Genomic analysis of the function of the transcription factor gata3 during development of the Mammalian inner ear
No full textWe have studied the function of the zinc finger transcription factor gata3 in auditory system development by analysing temporal profiles of gene expression during differentiation of conditionally immortal cell lines derived to model specific auditory cell types and developmental stages. We tested and applied a novel probabilistic method called the gamma Model for Oligonucleotide Signals to analyse hybridization signals from Affymetrix oligonucleotide arrays. Expression levels estimated by this method correlated closely (p<0.0001) across a 10-fold range with those measured by quantitative RT-PCR for a sample of 61 different genes. In an unbiased list of 26 genes whose temporal profiles clustered most closely with that of gata3 in all cell lines, 10 were linked to Insulin-like Growth Factor signalling, including the serine/threonine kinase Akt/PKB. Knock-down of gata3 in vitro was associated with a decrease in expression of genes linked to IGF-signalling, including IGF1, IGF2 and several IGF-binding proteins. It also led to a small decrease in protein levels of the serine-threonine kinase Akt2/PKB beta, a dramatic increase in Akt1/PKB alpha protein and relocation of Akt1/PKB alpha from the nucleus to the cytoplasm. The cyclin-dependent kinase inhibitor p27(kip1), a known target of PKB/Akt, simultaneously decreased. In heterozygous gata3 null mice the expression of gata3 correlated with high levels of activated Akt/PKB. This functional relationship could explain the diverse function of gata3 during development, the hearing loss associated with gata3 heterozygous null mice and the broader symptoms of human patients with Hearing-Deafness-Renal anomaly syndrome

No full text“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

Like Daniela Zorzi, the author considers herself a socially engaged linguist, and in memory of her friend’s work, offers a reflection on the key roles of languages and cultures and of multilingualism for professional success and social welfare today

No full textNational audienceParis est une fête di Sylvain George è un'opera audiovisiva in cui sottraendo, se si vuole, il termine agli studi foucaultiani, la rivolta è disciplina giornaliera che spinge a reinterpretare le istanze dei più affamati ricordi, infinito ripetitore ed acceleratore di sommosse interrotte e di battaglie perdute (la rievocazione continua, benché stilizzata, dell’esperienza della comune di Parigi, come pure del vincente movimento contro il CPE, dell’emigrazione per mare del giovane Camara, e infine di chi non c’è più, vittima del terrore poliziesco, quando la camera di Sylvain George rende visita alla stele commemorativa di Zyed e Bouna, adolescenti morti a Clichy-sous-Bois nell’ottobre del 2005). La memoria in lotta della città di Parigi si esprime nelle testimonianze della vita delle popolazioni e in quelle delle vite singolari, nelle lunghe durate e negli istanti di riappropriazione (ed aiuta anche il buon montaggio del suono, curato da Ivan Gariel, come pure la raffinata presenza musicale dei Godspeed You! Black Emperor, maestri dell’uso della lunga durata in quel che ancora chiamiamo post rock). Delle lunghe maree e delle veloci tappe che scandiscono questa balade audiovisiva e notturna non sapremmo coglierne l’inizio o la fine, o prevederne la durata. Si tratta, piuttosto, di motivi tutti intrecciabili di un unico, politico, film de ville, che a detta dell’autore è anche un omaggio moderno a quelle sinfonie d’antica pellicola (Elie Lotar, Alberto Cavalcanti e Paul Strand in particolare, ma anche agli esperimenti di Marey, o al Jean Vigo di A proposito di Nizza)

No full textThis paper presents an environmental diagnosis based on public complaints on environmental
issues submitted to the Environmental Department of the Aveiro City Council, Portugal, between
2000 and 2005. It discusses the potential influences of these in local environmental planning
and governance. The paper has been organised into five sections. The first of these introduces
the study. The second section focuses on the conceptual approaches relating to environmental
grassroots movements, the main actors involved in these movements and the role played by
local government. It also contains a brief review of the most recent urban environmental
quality challenges in the European context together with a description of the main features of
the associated political and legal framework in Portugal. The third section describes the case
study and the methodology used. The results of the empirical study are detailed in the fourth
section. The final section critically analyses these results with emphases on the temporal
evolution of the submission of complaints, the actors involved, the local environmental problems
and their associated spatial pattern as well as the responses given by the City Council. This
information may then be used to provide a useful indicator for the perception of environmental
quality as well as a credible instrument for the visualisation and evaluation of local performance

No full textResumen: Daniela Parisi analiza el impacto de la vida de San F rancisco de Asís desde la perspectiva de la historia del pensamiento económico. Haciendo referencia particularmente a la atención otorgada en los círculos franciscanos a los signos de los tiempos, la autora traza el camino desde la vida de San Francisco, pasando por la vida de la Orden hasta el presente, y revela los orígenes del movimiento franciscano como un intento de reforma social y religiosa. En primer lugar, el artículo presenta la vida que llevó San Francisco como una “pobreza material voluntaria” en el contexto de los cambios socio-económicos que tuvieron lugar en el siglo XIII, con el advenimiento de la sociedad comercial. Luego, explica cómo la propuesta de San Francisco creció hasta convertirse en una orden religiosa. Finalmente, el artículo intenta iluminar aquellos aspectos en que la Orden Franciscana puede todavía considerarse un signo de los tiempos a través de una existencia comprometida con la pobreza, eliminando lo superfluo de nuestra vida y viviendo en consonancia con el Evangelio.Abstract: Daniela Parisi analyses the impact of the life of Saint Francis of Assisi, from the perspective of the history of economic thought. By referring in particular to the attention given in Franciscan circles to the signs of times, the author draws the path from Saint Francis’s life to the life of the Order until the present, and reveals the origins of the Franciscan movement as an attempt for social and religious reformation. First, the article presents St. Francis’s life of a “voluntary material poverty” in the context of the social and economic changes that took place in the XIII century, with the rise of the commercial society. Then, the author explains how St. Francis’ proposal grew into an Order. Finally, the article aims at illuminating the aspects in which the Franciscan Order can be still considered a sign of the times by living the commitment to poverty and minority, eliminating the superfluous from our lives and living close to the Gospel
