    ABSTRACTS Optimalization protoplast isolation of the leaves mesophyll orchid Paraphalaenopsis layockii. The obyectives of the research was to investigate the effect of combination treatment of sucrose concentrations and incubation time on protoplast isolation of the leaves mesophyll orchid of Paraphalaenopsis layockii. The factorial experiment consisted of two factors that was arranged by completely randomized design (CRD) with ten replications. The first factor was concentrations of sucrose which consists of three levels namely 0.4 M, 0.5 M and 0.6 M. The second factor was incubation time which consists of four levels namely 2 hours, 4 hours, 6 hours and 8 hours. Data were analyzed using Kruskal-Wallis test followed by MannWhitney test. The results showed that there was effect of treatment for all variables observed. The highest protoplast density and number of viable protoplasts achieved in the combined treatment of 0.6 M sucrose and 4 hours of incubation time

    RINGKASAN Dendrobium lasianthera J.J.Sm adalah jenis anggrek yang dikenal sebagai anggrek stroberi dan sebagai tanaman obat. Dari 3 organ vegetatif yaitu akar, batang dan daun bersifat toksik dan berpotensi sebagai antikanker. Tujuan penelitian tahun pertama adalah untuk mengetahui pengaruh pemberian pepton terhadap perkecambahan biji dan pembentukan protocorm (embrio tahap 3) D. lasianthera, mendapatkan konsentrasi pepton dan waktu inkubasi yang tepat dalam upaya menyediakan eksplan protocorm di atas 70% sebagai target transformasi dan mengetahui kemampuan regenerasi eksplan (protocorm) secara in vitro. Penelitian ini menggunakan eksplan biji berumur 14 minggu hasil self pollination dan dikecambahkan pada media VW diberi pepton (0, 1, 2, 3 g/L). Pengamatan perkecambahan biji dilakukan pada minggu ke-4, ke-8 dan ke-12 setelah dikultur. Setelah 12 minggu dikultur, tunas yang berasal dari perkecambahan biji disubkultur secara individual pada media VW ditambah dengan nutrien organik 15% air kelapa, 2 g/L pepton, 150 g/L bubur pisang dan tanpa nutrient organik digunakan sebagai kontrol. Plantlets Dendrobium lasianthera J.J.Sm dengan sekitar 4-5 daun, 4-6 akar (tinggi sekitar 2-3 cm) dikeluarkan dari botol kultur dan dicuci dengan menggunakan air mengalir untuk menghilangkan agar. Plantlets selanjutnya ditanam dalam pot plastik berdiameter 10 cm yang mengandung campran serabut kelapa dan sphagnum moss (3:1). Semua pot disimpan dalam greenhouse dan disiram 2 kali sehari selama aklimatisasi. Penelitian ini dirancang menggunakan rancangan acak lengkap. Data yang diperoleh dianalisis menggunakan analisis varian. Jika ada pengaruh perlakuan dilanjutkan dengan uji Duncan’s Multiple Range Test (DMRT) pada tingkat signifikansi α = 0.05 (Duncan, 1955). Hasil penelitian menunjukkan dari berbagai konsentrasi pepton yang ditambahkan ke dalam media VW padat mempengaruhi perkecambahan dan pembentukan protocorm anggrek Dendrobium lasianthera. Konsentrasi pepton terbaik yang mampu menyediakan protocorm sebagai target transformasi > 70% adalah 3 g/L. Waktu inkubasi yang tepat dalam menyediakan eksplan protocorm sebagai target transformasi adalah selama 8 minggu. Protocorm yang dihasilkan pada penelitian ini mampu ber regenerasi membentuk plantlet

    ABSTRACT Research of the structure and development pattern of somatic embryos from callus of leaf explants moon orchid Phalaenopsis amabilis (L) Bl had been done. One year old of plantlets were used as explants sources. Basal leaf of these explants were cultured in Somatic Embryo Induction Medium (SEIM) e.i.: NP(New Phalaenopsis) medium added with 2 mg/L NAA, 1 mg/L BA, 10 g/L sucrose, and 2 g/L gellan gum. Then somatic embryos were transferred to EMM (Embryo Maturation Medium) e.i.: NP medium added with 1 mg/L NAA, 1 mg/L BA, 10 g/L sucrose, and 2 g/L gellan gum. Finally, mature somatic embryo were transferred to NP medium without plant growth regulator as Embryo Germination Medium (EGM). The origin of somatic embryos initially from single cell at the pheriphery of embryogenic callus. These cells then devided in mitotic repeatedly formed globular proembryo, elongation embryo, and completed embryo.     Induksi Regenerasi Phalaenopsis amabilis (L.) Bl pada Berbagai Dosis Zat Pengatur Tumbuh

    ABSTRACT The research was aimed to study the effect of plant growth regulator 2iP, NAA and its combination to regeneration of P. amabilis. Internodes and nodes segments from pedicellus were cultured in New Phalaenopsis (NP) media supplemented either with 2iP or NAA and its combination with concentrations of 0.0:0.0; 0.0:2.5; 0.5:2.0; 1.0:1.5; 1.5:1.0; 2.0:0.5, and 2.5:0.0 mg L-1 respectively. Eksplans were sub-cultured in the same medium once four weeks during twelve weeks. Callus initiation time was observed once a day. Shoot initiation time from internodes and nodes were observed once a week. The number of shoot and root were calculated twelve weeks after inoculation. The results showed that bud regeneration from internodes was indirectly due to callus phase, while bud and root regeneration from nodes were directly without callus phase. The combination between 2iP and NAA had higher effect on regeneration than single or without plant growth regulator. Nodes of explants produced more shoots and roots than internodes of explants

    RINGKASAN Penelitian Produksi Senyawa Antimalaria dari kalus Sonchus Arvensis L. : Upaya Penanganan Penyakit Inveksi pada tahun pertama ini bertujuan untuk 1) mengetahui zat pengatur tumbuh yang dapat menginduksi tebentuknya kalus, 2) mengetahui pengaruh pemberian sukrosa dan elisitor terhadap pertumbuhan kalus; 3) mengetahui jenis elisitor yang dapat memacu terbentuknya senyawa antimalaria, 4) mengetahui profil golongan senyawa yang terdapat dalam kalus Sonchus arvensis L., 5) mengetahui aktifitas antimalaria dari masing-masing elisitor yang terdapat dalam kalus Sonchus arvensis L.. Bahan penelitian adalah daun Sonchus Arvensis L. yang disterilisasi dengan menggunakan Natrium Hypoclorit 5,25% dan ditanam pada medium Murashige & Skoog (MS) dengan perlakuan zat pengatur tumbuh 2,4 D, IAA, IBA, NAA 1ppm dan atau tanpa BAP 0,5 ppm, sukrosa (1% ; 2% ; 3% ; 4% ; 5%) dan elisitor Glutamin (0,25 g dan 0,5 g); Ammonium nitrat (0,5 g dan 1,0 g) ; Kalium nitrat (0,5 g dan 1,0 g) ; Kalium phosphat (0,1 dan 0,2 g) ; Kontrol (tanpa elisitor). Uji aktivitas antimalaria dilakukan secara in vitro menggunakan Plasmodium falciparum. Identifikasi kandungan senyawa dilakukan dengan KLT. Hasil penelitian menunjukkan bahwa perlakuan hormon 2,4 D 1ppm + BAP 0,5 ppm + sukrosa 3% pada kondisi gelap menginduksi kalus tercepat (minggu ke-2). Pemberian elisitor tidak berpengaruh nyata terhadap pertumbuhan kalus, perlakuan elisitor (Glutamin 500 g ; NH 4 NO 0,5 g dan 1,0 g) menunjukkan aktifitas positif antimalaria dengan nilai IC 50 3 = 1-10 µg/mL, dengan kandungan golongan senyawa flavonoid

    Avicennia marina is one species used for building coastal ecosystem stabilization in Surabaya shores because of its tolerance to high salinity level and resistance to wave energy. Their existence is threatened by various factors i.e. land conversion, pollution and interisland bridge existence. Growth of A. marina is currently limited by seed dormancy. In an effort to increase propagation efficiency, seed viability and vigor were compared based on collection dates and pericarp removal methods. The results showed no relationship between salinity levels of soaking water or seed buoyancy and seed viability or seed vigor. Collection dates may influence seed viability and vigor. Seeds collected in October and November germinated more quickly than those collected in December, although there were no differences in total germination. Hand-peeled seeds germinated more slowly than soaked seeds for all collections, but with no differences in total germination. Seeds collected in October and November grew more vigorously than those collected in December

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.
