Abstract The 2S light chains of mammalian and avian slow muscle myosin, indistinguishable by two-dimensional gel electrophoresis, have been examined by peptide mapping. The fragments obtained with S. aureus V8 protease were analysed either by gel electrophoresis or by reverse-phase high performance liquid chromatography. The peptide maps of avian 2S light chains contain fragments distinct from those of mammalian 2S light chains. Chicken and turkey LC2S appear to be more similar to each other than those from mammalian species (rat and rabbit). These results are in agreement with the relative phylogenetic distances among the four species studied here. The 2S light chain of slow muscle represent further examples of polypeptides which comigrate in two-dimensional gel electrophoresis in spite of their different peptide maps.     El daño ambiental causado por la cantidad de Residuos Sólidos Urbanos (RSU) generados hace necesaria una rápida intervención para contener las actividades antropogénicas. La cuestión de la reducción y del reciclaje de residuos es, por tanto, una de las áreas fundamentales de investigación de nuestra actualidad. Este documento, fruto de la tesis doctoral, aún en curso, de Dolores Carvajal Caro, La industria del reciclaje en Europa. El uso de residuos plásticos sólidos urbanos en Arquitectura., y de la tesis fin de carrera de Giorgia Betto, Construir con Residuos. El uso de materiales plásticos reciclados como materia prima para la realización de un sistema constructivo ecosostenible., tiene como objetivo ilustrar la actual situación de los residuos en el ambiente, centrándose en los residuos plásticos y, en particular, en la importancia de su gestión, subrayando su potencial como materia prima para la realización de componentes para la construcción mediante su reciclaje y reutilización. Como resultado del análisis y comparación de los datos sobre RSU, y del estudio de diversos métodos sostenibles de gestión y reutilización de residuos plásticos en la construcción, este documento presenta además el diseño y la realización de un bloque en plástico reciclado, fruto de la tesis de Betto, que sigue los objetivos introducidos en 2015 por la Economía Circular de reintroducir materiales en nuevos ciclos de producción. Se trata un componente de construcción cuyo uso podría limitar la dispersión de residuos plásticos en el medio ambiente, explotando su potencial en el sector de la construcción. The environmental damage caused by the amount of Solid Urban Waste (MSW) generated rapid intervention to contain anthropogenic activities. The issue of waste reduction and recycling is, therefore, one of the fundamental areas of research today. This document, result of the Dolores Carvajal Caro’s PhD research, still in progress, The recycling industry in Europe. The use of solid urban plastic waste in architecture., and Giorgia Betto’s thesis degree Build with Waste. The use of recycled plastic materials as raw material for the realization of an eco-sustainable construction system., shows the current situation of waste in the environment, focusing on plastic waste and, in particular, on the importance of its management, underlining its potential as a raw material for the realization of components for construction through recycling and reuse. As a result of the analysis and comparison of the data on MSW, and the study of various sustainable methods of management and reuse of plastic waste in building sector, this document also present the design and realization of a block in recycled plastic, the result of the Betto’s thesis, which follow the objectives introduced in 2015 by the Circular Economy to reintroduce materials in new production cycles. It is a construction component whose use could limit the dispersion of plastic waste in the environment, exploiting its potential in the construction sector

    Characterization of the ATP-hydrolysing activity of α-sarcoglycan

    α-Sarcoglycan is a glycoprotein associated with the dystrophin complex at sarcolemma of skeletal and cardiac muscles. Gene defects in α-sarcoglycan lead to a severe muscular dystrophy whose molecular mechanisms are not yet clear. A first insight into the function of α-sarcoglycan was obtained by finding that it is an ATP-binding protein and that it probably confers ability to hydrolyse ATP to the purified dystrophin complex [Betto, Senter, Ceoldo, Tarricone, Biral and Salviati (1999) J. Biol. Chem. 274, 7907–7912]. In the present study, we present definitive evidence showing that α-sarcoglycan is an ATP-hydrolysing enzyme. The appearance of α-sarcoglycan protein expression was correlated with the increase in ecto-nucleotidase activity during differentiation of C2C12 cells. Approx. 25% of ecto-nucleotidase activity displayed by the C2C12 myotubes was inhibited by preincubating cells with an antibody specific for the ATP-binding motif of α-sarcoglycan. This demonstrates that α-sarcoglycan substantially contributes to total ecto-nucleotidase activity of C2C12 myotubes. To characterize further this activity, human embryonic kidney 293 cells were transfected with expression plasmids containing α-sarcoglycan cDNA. Transfected cells exhibited a significant increase in the ATP-hydrolysing activity that was abolished by the anti-α-sarcoglycan antibody. The enzyme had a substrate specificity for ATP and ADP, did not hydrolyse other triphosphonucleosides, and the affinity for ATP was in the low mM range. The ATPase activity strictly required the presence of both Mg2+ and Ca2+ and was completely inhibited by suramin and reactive blue-2. These results show that α-sarcoglycan is aCa2+,Mg2+-ecto-ATPDase. The possible consequences of the absence of α-sarcoglycan activity in the pathogenesis of muscular dystrophy are discussed

    The research work has dealt primarily with the analysis of features from Remote Sensing interpreted as roads in the Southern Valli Grandi Veronesi (Verona Province). The area is known in the literature as a central area of the local terramare "enclave" and, secondly, for the presence of a Roman landscape of marginal importance. The particular conditions of visibility of remote detected buried structures, resulting from prolonged neglect of the area due to extensive reclamation of the marshes until the midnineteenth century, configure the area as being particularly suited to the development of all the methods of investigation of Remote Sensing and surveys techniques. The reaserch was carried out by Remote Sensing on the widest range of platforms available (current and historical aerial photographs, digital orthophotos, satellite images, digital elevation models at different resolution) and through the more refined treatment and processing of images (image enhancing, image processing), or through the use of programs dedicated to the automatic and semi-automatic recognition of anomalies. The methodology of reading, organization and interpretation of features related to constructs road, but also to a number of features that can be interpreted as agricultural channels and ditches, crossed with field data has allowed us to propose a reconstruction of the unexpected complex landscape of the Bronze Age
