FOG-1, a transcriptional regulator within the haematopoietic system
Friend of GATA-1 (FOG-1) is a member of the friend of GATA (FOG)
family of proteins, which consists of large multitype zinc finger cofactors that
bind to the amino zinc finger of GATA transcription factors and modulate their
activity. FOG-1 also interacts with the C-terminal binding protein (CtBP),
mainly known as a corepressor and the nucleosome remodelling and histone
deacetylase repressive (NuRD) complex ; thus, integrating FOG-1 into the
transcription factor and chromatin modifier networks. Remarkably, the protein
activates or represses gene transcription by facilitating binding of GATA
factors to DNA, recruiting chromatin remodelling complexes, or by stabilizing
tissue-specific chromatin loops. Physical interaction between FOG and GATA
proteins in vivo is essential for the development of a broad array of tissues,
reflecting the overlapping expression patterns of these factors. Notably, within
the haematopoietic system, FOG-1 is absent in most of the myeloid lineages ;
it is expressed at high level in multipotent progenitors, erythroid and
megakaryocytic cells, low level in lymphoid and haematopoietic stem cells.
The cofactor is essential for differentiation of the erythroid and
megakaryocytic lineages, notably by interacting with GATA-1. FOG-1 also
plays a role in the T-lineage by repressing GATA-3 dependent induction of
Th2 development. Interestingly, overexpression of FOG-1 in avian
eosinophils, which do not normally express FOG-1, reprograms these
differentiated cells into multipotent cells. To study FOG-1 in mammals, we
used a novel transgenic mouse model strategy which we had designed to
generate mice with conditional overexpression of FOG-1. Our work with
enforced expression of FOG-1 in the whole murine haematopoietic system led
to a reduction in the number of circulating eosinophils, confirming and
extending to mammals the previously reported role of FOG-1 in repressing
this lineage development. Strikingly, we have identified the expression of
FOG-1 in early B lymphocytes, but not in late developmental stages such as
mature B cells and plasma cells. Moreover, FOG-1 function had never been
described in the B-lineage, where GATA factors are not expressed. Therefore,
we were intrigued by both the regulated expression of FOG-1 during B cell
development and its molecular mechanism of action in the absence of GATA
factors. Thus, we generated transgenic mice in which FOG-1 expression was
enforced at a physiologically relevant level in the B lymphoid system : in
mature B cells and from early B cell stages. We found that sustained FOG-1
expression in mature and late B cells did not affect their development or
function, contrary to our expected hypothesis. Although the mice
overexpressing FOG-1 from early B cell lineages showed only a weak
phenotype, we extensively studied FOG-1 partners in early B cell stages.
Indeed, describing FOG-1 molecular mechanism of action in the absence of
GATA factors is a question that warrant further investigation. We notably
found FOG-1 in complex with Ikaros, a transcription factor well described as
crucial for B cell development. The cofactor was also found associated to the
CtBP and NuRD epigenetic complexes in B cell lines

Media sosial TikTok dan Twitter (X) merupakan dua media sosial yang memiliki banyak pengguna di Indonesia. Berdasarkan lembaga survei We Are Social pengguna TikTok di Indonesia mencapai 109.9 juta sedangkan pengguna Twitter (X) di Indonesia mencapai 24 juta. Media sosial TikTok dan Twitter (X) seringkali menjadi tempat untuk menyampaikan pendapat atau komentar terhadap suatu hal. Universitas Terbuka merupakan suatu kampus yang memiliki media sosial TikTok dan Twitter (X) dengan ribuan pengikut. Penelitian ini dilakukan untuk mengetahui tingkat layanan bidang akademik dan non-akademik Universitas Terbuka. Data yang dianalisis sebanyak 685 data komentar pada media sosial TikTok dan Twitter (X) dengan kata kunci Universitas Terbuka. Metode yang digunakan adalah analisis menggunakan model pre-trained BERT. Pada model ini diperoleh nilai akurasi sebesar 90% dengan proporsi data latih dan data uji 80:20
Penerapan Learning Management System Untuk Karyawan Perusahaan Menggunakan Mesin Turing
Dimasa kini pembelajaran untuk mengetahui pengetahuan yang lebih yang dimiliki oleh suatu karyawan dalam perusahaan cukup sedikit hal ini dapat menimbulkan dari sisi perusahaan tidak mengetahui banyak hal tentang latar belakang ilmu pengetahuan yang dimiliki oleh seorang karyawan. Disisi karyawan dengan tidak adanya fasilitas pengembangan diri dari perusahaan suatu saat karyawan bisa saja keluar dari perusahaan tersebut karna minimnya fasilitas perusahaan. Oleh karena itu salah satu upaya untuk meningkatkan kualitas dari mutu karyawan adalah dengan menerapkan aplikasi Learning Management System (LMS) bagi karyawan dalam suatu perusahaan menggunakan model mesin turing. Sehingga dengan dibangunnya aplikasi ini diharapkan karyawan dapat belajar dengan mudah dan membantu perusahaan mengetahui seberapa jauh potensi ilmu yang dimiliki masing-masing dari karyawan dengan laporan ini berguna sebagai penilaian bagi karyawan. Aplikasi ini dilengkapi dengan fitur integrasi Application Programming Interface (API) data karyawan, untuk dapat membaca data tersebut dibutuhkan metode model mesin turing untuk membaca dan menulis ke dalam database sehingga langkah – langkahnya akan digambarkan pada diagram transisi dari tahap pembuatan kelas, sinkornasi API, hingga karyawan tersebut berhasil didaftarkan dalam kelas pembelajaran. GATA Factor-Dependent Positive-Feedback Circuit in Acute Myeloid Leukemia Cells
SummaryThe master regulatory transcription factor GATA-2 triggers hematopoietic stem and progenitor cell generation. GATA2 haploinsufficiency is implicated in myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML), and GATA2 overexpression portends a poor prognosis for AML. However, the constituents of the GATA-2-dependent genetic network mediating pathogenesis are unknown. We described a p38-dependent mechanism that phosphorylates GATA-2 and increases GATA-2 target gene activation. We demonstrate that this mechanism establishes a growth-promoting chemokine/cytokine circuit in AML cells. p38/ERK-dependent GATA-2 phosphorylation facilitated positive autoregulation of GATA2 transcription and expression of target genes, including IL1B and CXCL2. IL-1β and CXCL2 enhanced GATA-2 phosphorylation, which increased GATA-2-mediated transcriptional activation. p38/ERK-GATA-2 stimulated AML cell proliferation via CXCL2 induction. As GATA2 mRNA correlated with IL1B and CXCL2 mRNAs in AML-M5 and high expression of these genes predicted poor prognosis of cytogenetically normal AML, we propose that the circuit is functionally important in specific AML contexts

Analisis Sentimen Physical Distancing pada Twitter Menggunakan Text Mining dengan Algoritma Naive Bayes Classifier
Abstrak Physical distancing kini sedang ramai menjadi perbincangan publik sebagai salah satu cara pemerintah dalam menekan penyebaran virus covid-19 yang sedang melanda beberapa negara di belahan dunia. Tidak tersaringnya cuitan terkait physical distancing bisa memunculkan berbagai macam opini, tidak hanya opini yang positif tapi juga yang negatif. Maka dari itu, Twitter di anggap lebih diminati oleh masyarakat indonesia dikarenakan twitter dirasa lebih mudah untuk mengungkapkan opininya. Metode yang digunakan yaitu Naive Bayes Classifier (NBC). Data terkumpul dilakukan filter dari cuitan tersebut dengan menghapus data yang double maka setelah di filter data yang terambil yaitu sebanyak 547 tweets . Pada perhitungan analisis sentimen terhadap physical distancing di tengah pandemi covid-19 menggunakan metode NBC memperoleh hasil akurasi sebesar 50,26%. Tujuan dari penelitian ini, agar dapat mengkategorikan opini negatif atau positif, dari pembahasan physical distancing . Nantinya informasi terkait kebijakan Physical Distancing bisa sampai tepat informasinnya kepada masayarakat. There are public tweets related to physical distance that is free from various kinds of opinions, not only positive but also negative ones. Therefore, Indonesian people consider Twitter to be more desirable because it is easier for Indonesians to express their opinion. The method used is the Naive Bayes Classifier (NBC). The data collected was filtered from the tweets with double data, then after filtering the data were taken as many as 547 tweets. In calculating the sentiment analysis of physical distance in the middle of the Covid-19 pandemic using the NBC method, it gets an accurate result of 50.26%. The purpose of this study, to find and categorize negative or positive opinions, from the discussion of physical distancing. Methylation of GATA-4 and GATA-5 and development of sporadic gastric carcinomas
AIM: To understand the implication of GATA-4 and GATA-5 methylation in gastric carcinogenesis. METHODS: Methylation status of GATA-4 and GATA-5 CpG islands in human gastric mucosa samples, including normal gastric biopsies from 45 outpatients, gastric dysplasia [low-grade gastric intraepithelial neoplasia (GIN), n = 30; indefinite, n = 77], and 80 paired sporadic gastric carcinomas (SGC) as well as the adjacent non-neoplastic gastric tissues was analyzed by methylation specific polymerase chain reaction (MSP) and confirmed by denatured high performance liquid chromatography (DHPLC). Immunohistochemical staining was used to detect protein expression. The correlation between GATA-4 and GATA-5 methylation and clinicopathological characteristics of patients including Helicobacter pylori (H. pylori) infection was analyzed. \ RESULTS: GATA-4 and GATA-5 methylation was frequently observed in SGCs (53.8% and 61.3%, respectively) and their corresponding normal tissues (41.3% and 46.3%) by MSP. The result of MSP was consistent with that of DHPLC. Loss of both GATA-4 and GATA-5 proteins was associated with their methylation in SGCs (P = 0.01). Moreover, a high frequency of GATA-4 and GATA-5 methylation was found in both gastric low-grade GIN (57.1% and 69.6%) and indefinite for dysplasia (42.9% and 46.7%), respectively. However, GATA-4 and GATA-5 methylation was detected only in 4/32 (12.5%) and 3/39 (7.7%) of normal gastric biopsies. GATA-4 methylation in both normal gastric mucosa and low-grade GIN was also significantly associated with H.,pylori infection (P = 0.023 and 0.027, two-sides). CONCLUSION: Epigenetic inactivation of GATA-4 (and GATA-5) by methylation of CpG islands is an early frequent event during gastric carcinogenesis and is significantly correlated with H. pylori infection. Molecular cloning of grass carp (Ctenopharyngodon idellus) T-bet and GATA-3, and their expression profiles with IFN-gamma in response to grass carp reovirus (GCRV) infection
Both T-bet and GATA-3, Th1/Th2 lineage-specific transcription factors, play important roles in the development of T cells and Th1/Th2 differentiation. In this study, T-bet and GATA-3 genes were cloned from grass carp (Ctenopharyngodon idellus). The putative primary structure of the polypeptide deduced from the cDNA sequence of grass carp T-bet contained 608 aa, which possessed a T-box DNA binding domain. The putative primary structure of the polypeptide deduced from the cDNA sequence of grass carp GATA-3 contained 396 aa, which possessed two consensus zinc finger domains (C-X(2)-C-X(17)-C-X(2)-C). The YxKxHxxxRP motif, KRRLSA and LMEKs/n sequences were also conserved in this GATA-3. Phylogenetic analysis indicated that grass carp T-bet and GATA-3 group with their known counterparts with zebrafish T-bet and GATA-3 as the closest neighbor, respectively. RT-qPCR results showed that grass carp T-bet gene was highly expressed in head kidney, followed by spleen, and low expressed in gill, liver, kidney, and intestine, while GATA-3 gene was highly expressed in intestine, followed by spleen, and low expressed in gill, liver, kidney, and head kidney. Grass carp is one of the "four important domestic fish" in China and often infected by grass carp reovirus (GCRV). As yet, there is no evidence that T-bet and GATA-3 (Th1/Th2 subsets) are involved in anti-virus immune of teleost fish. In this study, by RT-qPCR, we analyzed the expression dynamics of grass carp T-bet and GATA-3 genes with IFN-gamma gene in response to GCRV infection for the first time. The expression dynamics showed that three genes might be crucially modulated by in vivo GCRV infection: (1) GCRV mainly induced a T-bet expression profile comparing to the GATA-3 expression, while the higher expression profiles of IFN-gamma correlated with the up-regulation of T-bet; (2) T-bet/IFN-gamma and GATA-3 expression changes suggest that in GCRV-infected grass carp, the common immune state of head kidney further heightens, whereas the common physiological state of intestine transforms to an anti-virus immune state.
