Phase I and Pharmacokinetic Studies of CYT-6091, a Novel PEGylated Colloidal Gold-rhTNF Nanomedicine

Purpose: A novel nanomedicine, CYT-6091, constructed by simultaneously binding recombinant human tumor necrosis factor alpha (rhTNF) and thiolyated polyethylene glycol to the surface of 27-nm colloidal gold particles, was tested in a phase I dose escalation clinical trial in advanced stage cancer patients.
Experimental Design: CYT-6091, whose dosing was based on the amount of rhTNF in the nanomedicine, was injected intravenously, and 1 cycle of treatment consisted of 2 treatments administered 14 days apart.
Results: Doses from 50 μg/m2 to 600 μg/m2 were well tolerated, and no maximum tolerated dose (MTD) was reached, as the highest dose exceeded the target dosage of 1-mg rhTNF per treatment, exceeding the previous MTD for native rhTNF by 3-fold. The first 2 patients on the study, each receiving 50 μg/m2, did not receive any prophylactic antipyretics or H2 blockade. A predicted, yet controllable fever occurred in these patients, so all subsequently treated patients received prophylactic antipyretics and H2 blockers. However, even at the highest dose rhTNF's dose-limiting toxic effect of hypotension was not seen. Using electron microscopy to visualize nanoparticles of gold in patient biopsies of tumor and healthy tissue showed that patient biopsies taken 24 hours after treatment had nanoparticles of gold in tumor tissue.
Conclusions: These data indicate that rhTNF formulated as CYT-6091 may be administered systemically at doses of rhTNF that were previously shown to be toxic and that CYT-6091 may target to tumors. Going Beyond Counting First Authors in Author Co-citation Analysis
Get PDFThe present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Data yang diperoleh dari putusan Hakim terhadap permohonan harta bersama di Pengadilan Agama Kabupaten Malang Nomor : 6091/Pdt.G/2013/PA.Kab.Mlg, yang mana menetapkan bagian suami lebih besar dalam pembagian harta bersama.Isi dalam putusan No. 6091/Pdt.G/2013/PA.Kab.Mlg. adalah suami menggugat cerai terhadap istri. Pembahasan dalam skripi ini, menitik beratkan pada pembagian harta bersama yang mana harta bersama tersebut hasil dari suami istri selama dalam ikatan perkawinan namun ketika terjadi perceraian harta bersama yang berupa sebuah tanah seluas 114 M2 dan diatasnya berdiri sebuah bangunan, yang terletak di Jalan Raya Sengkaling Desa Mulyoagung Kecamatan Dau Kabupaten Malang, melihat perkara ini majlis hakim mempunyai pertimbangan bahwasannya walaupun kaduanya (suami istri) ikut andil dan berpartisipasi dalam perolehan harta bersama namun suami yang lebih berhak mendapatkan bagian yang lebih besar dari istri karena istri telah menguasai harta bersama tersebut selama 2 tahun serta harta bersama tersebut di jadikan lahan usaha oleh istri akan tetapi hasil dari usaha tersebut tidak dibagi kepada suami. Pembagian harta bersama yang tidak seusai dengan ketentuan KHI Pasal 97 “janda atau duda cerai masing-masing berhar seperdua dari harta bersama sepanjang tidak ditentukan lain dalam perjanjian perkawinan, dengan dasar keadilan dan mengesampingkan KHI. Maka hakim memberikan putusan suami mendapat bagian lebih besar dari istrinya yaitu 2/3 untuk suami dan 1/3 untuk istri
