In situ gel: A Review
Oral route is technique which is used over a decades. It is most preferred and common technique for oral administration of drug in the body, but due to certain limitation such as absorption of drug, drug targeting to particular organ can cause problem for administration through oral route. To overcome these types of problem as well as for improvement of drug safety and efficiency a novel approach is developed for delivery of drug i.e. In-situ Nasal Drug Delivery System. In-situ gel is a process in which sol form before administration in the body, but once administrated, it undergo gelation in-situ, to form gel. Nasal drug delivery is one of alternative viable route of drug delivery is one of alternative and viable route of drug delivery. Nasal route is rich in vasculature and highly permeable. Nasal route is suitable for those drugs whose oral administration is problematic due to gastric irritation. The present review focused on anatomy of nasal system and criteria required of drug candidate to prepare a gel i.e. In-situ gel. Evaluation of Vitamin B1, B2 and B6 Tablets in Bangladesh by UV-Vis Spectrophotometer
Thiamine hydrochloride (vitamin B1), riboflavin (vitamin B2) and pyridoxine hydrochloride (vitamin B6) were assayed in pharmaceutical dosages form. Thiamine hydrochloride of 100 mg dose of six different companies, riboflavin of 5 mg dose of five different companies and pyridoxine hydrochloride 20 and 25 mg dosages of two different companies of Bangladesh were extracted from the tablets, cleaned up and their active ingredients were evaluated by UV-Vis spectrophotometer at 432, 445 and 292 nm, respectively. Evaluation was carried out with respect to calibration curves of their standard reference samples. Amount of thiamine hydrochloride in 100 mg tablets of six different companies were found to be in the range of 37.62±1.11 mg to 79.03±0.42 mg. For riboflavin in 5 mg tablets of five different companies, active ingredient was found in the range of 6.78±0.19 mg to 8.23±0.15 mg. In case of pyridoxine hydrochloride tablets, it was found that 20 mg tablet of one company contained 21.75±0.41 mg and 25 mg tablet of another company contained 29.72±0.59 mg of active ingredient. Recovery experiments were done by spiking excipients of the respective medicines at three different concentration levels with 5 replicate studies. Correlation coefficients (r2) were found to be 0.99 for three vitamins and recoveries were 82.26± 0.10,99.61± 0.06 and 109.91±0.12 for thiamine hydrochloride (vitamin B1), riboflavin (vitamin B2) and pyridoxine hydrochloride (vitamin B6) respectively
Preliminary Study of Hyphomycetes of Central India.
During the preliminary investigation of hyphomycetes from central India eight species of freshwater and freshwater-borne Hyphomycetes encountered in foam samples collected from the different streams and rivers. These fungi namely viz., Wiesneriomyces laurinus Kirk, Flabellospora multiradiata Nawawi, Isthmotricladia gombakiensis Nawawi, Triscelophorus acuminatus Nawawi, Flabellospora acuminata Descals and Webster, Sporidesmium leonense M.B. Ellis, Acrodictys globulosa Toth M.B. Ellis, and Acladium phoenicis Ramachar are being reported for the first time from central India. Brief notes and illustrations are also provided along with geographical distributions of these fungi in India
Neuroprotective Effect of Cinamomum zeylanicum in streptozotocin induced diabetes in Mice
Background: Diabetes is recognized as one of the leading causes of morbidity and mortality and representing as an emerging global epidemic worldwide. Diabetes and stroke both are the conditions which share various common threats. A patient with Diabetes has higher risk of stroke as compared to non Diabetics. Diabetes and stroke both affects the blood vessels. There are several biochemical pathways which are responsible for the developments of vascular complications due to hyperglycemia. Thus prevention and treatment of diabetes and stroke can reduce the risk of various vascular diseases. Cinnamomum zeylanicum has been used for the treatment of diabetes since long time. Cinnamomum zeylanicum rich in phenolic contents (antioxidants) have been identified as a major active component exhibiting antioxidant, and neuroprotective effects. The present study evaluate the neuroprotective effect of Cinnamomum zeylanicum in streptozotocin induced diabetes in mice and experimentally induced global cerebral ischemia/reperfusion injury in mice. Materials and methods: Swiss albino mice (male) weighing 20-30 g were randomized into eight groups. Diabetes was induced by administering streptozotocin (65 mg/kg, i.p).Starting from seven days after streptozotocin injection, insulin and Cinnamomum zeylanicum were administered for 3 weeks. Serum glucose level and body weight were measured weekly. Neuroprotective activity was carried out by global cerebral ischemia on swiss albino mice by carotid artery occlusion for 10 min followed by 24 hr. reperfusion. Measurement of Triglycerides, Malondialdehyde , cerebral infarct size and glutathione level was carried out at the end of the study. Results: After 21 days of treatment with Cinnamomum zeylanicum (75 mg/kg, 150 mg/kg and 300 mg/kg, p.o) significant increase in body weight and glutathione level and a significant reduction in serum glucose level, triglycerides, MDA levels and cerebral infarct size was observed as compared to diabetic control and diabetic+ stroke group. Conclusion: The present study concludes that Polyphenolic fraction of Cinnamomum zeylanicum not only attenuates the diabetes but also reverse the cerebral infarction in mice through its neuroprotective actions and thus Cinnamomum zeylanicum may serves as a new therapeutic alternative for management of brain damage associated with type-1 diabetes
Comparative pharmacognostical and pharmaceutical evaluation of Vyaghri haritaki avaleha - an ayurvedic formulation
Kasa is well-defined clinical condition in Brihadtrayi, clearly correlate with cough and its pathophysiology exactly correlates with the mechanism of cough reflex in contemporary medical science. Everywhere, Paediatric Outdoor Patients Department (OPD) has more than half of the total patients having respiratory tract complaints. So, to treat the disease Kasa, Vyaghri Haritaki Avaleha (VHA) was taken from Bhaishajya Ratnavali. The present work was carried out to standardize the raw drugs and finished product-VHA with comparing API standards and previous research work done in same institute to conform its identity, quality and purity of the final product. The pharmacognostical work reveals that presence of Epidermis, Cork, Pericycle, Phloem etc. from Kantakari; Lignified fibre, Stone cells etc. of Haritaki; Aluerone grains, Prismatic crystals etc. of Shunthi observed microscopically. Organoleptic features of VHA made out of the crude drugs were within the standard range. The pH value of VHA was 4.5, Water soluble extract was 71.9 %w/w, Loss on drying was 30.41 %w/w, Reducing sugar was 27.92 %w/w and High Performance Thin Layer Chromatography (HPTLC) at 254nm and 366nm resulted into 2 and 5 spots respectivel
Growth Factor Receptor Tyrosine Kinase Inhibitors in Non-small cell lung cancer
Aberrant increased expression and activation of receptor tyrosine kinases occur frequently in human carcinomas. Several small molecules targeting receptor tyrosine kinases, which have crucial roles in the growth factor signaling that promote tumor progression in various malignancies, including non-small cell lung cancer (NSCLC), are currently in clinical development. Therapeutic strategies include inhibition of growth factor tyrosine kinase function. Drugs of this type include those that target the epidermal growth factor receptor tyrosine kinase, those that target vascular endothelial growth factor receptors tyrosine kinase and those that target anaplastic lymphoma receptor tyrosine kinase. In this review we first discuss the role of receptor tyrosine kinases in human malignancies, and focus on discussing the potential use of epidermal growth factor receptor tyrosine kinase inhibitors and the vascular endothelial growth factor receptors tyrosine kinase inhibitors in NSCLC. In addition, we discuss the contribution of growth factor receptor tyrosine kinase inhibitors to the clinically observed resistance, and toxicity
Spices and condiments: safer option for treatment of hyperlipidemia
Hyperlipidemia is a lipoprotein metabolic disorder characterized by high serum Low density Lipoprotein and blood cholesterol. It is a major risk factors in the development and progression of atherosclerosis that eventually lead to cardiovascular diseases. This poses a major problem to majority of society because of the close correlation between cardiovascular diseases and lipid abnormalities. There are various features which are associated directly or indirectly as etiological factors viz. heredity, age, obesity, sex, diet, physical inactivity, hypertension, lifestyle disorders and various stress factors. For alleviation and treatment there are many ways such as allopathic medications, alternative systems like Ayurvedic, Diet control, lifestyle discipline etc. Recently Spice therapies are seen useful and effective. In India, Ayurveda and other Indian literature mentions the use of various plants and spices. Spices in diet are useful as they play effective role in the functioning of various body systems such as gastrointestinal, cardiovascular and nervous system. Along with proper food habits, diet which contains variety of spices which have been proved as hypolipidemic, can be effective in controlling hyperlipidemia. Spices used in day-to-day life as food, can also be used in the treatment of various human ailments. Along with the taste, flavor, colour and preservative property, spices also possess hypolipidemic effects. This review is focused mainly on the beneficial hypolipidemic effect of five spices (Dill, Garlic, Fenugreek, Ginger, Coriander) in the management of hyperlipidemia. This article is based on the traditional knowledge, mechanism of action for hypolipidemic activity and some experimental scientific studies done to support the use of these spices in the management of hyperlipidemia
Association of altered serum levels of Chemerin, Paraoxonase-1 (PON1), Asymmetric Dimethyl arginine (ADMA) and obesitin the development of Polycystic Ovarian Syndrome (PCOS) in Egyptian women
Chemerinwas recently added to the adipokine family and was identified in human ovarian follicles and follicular fluid that suggests a direct correlation between chemerin and PCOS. Asymmetric dimethyl arginine (ADMA) is involved in endothelial dysfunctionthe atherogenic potential of ADMA has been investigated in young patients with PCOS. Oxidative stress is considered to be implicated in the pathophysiology of PCOS.Paraoxonase 1 (PON1) is an antioxidant enzyme and its concentration has been shown to be inversely associated with oxidative stress. Objectives: Evaluation of serum chemerin, ADMA, PON1in obese and non-obese polycystic ovarian patients to postulate their role in pathogenesis of PCOS. Methods: Ninetynuligravida women aged 20-35 (60 with PCOS and 30 controls) were recruited. Fasting blood was obtained on day 2 or 3 of the menstrual cycle. Clinical evaluation, hormonal profile, Chemerin, ADMA and PON1 were assessed. Results: There was a significant increase in serum chemerinlevels in PCOS obese group when compared with PCOS non obese patients and healthy controls non obese and obese respectively. Serum ADMA level was increased significantly in PCOS obese group as compared to the PCOS non obese group , control non obese and control obese. Paraoxonase was decreased stepwise significantly from the control non obese group and control obese group to PCOS non obese patients then PCOS obese patients to. Conclusions: it could be suggested that increased chemerin has a role in PCOS development andaltered ADMA and PON1 associated withobesity and oxidative stress may exacerbate the condition
Pharmacognostic studies of the stem bark of Chloroxylon swietenia DC
Chloroxylon swietenia DC. (Family: Rutaceae) is an important traditional medicinal plant used in the treatment of various ailments like fungal infection of skin, rheumatism, common cold, cough, ophthalmic infection and cataract, wounds and as an astringent. However, detailed scientific information is not available to identify the plant material, in order to ascertain its quality and purity. In this paper, we report the pharmacognostic evaluation of the bark for the purpose of its identification and differentiation from related species. The macroscopy, microscopy, physicochemical parameters such as moisture content, ash values, extractive values, fluorescence analysis and preliminary phytochemical analysis of the bark were investigated. Transverse section of the bark shows presence of cork, cortex, medullary rays, and stone cells. Presence of cork, stone cells, phloem fibres and parenchyma cells which were also observed in the powdered sample of the bark. The result of preliminary phytochemical screening indicated presence of alkaloids, steroids, carbohydrates, proteins, phenolic compounds, tannins flavonoids and triterpenoids. The present study will be useful for its identification prior to carrying out further research work. The findings of this study will facilitate pharmacognostic standardization of the plant material and aid in the preparation of a herbal monograph for the species
