The purpose of this study was to develop a viable FV-deficient mouse model using CRISPR/Cas9 technology. A viable pathological model of the disease was not available to develop new therapies. A previous in silico study was performed to select a mutation causing a mild disease phenotype in humans (Thr1898Met missense). Such mutation was replicated in mice by CRISPR-mediated homology directed repair. Following crossing, homozygous individuals were subjected to coagulometry assays, including FV levels, prothrombin time (PT), and activated partial thromboplastin time (aPTT). The in silico study suggested that the mutation destabilizes FV structure of both mouse and human variants, putatively producing a mild phenotype of the disease in mice. Mendelian inheritance was observed in the offspring. No spontaneous signs of blood clotting disturbances, premature deaths or gestational dysfunctions were observed. FV levels in homozygous animals were 24.5% ± 5.1; 39.7 sec ± 2.8; PT was 61.8% ± 6.3; 23.4 sec ± 1.6 (INR = 1.47 ± 0.12); and aPTT was 46.9 sec ± 3.2. A viable FV-deficient mouse model was generated by introducing a missense mutation in FV. In Vitro Activities of MMV Malaria Box Compounds against the Apicomplexan Parasite Neospora caninum, the Causative Agent of Neosporosis in Animals.
No full text(1) Background: Neospora caninum is a major cause of abortion in cattle and represents a veterinary health problem of great economic significance. In order to identify novel chemotherapeutic agents for the treatment of neosporosis, the Medicines for Malaria Venture (MMV) Malaria Box, a unique collection of anti-malarial compounds, were screened against N. caninum tachyzoites, and the most efficient compounds were characterized in more detail. (2) Methods: A N. caninum beta-galactosidase reporter strain grown in human foreskin fibroblasts was treated with 390 compounds from the MMV Malaria Box. The IC50s of nine compounds were determined, all of which had been previously been shown to be active against another apicomplexan parasite, Theileria annulata. The effects of three of these compounds on the ultrastructure of N. caninum tachyzoites were further investigated by transmission electron microscopy at different timepoints after initiation of drug treatment. (3) Results: Five MMV Malaria Box compounds exhibited promising IC50s below 0.2 µM. The compound with the lowest IC50, namely 25 nM, was MMV665941. This compound and two others, MMV665807 and MMV009085, specifically induced distinct alterations in the tachyzoites. More specifically, aberrant structural changes were first observed in the parasite mitochondrion, and subsequently progressed to other cytoplasmic compartments of the tachyzoites. The pharmacokinetic (PK) data obtained in mice suggest that treatment with MMV665941 could be potentially useful for further in vivo studies. (4) Conclusions: We have identified five novel compounds with promising activities against N. caninum, the effects of three of these compounds were studies by transmission electron microscopy (TEM). Improving navigation and preventing erosion near San Pablo, Bolivar
No full textIn this report a study is carried out on the Magdalena River near the village of San Pablo, Colombia. The Magdalena River near San Pablo experiences two major issues which are related to navigation and erosion. The major problem for inland navigation is the sharp bend of the Magdalena River at San Pablo, which has too small a radius, causing problems for navigation. Furthermore, there are some parts in the considered river reach where the water depth is insufficient according to the requirements for navigability. Regarding the erosion problem near San Pablo, the main driving force is assumed to be high flow velocities and large near-bank water depths due to spiral flow in the outer bend. The main flow passing by the river bank of San Pablo gradually takes soil particles away, resulting in a continuous erosion process. In addition, the developed steep slope and height of the river bank results in mass failure mechanisms, increasing the erosion problems even further in the future. Several possiblemeasures have been analysed to solve this navigational problem, such as river training works consisting of groynes, longitudinal dams and a spit, where the latter one consists of a land tongue and a longitudinal dam connected to it. Measures with an even bigger impact are also considered, such as a bend cut-off and a river bypass. There are several possible measures to solve the erosion problem, such as a revetment, groynes and geogrids. Both the navigability and erosion problem can be solved in an integral way by a combination of different measures. In this study five alternatives are proposed, which are groynes, a spit, longitudinal dams, a bend cut-off and a river bypass. With the help of a multi-criteria analysis the most promising alternatives have been selected, which showed the spit to be the best in terms of reducing erosion and increasing navigation possibilities. With the help of the Delft3D model the general up- and downstream consequences have been explored of the most promising alternatives. The large river training structures, such as longitudinal dams and a bend cut-off, do not result in the desired behaviour of the river reach. Therefore, it is not advised to completely restrict the river in its flow conditions since this in general leads to more difficulties as the restriction does not allow the river to get to its equilibrium within a short period of time. However, both the groynes and the spit pose fewer restrictions to the river flow and show large improvements of the current situation in terms of navigability and erosion. The groynes are especially successful in reducing erosion problems at the river bank of San Pablo. However, the spit is more effective in driving the main part of the flow through the current navigation channel, which results in a deeper and wider channel. Therefore, it is recommended to combine the spit and the groynes to obtain an integrated solution for both erosion and navigation problems. However, for both river training works, erosion problems are encountered downstream of San Pablo. Although the author is now forgotten, he is an important figure regarding the struggles between Modernist writers and their critics. Pablo Parellada was one the key detractors of literary Modernism, a movement which he attacked through his parodies as evinced in his poetry, drama and short stories. His works contain the main pejorative features that would become the standard critique of these young poets of Spain in the early 1900's. Through the work of Pablo Parellada, this study seeks to understand the literary debates of Modernism in Spain that took place in the early twentieth century. Through this understanding, this study becomes important for it reveals the many personal characteristics that were attributed to the young Modernist poets. Some looks scream for others, even for mine... The characters that appear in this gallery of faces have caught me in some way; some are friends, or were. I have never met some of the others. However, they are all a part of me. This collection of intervened portraits is a humble homage to all anonymous beings in their purest essence. In this way, Pablo Bielli (Montevideo, 1969) introduces this series of photographs for dixit. This selection is part of the “Innominate” project, which he began in 2010, and continues developing it in different stages called albums. The author started his career in photography in 1987. He worked for several national and international media --diario El Obervador, Revista Posdata, Diario Clarín, América Economía, Editorial AVER, Revista Bla, Revista Cava Privada. Since 1998 he is a freelance photographer in the fields of advertising, editorials and international news agencies. For six years, he gave Lighting Workshop lessons at Foto Club Uruguayo. He was a founding member of Simbad Foto producciones (advertising photography) and of Fototeca Sur (archive and contents).“Sin nómina, innominados, entes autónomos deambulando. Desde el silencio más profundo encuentro miradas perdidas, hambrientas; miradas que cortan, otras ausentes. Anonimato... algunas miradas llaman a gritos a otras, incluso a la mía”.
