Espólio de José Leite de Vasconcelos: corpus linguístico: o «Dialeto Brasileiro»
Este relatório tem como foco o desenvolvimento e criação de um corpus de provas tipográficas pertencentes ao espólio1 de José Leite de Vasconcelos conservado no Museu Nacional de Arqueologia.
Os pacotes2, que compõem o Legado de Leite de Vasconcelos, utilizados neste projeto foram os respeitantes à matéria de Filologia3.
Numa primeira fase, identificou-se e organizou-se os materiais por tema e criou-se um catálogo dos mesmos para facilitar a posterior pesquisa. Numa segunda fase, fez-se fotografia de todas as provas tipográficas encontradas e conseguinte atribuição de cotas e edição de imagens. Ulteriormente, identificou-se a que obras pertenciam as provas e elaborou-se com essa informação tabelas de «correspondência»4.
Por fim, este trabalho terá uma parte mais interpretativa, onde o foco será o chamado «dialeto brasileiro». Este foi um tema pouco tratado por Leite de Vasconcelos e, no entanto, teve relevância bastante para estar presente nos seus apontamentos e investigações. Foram encontrados, em específico, dois envelopes, que continham informação recolhida pelo autor, sobre este tema. Daí enveredou-se pelo estudo do seu conteúdo, fazendo uma descrição detalhada dos suportes materiais lá contidos.This report focuses on developing and creating a corpus of typographical proofs belonging to the estate of José Leite de Vasconcelos kept at the National Museum of Archaeology.
The parcels which compose Leite de Vasconcelos' Legacy used in this project were those related to Philology.
In the first phase, the materials were identified and organized by theme, and a catalog of them was created to facilitate later research. In a second phase, all the typographical proofs found were photographed, and, consequently, quotations and image editing were done. Later, the works to which the proofs belonged were identified and "correspondence" tables were created with this information.
Finally, this work will have a more interpretative part, focusing on the so-called "Brazilian dialect". This dialect was a theme little dealt with by Leite de Vasconcelos, and, nevertheless, it was relevant enough to be present in his notes and research. The archaeological explorations carried out in Monte Real (Leiria) in 1865 by Frederico Augusto de Vasconcelos Pereira Cabral or the history of a schist prehistoric engraved plaque
In this paper we publish the report prepared in 1866 by Frederico de Vasconcelos Pereira Cabral (1820-1886) after his visit in 1865 to Monte Real, concerning the characterization of the geological conditions of a funerary deposit of Neolithic age occasionally found in the previous year and whose human remains were then buried in the local cemetery.
Among the scarce spoils that the author obtained in 1865 from the inhabitants of Monte Real, there is a schist plaque that was, together with the exemplars recovered in the excavations of Nery Delgado held in the same year in the cave of the Casa da Moura the first to be duly recognized as prehistoric artifacts.
The scientific quality of this report would suffice to place its author among the pioneer archaeologists of Portugal, confirming his priority in other contributions of a geological and archaeological nature, such as the publication in 1881 of the first Paleolithic artefacts collected in Portuguese territory.
This study is also a contribution to draw attention to the pioneer activity of its author in the field of Archeology, as a member of the Geological Commission of Portugal.info:eu-repo/semantics/publishedVersio

Luís Mendes de Vasconcelos publishes in 1608 Do Sítio de Lisboa. Diálogos [About Lisbon. Dialogues]. He hoped that King Philip III of Spain, and II of Portugal, would turn Lisbon into the head of the Hispanic Empire. It situates the Dialogue between an advisor to the king, a captain, and a philosopher in the reign of D. João III, drawing inspiration, as we affirm, from historical figures of that time.

Os 4P's do design: uma proposta metodológica não linear de projeto
Tese (doutorado) - Universidade Federal de Santa Catarina, Centro Tecnológico. Programa de Pós-Graduação em Engenharia de Produção.A tese apresenta uma Metodologia de Projeto em Design, de estrutura não linear proposta para o ensino e mercado profissional. A grande maioria das metodologias de projeto hoje utilizadas na academia tem características similares de linearidade com foco no problema. A metodologia apresentada na tese não invalida a importância da resolução de problemas durante a execução de um projeto, porém, a máxima: "defina primeiro o problema" é questionada e sugere que o processo metodológico deve seguir o pensamento de seu executor (designer). É discutida a linearidade do pensamento humano e apresentada uma metodologia não linear baseada em 4 etapas do processo de design, que podem ser gerenciadas de acordo com o pensamento de um grupo colaborativo de profissionais ou acadêmicos. Algumas metodologias tradicionais são apresentadas para fundamentar a tese de que o pensamento humano não é linear e imprescinde de estrutura fechada para obter resultados organizados e de acordo com as expectativas do mercado. Como o mercado de design está cada dia mais se profissionalizando, e essa profissionalização passa pela Academia, a metodologia é apresentada de forma didática e com objetivo no ensino superior de design. Porém, o ensino está mudando e a Universidade tem buscado novas alternativas para ampliar seus horizontes. A Educação à Distância através da rede mundial de computadores (Internet) está cada vez mais presente no cotidiano de educadores e estudantes dos mais diferentes níveis. O Design não pode ficar alheio a esse processo evolutivo. Essa pesquisa então, através de avaliação e uso de ferramentas já disponíveis na rede e da gestão integrada do design baseada na metodologia "4 P's do Design", procura sugerir, com apoio de um grupo de teste real, um Ambiente Virtual de Aprendizagem baseado em trabalho colaborativo através da Internet

Polysaccharides from plant cell walls are the most abundant biomass on Earth and are important resource for biofuel production. The main components of plant cell walls are cellulose, hemicellulose and lignin. Hemicellulose is the second most abundant component of renewable biomass and as arabinoxylan, is mainly composed of xylose and arabinose. Filamentous fungi, such as Trichoderma reesei, produce gram-per-liter levels of glycoside hydrolases (GHs). GH enzymes are required for hydrolysis of the glycosidic bonds present in complex polysaccharides, releasing fermentable sugars. In this project, we investigated the biochemical characteristics of two endoxylanases in two model filamentous fungi, Neurospora crassa and Aspergillus nidulans. Putative endoxylanase genes from N. crassa (ncu05924) and A. nidulans (an1818) were expressed homologously and heterologously in both filamentous fungi. Here, we demonstrate that A. nidulans was able to expressed and secrete at the same levels, both the recombinant homologous (AN1818) and heterologous (NCU05924) proteins, while N. crassa expressed the recombinant homologous protein at 26-fold more than the recombinant heterologous protein. All 4 endoxylanases had similar optimal pH (~5.8) and temperature (50 to ~55°C), similar secondary structures, and comparable glycosylation patterns. High performance liquid chromatography (HPLC) was used to identify the end products released by each enzyme from xylan substrates. The specific activity of AN1818 was ~50% higher than NCU05924 on different model xylans.
Xylan degrading enzymes from human colonic bacteroides intestinalis1
Many human diets contain arabinoxylan, and the ease of genome sequencing coupled with reduced cost have led to unraveling the arsenal of genes utilized by the colonic Bacteroidetes to depolymerize this polysaccharide. The colonic Bacteroidetes with potential to ferment arabinoxylans include Bacteroides intestinalis. In this study, we analyzed the hydrolytic activities of members of a xylan degradation cluster encoded on the genome of Bacteroides intestinalis DSM 17393. Here, it is demonstrated that a cocktail of the xylanolytic enzymes completely hydrolyze arabinoxylans found in human diets. Fascinatingly, this bacterium and other relatives have evolved and secrete a unique bifunctional endoxylanase/arabinofuranosidase in the same polypeptide. The bifunctional enzyme and other secreted enzymes attack the polysaccharides extracellularly to remove the side-chains, exposing the xylan backbone for cleavage to xylo-oligosaccharides and xylose. These end products are transported into the cell where a β-xylosidase cleaves the oligosaccharides to fermentable sugars. While our experiments focused on B. intestinalis, it is likely that the extracellular enzymes also release nutrients to members of the colonic microbial community that practice cross-feeding. Polysaccharide utilization by the human colonic bacterium, bacteroides intestinalis DSM 17393
The human gastrointestinal microbiome has co-evolved with the host for an extended period, generating an intrinsic metabolic network capable of modulating both the microbial community and host physiology. Changes in the gut environment shape the microbial community with one of the main driving characteristics being competition for nutrients. A key aspect for nutrient acquisition in the gut is the ability of the microbes to degrade and ferment dietary fiber. These chemically diverse polysaccharides are the main components of the plant cell wall and function as the primary energy source for the microbiota. The dietary polysaccharides are largely indigestible by the digestive enzymes of the host. The phylum Bacteroidetes, one of the main colonizers of the gastrointestinal tract (GIT), has evolved one of the largest arrays of carbohydrate-associated enzymes (CAZymes) responsible for degrading polysaccharides. Furthermore, these bacteria have highly organized gene clusters, known as polysaccharide utilization loci (PUL), encoding the enzymes required for the degradation and transport of the sugar components of dietary fiber. These PULs also possess an unusual regulatory mechanism found only in Bacteroidetes, similar to the canonical two-component system encoded by organisms in all three domains of life. The two-component system is involved in signal transduction, mediated by a histidine-kinase sensor and a response regulator. Interestingly, the Bacteroidetes encode this system in a single polypeptide that can be delineated into a sensor, a Y_Y_Y, a histidine-kinase, a histidine-ATPase, and response regulator modules, respectively.
Through our work, we aim to understand the degradation of esterified arabinoxylan by diverse Bacteroides spp. and to develop a rapid approach for identifying the potential target of a vast number of uncharacterized PULs. Our findings demonstrate that some Bacteroides spp. highly express an esterase-enriched cluster during growth on esterified arabinoxylan compared to the mixture of its component monosaccharides xylose and arabinose. Biochemical analysis of the proteins encoded by the esterase-enriched cluster demonstrated diverse enzymatic activities and substrate specificities capable of working synergistically to fully depolymerize complex arabinoxylan substrates. Interestingly, the hypothetical domain encoded by BACINT_01040 demonstrated one of the most versatile feruloyl esterase activities, and the enzyme was able to cleave the ferulic acid side chain of every substrate tested in our studies. Moreover, we demonstrated that the bacteria harboring the esterase-enriched cluster do not metabolize the ferulic acid during growth on wheat bran. Thus, the plant phenolic compound accumulated in the spent medium. Furthermore, the accumulated ferulic acid was able to modulate the immune system and induce a Th1-type immune response and viral defenses in both gastrointestinal cell lines and mouse model.
Due to the vast number of uncharacterized PULs encoded in the microbiome, it is important to develop a rapid means to assign function to these gene clusters. Our current hypothesis relies on the observation of a potential endo-acting enzyme near the susC/susD-like genes. We hypothesized that these enzymes “scout” the environment for their associated target polysaccharides. In this work, we biochemically characterized the activity of two “scouting enzymes” on arabinoxylan and arabinan, thus showing that a PUL-associated hybrid two-component system (HTCS) sensor domain is able to bind and sense the products of its associated scouting enzyme. This hypothesis was further corroborated by transcriptomic analysis of B. intestinalis grown on each polysaccharide compared to its monomeric components. Thus, the results demonstrated in this study may advance the understanding of polysaccharide utilization by Bacteroidetes species by rapidly assigning function to uncharacterized PULs. Deslocamentos de fronteiras: percurso e produção musical de Gonzagão e Gonzaguinha
O presente trabalho pretende discutir de que forma os percursos, permeados por diversas fronteiras que hierarquizam saberes, poderes e enunciados, interferiram na produção artística de dois músicos brasileiros, oriundos de lugares historicamente marginalizados no Brasil, a saber: Luiz Gonzaga do Nascimento (conhecido como Gonzagão), vindo do sertão de Pernambuco para ganhar as paradas de sucesso no Rio de Janeiro nos anos 1940/50 e Luiz Gonzaga do Nascimento Júnior (o Gonzaguinha), nascido no Morro de São Carlos/Rio de Janeiro, que alcançou grande sucesso entre as décadas de 1970/80. Interessa investigar quais estratégias e táticas cada um desses artistas utilizou para driblar as fronteiras que separam o "centro" e a "periferia" e alcançar o cobiçado panteão da Música Popular Brasileira. Tal discussão é parte da pesquisa de doutorado em Estudos de Cultura (em curso) intitulada: Deslocamentos de fronteiras: percurso e produção musical de Gonzagão e Gonzaguinha na construção das brasilidades, realizada na Universidade de Lisboa. Para compreender a relação entre música, percurso e identidade, assim como para pensar a fronteira como construção simbólica serão tomados como aportes teóricos conceitos e reflexões oriundos do pensamento pós-colonial e decolonial
