Bu görüşme, Aziz Elbas ve Ahmet Erdönmez koordinatörlüğünde Bursa Büyükşehir Belediyesi bünyesindeki Bursa Araştırmaları Merkezi tarafından Çarşının Öyküsü projesi başlığı altında gerçekleştirilmiştir. Oturdukları evden bahseden Şekercioğlu, o dönemde mahallede 6-7 taksi olduğunu söyler. Mahalledeki sütçü ve yoğurtçular hakkında bilgi verir. Changes in essential oil composition, phenylalanine ammonia lyase gene expression and rosmarinic acid content during shoot organogenesis in cytokinin-treated Satureja spicigera (C. Koch) boiss. shoots
WOS: 000516134000001This study investigated the effects of different concentrations of zeatin and thidiazuron on the accumulation of phenolics and activity of phenylalanine ammonia lyase gene in the micropropagated Satureja spicigera shoots. S. spicigera clones were generated from cultured nodal segments on Murashige and Skoog including vitamins medium supported with zeatin and thidiazuron concentrations. Shoots were subcultured three times in the same medium and harvested at the end of the third month. The expression levels of phenylalanine ammonia lyase gene were conducted using semi-quantitative reverse transcription polymerase chain reaction technique. Rosmarinic acid contents were determined using reverse phase-high performance liquid chromatography with a diode array detector. Genetic stability of regenerants was performed using random amplified polymorphic DNA markers. Essential oils from shoots were identified by gas chromatography-mass spectrophotometry analysis. It has been detected that 0.1 mg/L thidiazuron is more effective on the node formation and shoot elongation. Contrarily, 4.0 mg/L thidiazuron caused a significant increase in shoot multiplication. The highest biomass accumulation was calculated from medium including 1.0 mg/L zeatin. According to random amplified polymorphic DNA analysis, all banding profiles from micropropagated plants were monomorphic. Gas chromatography-mass spectrophotometry analysis showed that rising concentrations of zeatin and thidiazuron increased the thymol and decreased the carvacrol content. It was also found that rosmarinic acid production increased significantly at 1.0 mg/L zeatin application. However, thidiazuron applications had no positive effect on rosmarinic acid accumulation. In vitro seed germination, plantlet growth, tuberization, and synthetic seed production of Serapias vomeracea (Burm. f.) Briq.
WOS: 000390711200004Orchids are considered recalcitrant plants in in vitro propagation. Due to the lack of appropriate micropropagation techniques for mass production and damage to their ecological distribution posed by local gatherers, these species are threatened with extinction, including Serapias vomeracea (Burm. f.) Briq. In this research, we put forward a complete micropropagation method covering in vitro micropropagation, synthetic seed formation, germination in soil, and acclimatization to ambient conditions. To the best of our knowledge this is the first report of successful synthetic seed formation and germination of S. vomeracea. Initially, seeds were germinated in different culture media and also media supplemented with different concentrations of plant growth regulators. Effects of plant growth regulators on tuber formation, glucomannan contents, and different growth parameters were evaluated throughout the study. The best germination rate (84.03%) was achieved on Orchimax including activated charcoal medium and supplemented with 2.0 mg/L 6-benzyladenine. The longest shoot elongation amongst plantlets was observed on the same medium supplemented with 0.25 mg/L thidiazuron, whereas 2.0 mg/L indole-3-butyric acid favored leaf formation. Higher indole-3-butyric acid concentrations were found to be more effective in the formation and elongation of roots. Orchimax medium supplemented with zeatin (2.0 mg/L) was superior to the others in terms of tuber formation and glucomannan content therein. Dactylorhiza urvileana'n n in vitro asimbiyotik çimlendirilmesi ve fidelerinin olu turulmas
Bu çal mada Orchidaceae familyas na ait Dactylorhiza urvillenana (Steud.) H.Baumann & Künkele'nin doku kültürü yöntemleriyle ço alt lmas amaçland . Ba lang ç eksplant olarak türe ait olgunla m tohumlar kullan ld . Sterilizasyonlar ve canl l k testleri yap lan tohumlar n çe itli temel besi ortamlar ndaki çimlenme, protokorm ve kallus olu um oranlar tespit edildi. Çimlenen tohumlardan olu an protokormlar, Zeatin'in farkl deri imleri ile desteklenen Orchimax aktif kömürlü besi ortam nda (OAM) organogeneze te vik edildi ve sürgün uzama miktarlar ve ortalama kök say lar belirlendi. Elde edilen sonuçlara göre, çimlenme, protokorm ve kallus olu umunda denenen temel besi ortamlar ndan Orchimax aktif kömürlü besi ortam n n daha etkili oldu u görüldü. Bu ortamdaki çimlenme oran %62,39, protokorm olu um oran %73,18 ve kallus olu um oran %8,6 olarak belirlendi. Farkl zeatin deri imlerinin sürgün uzamas ve kök say s üzerine etkilerine bak ld nda, 1,0 mg/L deri iminin di erlerinden daha olumlu sonuçlar verdi i görüldü ve bu ortamdan elde edilen ortalama sürgün boyu ve kök say s de erleri s ras yla 36,05 mm ve 3,4 adet olarak hesaplandIn this study, in vitro production of Dactylorhiza urvilleana (Steud.) H.Baumann &Künkele by using plant cell and tissue culture techniques were carried out. Mature seeds were used as starting explant. After the surface sterilization and viability tests, seeds were germinated in different basal culture media then germination, protocorm and callus formation rates were determined. Protocorms were encouraged to organogenesis in Orchimax medium including activated charcoal (OAM) and supported with different concentrations of Zeatin and shoot length and root numbers of seedlings were detected. It has been determined that the most effective culture medium for germination, protocorm and callus formation rates of seeds is the OAM. In this medium, germination, protocorm and callus formation rates were determined as 62.39%, 73.18%, 8.6%, respectively. It has been determined that the media including 1.0 mg/L Zeatin were effective on the elongation of shoots and root formation. In this work, a question related to the concept of "folk" which was suggested by Alan Dundes in his work "Who is Folk?" has been discussed.
