    The focus of the thesis is on the evaluation of the effect of the implementation of a three-hour per week problem-based learning (PBL) module for 1SI year undergraduate students. The research questions are outlined below: • What approaches to learning are undergraduate students adopting at the initial stage of tertiary education? • Are student approaches to learning related to age/gender/ time in university/achievement in examinations? • Can a PBL module in chemistry be developed that can provide an effective teaching and learning environment, which develops students’ understanding in chemistry and engages the students with the context and processes o f chemistry? • Will the introduction of such a PBL module in chemistry have an effect on students’ approaches to learning? The main evaluation tool for determining student approaches to learning was the learning style inventory - Approaches and Study Skills Inventory for Students. Other evaluation tools employed were student surveys, interviews and assessment data. Key findings were that students on entry to tertiary level report a preference for deep and strategic approaches to learning over a surface approach. However, with time in university, the profile shifts with students indicating increasing use o f a surface approach. Mature students tended to prefer a deep approach in comparison to their younger counterparts and female students were more strategic than males in their approach. An introductory year-long chemistry PBL laboratory module was developed, implemented and evaluated. Interestingly, students who took part in the PBL module showed a lower preference for a surface approach having successfully completed the PBL module compared to those who followed the traditional approach despite showing similar trends at the start of the study. The effect on a deep approach to learning, after taking the PBL module, one sixth of the 1st year course, was not evident however. Conversely, the PBL students did significantly better in a non-formal exam designed to assess students learning in the first year chemistry laboratory. The students also successfully engaged with the chemistry content, context and processes and reported the benefit of the pre-lab, group work and practical aspects of the PBL approach

    Wetlands and future change—Implications and opportunities with the Ramsar Convention

    In the face of global-scale decline in the extent and state of the world’s wetlands, the Ramsar Convention has mobilised most of the world’s nations to support the conservation and wise use of wetlands. Whilst wetlands continue to be lost and their condition overall continues to decline, the Convention has played a significant part in assisting nations to mitigate pressures and preserve values and ecological assets. The pressures on wetlands will continue, and the Convention could continue its valuable contribution by increasing the awareness of the value of, and risks to wetlands, assisting Contracting Parties to plan for and accommodate change in wetland state, including that driven by climate change, recognising wetlands as socio-ecological systems, and refining the practice of wetland restoration. There remains much opportunity to extend the technical and management capacity that can be catalysed by the Convention to all nations to address the conservation and wise use of wetlands across the globe. © 2023 Elsevier Inc.     Measurement of the ratio of prompt χ c to J / ψ production in pp collisions at √s = 7 TeV

    The prompt production of charmonium χ c and J / ψ states is studied in proton-proton collisions at a centre-of-mass energy of √s = 7 TeV at the Large Hadron Collider. The χ c and J / ψ mesons are identified through their decays χ c → J / ψ γ and J / ψ → μ + μ - using 36 pb - 1 of data collected by the LHCb detector in 2010. The ratio of the prompt production cross-sections for χ c and J / ψ, σ (χ c → J / ψ γ) / σ (J / ψ), is determined as a function of the J / ψ transverse momentum in the range 2 < p T J / ψ < 15 GeV / c. The results are in excellent agreement with next-to-leading order non-relativistic expectations and show a significant discrepancy compared with the colour singlet model prediction at leading order, especially in the low p T J / ψ region

    Investigation of FEAR and MEN pathway homologs in multinucleate cells

    The cell cycle is a sequence of events enabling a cell to replicate and proliferate. Common landmark events in most eukaryotic cell cycles are duplication of the DNA, mitosis, and cell separation. The cell cycle lays the basis for development in multicellular organisms, and is of course important for cell or tissue renewal. Disregulation of the cell cycle can lead to uncontrolled growth and tumor formation. Mitosis, or nuclear division, is tightly regulated to ensure proper segregation of nuclei to daughter cells. In budding yeast (S. cerevisiae), the timing of mitosis is coupled to cytokinesis by the action of the FEAR and MEN regulatory networks, which mediate exit from mitosis by activation of the phosphatase ScCdc14. In this thesis, we investigate the function of homologs of these pathways in the filamentous fungus, Ashbya gossypii. This organism provides a unique cellular setting to study these matters, as it is a close relative of S.cerevisiae and shares most of the molecular machinery. However within its multinucleate hyphae, mitosis is uncoupled from cytokinesis, and cytokinesis is not followed by cell separation, thus leading to compartments containing 8-10 nuclei, separated by septa. We look into how exit from mitosis is affected in such a cellular environment where nuclei are free to divide without the spatial and temporal constraints as described in budding yeast, where each mitosis is followed by cytokinesis and cell separation. First we study the core component of exit from mitosis signalling. We examinethe phosphatase AgCdc14 and discover that its regulation is very similar to S. cerevisiae, with the phosphatase being sequestered within the nucleolus throughout interphase, and released during anaphase. Furthermore, we find it to be an essential component in nuclear cycle progression in A. gossypii, with nuclei in null mutants failing to undergo mitosis. We then look at MEN homologs in A. gossypii and present evidence that the kinase cascade function of the pathway is likely conserved. We however show MEN homologs to be non-essential and play no role in AgCdc14 regulation. On the other hand, we detect sporulation deficiencies in our mutants, which we could attribute to septation defects. More interestingly, we observe a partial mitotic arrest in MEN deficient cells. MEN homologs thus seem to have diverged from the primary role of their counter-parts in budding yeast. The final chapter deals with homologs of the FEAR pathway. It had been previously suggested that, in primitive cells, the task of Cdc14 phosphatase regulation may be solely bestowed upon this network of genes. We find this to hold true for A. gossypii, where we report severe mitotic defects upon deletion of FEAR homologs, and in particular, disruption of control over AgCdc14 release. We interpret the results as evidence for a simpler system regulating exit from mitosis in A. gossypii and lay out potential implications for the more complex system in S. cerevisiae
