    A Review on organogel for skin aging

    Skin aging is one of the prominent problems associated with skin as each part of body ages with the time, skin is the external organ where the sign and symptoms of aging are readily evident. However cosmetics as well as pharmaceutical approaches delayed skin aging. Gel are best fitted in all these essential criteria because of their excellent appearance, smoothness, desired consistency, fast drug release, ease of manufacturing and quality assessment and admirable stability. Recently gel formulation have been modified to yield an advance drug delivery system known as ―organogels‖. Gel define as a semi-solid preparation having an external solvent phase, apolar [organogel] or polar [hydrogel] immobilized within the space available of a three dimensional network structure. Lecithin is a natural surfactant isolated from eggs or soya bean, when it combined with water and non-polar solvent, it form gels. PLO gels have gained importance in recent years as transdermal drug delivery system. It is a thermodynamically stable, visco-elastic system, which is non-irritating, odorless and biodegradable. Pluronic F127 or poloxamer is a copolymer of polyoxyethylene and polyoxypropylene which forms a thermoreversible gel in concentrations between 15-30%w/v. Water plays the role of a structure-forming agent and stabilizes the process of gel formation as it solubilizes the pluronic and other hydrophilic drugs. PLO gel system facilitates the delivery of hydrophilic as well as lipophilic drugs owing to the presence of both oil and aqueous phases within the gel system

    Role of Rauwolfia serpentina in stroke induced experimental dementia

    Objective: To investigate the Role of Rauwolfia serpentina in stroke induced experimental dementia. Methods: Methanolic root extract of Rauwolfia serpentina(RS) at a dose of 10mg/kg,20mg/kg and 40mg/kg;p.o. was studied against global cerebral ischemia induced dementia by occluding both common carotid arteries. RS at a dose of 10mg/kg, 20mg/kg and 40mg/kg;p.o. were administered for 14 days before and 7 days after both common carotid arteries occlusion(BCCAO) and were continued during behavioral testing i.e. Morris water maze test and elevated plus maze test. At the end of all experiment mice brain were removed and TBARS level, SOD level and infract size were determined.Results : Occlusion of both common carotid arteries significantly increased escape latency time(ELT) during acquisition trial and decrease, time spent in target quadrant on morris water maze, an increase in escape latency time was also observed on elevated plus maze when measured after 7 days of occlusion.Furthermore, TBARS levels, SOD levels and infract size were also increased when measured after 12 days of occlusion. Administration of RS specially at a dose of 20mg/kg and 40mg/kg;p.o. significantly attenuated these alteration and show neuroprotective effect against oxidative stress, infract size and learning and memory impairment. Conclusions: These finding suggest that Rauwolfia serpentina may have a promising role as a prophylactic drug in stroke induced experimental dementia due to its neuroprotective effect.     Pharmacogenomics – The New Trend for Personalized Medicine

    Pharmacogenetics and pharmacogenomics are two major emerging trends in medical sciences, which influence the success of drug development and therapeutics. In current times, though pharmacogenetic studies are being done extensively for research, its application for drug development needs to get started on a large scale. Pharmacogenetic studies can be used at various stages of drug development. The effect of drug target polymorphisms on drug response can be assessed and identified. In clinical studies, pharmacogenetic tests can be used for stratification of patients based on their genotype, which corresponds to their metabolizing capacity. This prevents the occurrence of severe adverse drug reactions and helps in better outcome of clinical trials. This can also reduce attrition of drug compounds. Recognition of inter-individual differences in drug response is an essential step towards optimizing therapy. Over the past decades, much evidence has emerged indicating that a substantial portion of variability in drug response is genetically determined, with age, nutrition, health status, environmental exposure, and concurrent therapy playing important contributory roles. To achieve individual drug therapy with a reasonably predictive outcome, one must further account for different patterns of drug response among geographically and ethnically distinct populations

    Formulation and Charcterization of Lake Color Obtained From Red Cabbage

    Colorants are mainly used to impart a distinctive appearance to the pharmaceutical dosage forms. The present study was carried out to develop aluminium lake color of a dye obtained from red cabbageusing different adsorbent (s) (Aluminium oxide or aluminium hydroxide or combination of both) in different ratio by simply mixing them to get dye adsorbed onto the surface of adsorbent.Batch Mode studies were carried out for 60 minutes. Aluminiumhydroxide at a concentration level of 30% w/v found to be the choice of adsorbent after optimization because maximum adsorption of dye from reaction mixture after 60 minutes and maximum % yield.After optimization of adsorbent the effect of pH and temperature also studied by formulating lake by varying these two parameters. It was observed that a higher temperature (50oC) and a slight acidic pH (6) favor the adsorption. The lake then characterized for various physicochemical properties like angle of repose, Carr’s index, hausner’s ratio, loss on drying, particle size and limit test for heavy metals. The lake was found to follow pseudo second order kinetics

    Suspension is a heterogeneous mixture including solid particles that are satisfactorily bulky for sedimentation. Generally they must be larger than one micrometer. It is when particles are left floating around freely in a solvent. Metronidazole or 2-(2 methyl-5 nitro-1 H –imidazol-1 yl) ethanol belongs to the nitro imidazole group of antibiotics whose antimicrobial property through derive from the formation of toxic free radicals by intracellular reduction. The objective of present study was to prepare suspension of Metronidazole by using different suspending agents and stability testing was to provide evidence on how the quality of suspensions varies with the time under of influence of environmental factors such as temperature, light, oxygen, moisture, other ingredient or excipients in the dosage form, particle size of drug, microbial contamination etc. and to establish a recommended storage condition. The FTIR and DSC techniques are use for characterization of active ingredient MBZ. This suspension was evaluated for appearance, pH, sedimentation volume and dissolution study for in vitro drug release. Result of evaluation indicates 1% w/v xanthan gum give optimal characteristic suspension. Suspension containing 1% w/v was exposed for the stability studies as per ICH guidelines. The suspensions were evaluated for their physicochemical parameter, particle size, drug release, drug content and microbiological assessment. The suspension shows good stability at 25ºC ± 2 ºC /60%± 5% RH, 30 ºC ± 2 ºC /65%± 5% RH and good quantitative analysis result throughout the period of study. Suspension stored at 40 ºC ± 2 ºC /75%± 5% RH shows noncompliance with the analysis parameter after 4 week of storage. This indicate high temperature storage was shows degradation of the suspension which results in crystal growth in formulation and affect the physicochemical parameter as Appearance, pH, viscosity, specific gravity, drug dissolution profile and drug content

    A neglected case of treatable genetic disorder

    Congenital adrenal hyperplasia(CAH) is a autosomal recessive genetic disorder involving adrenal hormones resulting in excessive production of androgens and hence their effects. Here we report a case of CAH which was diagnosed very late but was treated successfully. 12 years old female child came to us with ambiguous genitalia. Examination showed praders stage 4 external genitalia. Evaluated further and confirmed as a case of classic type of simple virilising congenital adrenal hyperplasia due to 21 hydroxylase deficiency. She was successfully treated with steroids and surgical correction was also done. Now child has also attained menarche and on follow up at our pediatric out patient department

    Antioxidant and antimicrobial activity of whole seed extracts of Perseaamericana Mill

    Perseaamericana Mill used in traditional medicine for the treatment of various ailments and has antibacterial, antifungal, anti-viral and wound-healing properties. In the present study, antioxidant and antibacterial activity of different extracts of Perseaamericana Mill were investigated. Antioxidant potential of plant extracts was evaluated by means of total phenolic, total flavonoids content and DPPH radical scavenging activity. The highest phenolic and flanovoid content was observed in methanol extract while the lowest was achieved in aqueous extract. At concentration of 500 SOH g/mL, DPPH radical scavenging activity was found to be highest in methanol extract (70%) and lowest in aqueous extract (51%). Antibacterial activity of different extracts was evaluated by using the disk diffusion method. Highest antibacterial activity was observed with methanol extract against S. pyogenes, while minimum activity was observed with aqueous extract against E.coli
