition, respectively, of a proteolytic machinery that efficiently degrades the unstable GudBCR protein in B. subtilis

No full textMit der Absicht, eine Brücke zwischen deutschsprachiger Literatur und Humanistischer Psychologie zu konstruieren, geht die vorliegende Arbeit der Frage nach, ob es möglich ist, eine fiktive Person, die vor dem Hintergrund der Weltwirtschaftskrise von 1929 im Verlauf einer Romanhandlung arbeitslos geworden ist, durch Anwendung einer humanistischen Therapieform wieder in die Arbeitswelt zu integrieren. Die fiktive Person ist Jakob Fabian aus Erich Kästners neusachlichem Zeitroman Fabian. Die Geschichte eines Moralisten, erschienen 1931, und bei der humanistischen Therapieform handelt es sich um den „Klientenzentrierten Ansatz“ nach Carl Rogers. Die zentrale Forschungsfrage wird in zwei großen Schritten beantwortet: Im ersten Schritt werden themenrelevante Aspekte hinsichtlich Roman, Autor sowie Protagonist erläutert und analysiert, bis die Romanfigur Jakob Fabian zum Klienten Jakob Fabian wird und ein Anamnesebericht über diesen vorliegt. Basierend darauf, werden mit dem Ergebnis der beruflichen Rehabilitation im zweiten Schritt klientenzentriert-orientierte Therapiegespräche in einem realen Setting durchgeführt und transkribiert. Das Konzept dieser Arbeit kann als Modell auf weitere Arbeitslosenromane der Weimarer Republik angewendet werden. Zusätzlich besteht auch die Möglichkeit, das Setting an Beratungssituationen anzupassen.With the intention of constructing a bridge between German literature and “Humanistic Psychology”, the present thesis analyses the question, whether it is possible to reintegrate a fictitious man into the working environment by applying a humanistic type of therapy, who, against the background of the world economic crisis of 1929, in the course of a novel's plot has lost his job. Jakob Fabian is the fictional character in Erich Kästner’s new objectivity period novel Fabian. The story of a moralist, pub-lished in 1931, and the humanistic therapeutic method is the Rogerian Client-centered approach. The core issue is answered in two major steps: In the first step, issue-specific aspects regarding novel, author and protagonist are explained and analysed until the fictional character Jakob Fabian turns into the client Jakob Fabian and until an anamnesis report is available. Based on this, with the outcome of vocational rehabilitation in the second step, client-centred-oriented therapy sessions are conducted in a real setting and transcribed. The concept of this work can also be used as a model for other novels about unemployment during the Weimar Republic. In addition, it is also possible to adjust the setting of advisory situations
