Analysis of Nucleocytoplasmic Protein Shuttling by Imaging Flow Cytometry
No full textMany intracellular signal transduction events involve the reversible shuttling of proteins between the cytoplasm and the nucleus. Study of these processes requires imaging information on the protein localization in a given cell and a large number of measurements to obtain sufficient statistics on the protein localization in the whole population. The protocol describes method for quantitative imaging flow cytometry analysis of intracellular distribution of NF-kappaB in ARPE-19 cells stained with specific fluorochrome-conjugated antibodies. The described technique alone or in combination with standard flow cytometry methods can be applied to study any protein undergoing translocation from cytoplasm into the nucleus in a variety of cell lines as well as in heterogeneous primary cell population

Ivan Žuvela Šabo and His Etnographic Record from 1892
No full textU ovom tekstu otkriva se identitet autora jednog starijeg etnografskog zapisa nastalog na otoku Korčuli. Riječ je o rukopisu „Narodne pjesme junačke i žetelačke“ iz 1892., a koji se čuva u Arhivu Odsjeka za etnologiju Hrvatske akademije znanosti i umjetnosti u Zagrebu. Iako je u tom rukopisu autor potpisan kao „Ivan Žuvela, pravnik“, dosada ga se nije moglo točnije identificirati, iznesena je kriva pretpostavka, a čak se i spekuliralo da je riječ o pseudonimu. Istraživanjem dostupnih izvora i novinskih zapisa ipak se utvrdilo kako je zaista riječ o Velolučaninu Ivanu Žuveli iz grane Šabo, studentu prava koji je umro u svojoj 23-oj godini života, samo godinu dana po nastanku ovog rukopisa. Uz autora, u tekstu se detaljno progovara o rukopisu i njegovom iznimnom značaju za proučavanje nekadašnje svakodnevice, a posebno žetelačkih običaja na zapadnom dijelu otoka Korčule.This text reveals the identity of the hitherto unknown author one of the oldest known ethnographic record from the island of Korčula. The manuscript titled “Narodne pjesme junačke i žetelačke” (Folk Songs of Heroes and Harvesters) dates back to 1892 and is preserved in the Archive of the Ethnology Division at the Croatian Academy of Sciences and Arts in Zagreb. Although the author of the manuscript is signed as “Ivan Žuvela, legal scholar,” his precise identification has remained elusive, with incorrect assumptions and even speculation about a possible pseudonym. Through research of available sources and newspaper records, it has now been conclusively determined that the author is indeed Ivan Žuvela from Vela Luka, a member of the Šabo family, and a law student who passed away at the age of 23, just a year after creating this manuscript. In addition to introducing the author, this text provides a detailed exploration of the manuscript and its exceptional significance for studying the everyday life of the past, particularly the harvesting customs on the western part of the island of Korčula
Towards an Augmented Reality Game for Learning about Ivan Vazov's Literary Works

“Ivan Vazov” in Plovdiv, Bulgaria. The mural features the renowned Bulgarian author Ivan Vazov in the center, surrounded by characters from his literary works.
By using a head-mounted display in front of the mural, the players would see movement of the characters and will be able to interact with them in a gamified augmented reality environment. The serious game proposed in this paper offers a solution to the engagement of players with literature and textual heritage within a broader cultural context

No full textPrilog je napisan povodom sedamdesete godišnjice života isusovca Ivana Macana, cijenjenoga profesora filozofije na Filozofskom fakultetu Družbe Isusove u Zagrebu i njegova prvog dekana, uglednoga člana Hrvatske pokrajine Družbe Isusove te velikoga zaslužnika časopisa Obnovljeni život, s kojim je počeo surađivati već u prvoj godini njegova ponovnog izlaženja (1971). Autor želi najprije upoznati čitatelje s osnovnim biografskim podacima jubilarca, zatim se osvrnuti na njegov lik kao profesora i pedagoga, a potom iznijeti na vidjelo i njegovo filozofsko stvaranje, u kojemu osobito mjesto zauzimaju bavljenje L. Wittgensteinom, izvrsno sročeni udžbenici nekih filozofskih disciplina te prilozi iz teoretske i praktične filozofije, kojima valja pribrojiti i neke adhortativne napise. Naposljetku se donosi i cjelokupna Macanova bibliografija.This article was written to mark the seventieth year of life of Ivan Macan, S.J., the esteemed Professor of Philosophy at the Faculty of Philosophy of the Society of Jesus in Zagreb and the Faculty's first dean, whose merits include his continual contributions to the Journal Obnovljeni život since the first year of its renewed publication (1971). The author wishes to familiarize the readers with the jubilarian's basic biographical data, to portray his work as professor and pedagogue, to bring to the fore his philosophical creativeness, among which his reflections on Wittgenstein take a prominent place, as well as his excellently worded textbooks on certain philosophical disciplines and articles on theoretical and practical philosophy, which include also his adhortative writings. Finally, Ivan Macan's entire bibliography is presented

No full textThe thesis deals with photographic work of conceptual artist Ivan Kafka and questions regarding the thin line between technical documentation and its artistic quality. It focuses primarily on issues related to the documentation of art and presentation of installa-tion records. Issues accompanying photographic documentation of art are presented using examples of specific works by Ivan Kafka and his approach from a technical and artistic point of view. The text is built around thematic interviews with the author Ivan Kafka, art photographer Martin Polák and art historian Pavlina Morgan. The purpose of these interviews was to present a specification of Kafka's documentary approach from different perspectives and point to the exceptional, as well as problematic aspects of his photographic work. Individual subject areas are developed considering the problematic questions concerning documentation of photography

No full textAutor daje osvrt na pjesnički rad Ivana Mažuranića (1814. – 1890.) istaknutog htvatskog književnika i političara rođenog u Novom Vinodolskom i poznatog kao ban-pučanin zbog svojeg neplemićkog porijekla. Članak je nastao u povodu nekoliko nedavnih obljetnica vezanih uz njegov život, 125 godina od njegove smrti (1890.) te 170 godina od nastanka njegovog najznačajnijeg knjiženog djela, epa „Smrt Smail-age Čengića“ (1845.). Bio je izuzetna osoba u kojoj se sjedinilo domoljblje, talent pjesnika i politička mudrost. Uspješno je nadopunio završne dijelove Gundulićeva djela „Osman“ i pritom oživio autentičan Gundulićev pjesnički stil. Pjesništvom se intenzivno bavio tijekom jednog desetljeća, od 1835. do 1845. kada nastaje ep „Smrt Smail-age Čengića“. Pisao je o ljudskom nasilju i oholosti, a u književnom i političkom djelovanju nadahnula ga je borba Iliraca za uvođenje hrvatskog kao službenog jezika u Hrvatskom saboru 1847. godine.The author presents an overview of the work of Ivan Mažuranić (1814-1890) the prominent Croatian writer and politician born in Novi Vinodolski and famous as ban-commoner due to his non-aristocratic origin. The article was written upon the occasion of the recent anniversaries connected to his life, 125 years since his death (1890) and 170 years from the creation of his most important literally work, the epic poem ‘The Death of Smail-Age Čengić’ (1845).He was an exceptional person in whom patriotism, a talented poet and political wisdom were all combined. He successfully completed the final parts of Gundulić’s work ‘Osman’ and in doing so revived Gundulić’s authentic poetic style. He dealt intensively with poetry for a decade, from 1835 to 1845 when the epic poem ‘The Death of Smail-Age Čengić’ was created. He wrote about human violence and arrogance, and in literally and political activities the fight of the Illyrians for the introduction of Croatian to be the official language in the Croatian parliament in 1847 inspired him
Imaging Flow Cytometry of Legionella-Containing Vacuoles in Intact and Homogenized Wild-Type and Mutant Dictyostelium
No full textThe causative agent of a severe pneumonia termed "Legionnaires' disease", Legionella pneumophila, replicates within protozoan and mammalian phagocytes in a specialized intracellular compartment called the Legionella-containing vacuole (LCV). This compartment does not fuse with bactericidal lysosomes but communicates extensively with several cellular vesicle trafficking pathways and eventually associates tightly with the endoplasmic reticulum. In order to comprehend in detail the complex process of LCV formation, the identification and kinetic analysis of cellular trafficking pathway markers on the pathogen vacuole are crucial. This chapter describes imaging flow cytometry (IFC)-based methods for the objective, quantitative and high-throughput analysis of different fluorescently tagged proteins or probes on the LCV. To this end, we use the haploid amoeba Dictyostelium discoideum as an infection model for L. pneumophila, to analyze either fixed intact infected host cells or LCVs from homogenized amoebae. Parental strains and isogenic mutant amoebae are compared in order to determine the contribution of a specific host factor to LCV formation. The amoebae simultaneously produce two different fluorescently tagged probes enabling tandem quantification of two LCV markers in intact amoebae or the identification of LCVs using one probe and quantification of the other probe in host cell homogenates. The IFC approach allows rapid generation of statistically robust data from thousands of pathogen vacuoles and can be applied to other infection models
