    Replication Data for Exploring the fate of antibody-encoding pDNA after intramuscular electroporation in mice

    This dataset contains raw data contributing to the publication 'Exploring the fate of antibody-encoding pDNA after intramuscular electroporation in mice'. This publication is about DNA-based antibody therapy, which seeks to administer the encoding nucleotide sequence, rather than the antibody protein. To further improve the in vivo monoclonal antibody (mAb) expression, a better understanding of what happens after the administration of the encoding plasmid DNA (pDNA) is required. This study reports the quantitative evaluation and localization of the administered pDNA over time and its association with corresponding mRNA levels and systemic protein concentrations. pDNA encoding the murine anti-HER2 4D5 mAb was administered to BALB/c mice via intramuscular injection followed by electroporation. Muscle biopsies and blood samples were taken at different time points (up to 3 months). In muscle, pDNA levels decreased 90% between 24 hours and one week post treatment. In contrast, mRNA levels remained stable over time. 4D5 antibody plasma concentrations reached peak levels at week two followed by a slow decrease (50% after 12 weeks). Evaluation of pDNA localization revealed that extranuclear pDNA was cleared fast whereas the nuclear fraction remained relatively stable. This is in line with the observed mRNA and protein levels over time, and indicates that only a minor fraction of the administered pDNA is ultimately responsible for the observed systemic mAb levels. Therefore, efforts to increase the protein levels upon pDNA-based gene therapy should focus on strategies to increase both cellular entry and migration of the pDNA into the nucleus. The currently applied methodology can be used to guide the design and evaluation of novel plasmid-based vectors or alternative delivery methods, to achieve a robust and prolonged protein expression

    2012 National Assessment of Information Acquisition and Processing in Primary Education in Flanders

    In 2012 a national assessment was carried out, which was commissioned by the Flemish Government, to assess whether pupils have sufficient skills to acquire and process information at the end of primary education. Data were collected through two written tests: a test on working with tables and graphs and a test on working with plans and drawings. A representative sample of 2383 pupils from 92 Flemish primary schools participated. In addition, the extent to which pupils have mastered certain ICT competencies was assessed through a ICT prformance assessment. A subsample of six pupils from each school participated in this performance assessment. Tests were supplemented with background questionnaires for pupils, parents, teachers and ICT coordinators. The pupil and parent questionnaires collected, among other things, information on socio-economic background, diagnosis of disabilities and circumstances that may stimulate learning. The questionnaire for teachers collected, among other things, information on their teaching experience, the use of educational materials and ICT during the classes. The questionnaire for the ICT coordinator asked about the ICT policy and infrastructure at school. The response rates to these questionnaires were almost 100% for the pupil questionnaire, 93% for the parent questionnaire, 95% for the teacher questionnaire and 93% for the ICT coordinator.In 2012 vond een peiling plaats in opdracht van de Vlaamse overheid om na te gaan in welke mate leerlingen op het einde van het basisonderwijs beschikken over voldoende vaardigheden om informatie te verwerven en verwerken. De gegevens werden verzameld door middel van twee schriftelijke toetsen: een toets over het werken met tabellen en grafieken en een toets over het werken met plannen en tekeningen. Deze toetsen werden afgenomen bij een representatieve steekproef van 2383 leerlingen uit 92 Vlaamse basisscholen. Daarnaast werd ook nagegaan in hoeverre leerlingen bepaalde ICT-competenties beheersen door middel van een praktische proef. Deze werd afgenomen bij een deelsteekproef van zes leerlingen per school. Naast de toetsen werden ook vragenlijsten afgenomen bij leerlingen, ouders, leerkrachten en ICT-coördinatoren. De leerling- en oudervragenlijsten verzamelden onder meer informatie over de sociaal-economische achtergrond, eventuele diagnoses van beperkingen en omstandigheden die het leren kunnen stimuleren. Leerkrachtvragenlijsten verzamelden informatie over hun diploma, hun onderwijservaring, het gebruik van leermiddelen en ICT tijdens de lessen. Vragenlijsten voor de ICT-coördinator gingen over het ICT-beleid en de infrastructuur op school. De respons op deze vragenlijsten was bijna 100% voor de leerlingen, 93% voor de ouders, 95% voor de leerkrachten en 93% voor de ICT-coördinator

    Data and table for paper titled "Bidirectional Onboard Chargers for Electric Vehicles: State-of-the-Art and Future Trends" in IEEE Transactions on Power Electronics (10.1109/TPEL.2023.3319996) Data was collected through a systematic search using search strings in IEEE Xplore, MDPI, and Springer. Over 500 publications are analysed for the following criteria: A) galvanic isolation, B) experimental setup with measurements of at least the operating efficiency, C) rated power of >1 kW, D) bidirectional, E) published in English language, F) published in peer-reviewed paper or the manufacturer's documentation. On-board charger prototypes from academia, industry, and OEM electric vehicle manufacturers are included. This data is discussed and represented graphically in written documents. Hence this repository serves as full, transparent documentation of the extracted data and support for future research, statistical analyses, and contributions

    Power during functional tests in older adults: reliability, reference values and relationship to other sarcopenia-related outcomes

    Aging is associated with declines in lower-limb muscle strength and power. Muscle power (i.e., force x velocity) starts to decline earlier in life and at a faster rate compared to muscle mass and strength. Low values of muscle power in old age have been associated with mobility limitations and mortality, stressing the importance to include muscle power assessments to prevent these negative outcomes. Lower-limb power is typically measured with specialized and expensive exercise equipment that requires skilled lab personnel (e.g., pneumatic or isotonic resistance machines, force plate analysis, Nottingham Power Rig) and is often limited to single joint movements (e.g., isokinetic dynamometers). These measurements are unable to capture the complex functioning of muscles in daily life activities and large-scale applicability in clinical settings is limited. Clinically feasible measurements of lower-limb power should be easy to administer, limited in time, low cost, functionally relevant and suitable for elderly. In this regard, simple field tests, such as sit-to-stand and stair-climbing tests, are preferable. In addition to accurate timing, inertial measurement units can provide information on sub-phase durations and kinematic properties of a functional movement. Previously, researchers have used sensor-based trunk kinematics to estimate the vertical power required to elevate the body’s center of mass during a transfer from sit to stand with a single sensor worn at the waist. Recently, we have used a similar approach to evaluate power during stair ascent in healthy adults (20-70 years). Results showed that stair ascent power is strongly related to leg-extensor power (r = 0.80), underlining the added value of sensor-based analysis of power over duration parameters (r = -0.53 with leg-extensor power). However, the reliability of the method, its sensitivity to detect age-related differences and its link to other sarcopenia-related outcomes remains to be investigated. In addition, many clinical settings have limited space or no access to a full flight of stairs. A simple standardized stair model of 3 steps with fixed dimensions would improve applicability in clinical settings. To our knowledge, no research has investigated the potential of a sensor-based analysis of stair ascent power on a 3-step stair model

    2022 National Assessment of Dutch in the First Grade of Secondary Education (A- and B-stream) in Flanders

    In 2022 a national assessment was carried out, which was commissioned by the Flemish government, to assess the extent to which students have achieved the attainment targets for Dutch (reading, listening and writing) at the end of the first grade of secondary education. Data were collected through two digital tests for reading and listening. In addition, a digital practical test was administered to assess students' writing skills. A representative sample of 1520 students from the A-stream and 4048 students from the B-stream participated. Tests were supplemented with background questionnaires for students, parents, teachers, and the coordinator of the didactic team for Dutch. The questionnaires collected, among other things, information on general background characteristics of the students and their families, characteristics of the teachers and the Dutch language curriculum. The response rates to the student questionnaire were 95% for the A-stream and 84% for the B-stream. For the parent questionnaire, the response rates were 91% and 71%, respectively. For the teacher questionnaires, the response rates were 87% and 73%, respectively. For the coordinators of the didactic teams, the response rates were 72% and 63%, respectively.In 2022 vond een peiling plaats in opdracht van de Vlaamse overheid om na te gaan in welke mate leerlingen op het einde van de eerste graad van het secundair onderwijs de eindtermen voor Nederlands (lezen, luisteren en schrijven) hebben behaald. De gegevens werden verzameld door middel van twee digitale toetsen voor lezen en luisteren. Daarnaast werd ook een digitale praktische proef afgenomen om de schrijfvaardigheid van de leerlingen te evalueren. Aan de peiling nam een representatieve steekproef van 1520 leerlingen uit de A-stroom en 4048 leerlingen uit de B-stroom deel. Naast toetsen en praktische proeven werden ook vragenlijsten afgenomen bij leerlingen, ouders, leerkrachten en de coördinator van de vakgroep Nederlands. Deze vragenlijsten verzamelden onder andere informatie over de algemene achtergrondkenmerken van de leerlingen en hun gezin, kenmerken van de leerkrachten en het onderwijsaanbod Nederlands.     Dataset and supplementary material related to the Accountability in Research paper describing the results of the PRICe survey (Perceptions of Research Integrity Culture) at KU Leuven. In the PRICe project, a questionnaire assessing KU Leuven's research integrity culture was developed and launched university-wide. The questionnaire assessed respondents' attitudes towards research integrity, as well as the research culture in their immediate research environment (research group / lab / unit), their department and the university as a whole. Specific emphasis was placed on the mentoring and supervision of junior researchers. The supplementary material contains the data, 7 supplementary figures, as well as the questionnaire in pdf and Qualtrics-format and the communication surrounding the survey.
