Genome Sequencing of Strain Vm263, a Hypotoxic Strain of Valsa mali Causing Apple Valsa Canker
Apple valsa canker, caused by Valsa mali, has seriously endangered the sustainable development of the apple industry in China. Our previous study revealed that low-temperature induction increased the pathogenicity of strain Vm263, which is a hypotoxic strain of V. mali causing apple valsa canker. However, the regulatory mechanism of this phenomenon was still unclear. In this study, whole genome sequencing of strain Vm263 was performed using a combination of long-read single-molecule real-time sequencing technology (Pacific Biosciences) and MGISEQ sequencing. The genome size of strain Vm263 was 42.83 Mb and the assembly N50 was 5.09 Mb. The genome provides a valuable resource for facilitating our understanding of the response of V. mali to low temperature. [Figure: see text] Copyright © 2023 The Author(s).
A BRAZILIAN FAUST? AN INTERTEXTUAL ANALYSIS OF VALSA PARA BRUNO STEINThis paper is an intertextual analysis of the Brazilian book Valsa para Bruno Stein (1986), written by the Gaucho author Charles Kiefer, which narrative presents, since the beginning, many quotations from the book Faust (1808), written by the German author Johann Wolfgang von Goethe. The aim of this work is to comprehend the meaning of these quotations in this context, which can be considered like clues to other possible interpretations, certainly different from that ones that ignore these intertextual elements. Considering these references to Faust as an important intertextual dislocated element, it was analyzed what was the relationship between both narratives, which meanings were produced by these quotations in the Brazilian book and if these quotations confirm or subvert the Goethean narrative
Our object of study is the piece Valsa n° 6, by Nelson Rodrigues. Initially, the context of creation of the work is discussed based on the play Vestido de Noiva, by Nelson Rodrigues, when Brazilian theater undergoes structural changes, both in terms of dramaturgy and interpretation and direction. The elements that influenced the construction of the plot of Waltz n° 6 and its first montage are investigated. Next, the direction of three stagings of the play is examined, including the premiere, in 1951. As a source of research, works about the author, the play, and, with regard to the direction, sources that describe the stagings of 1951, 2012 and 2017. The purpose is to achieve a significant knowledge of the playwright Nelson Rodrigues' work and the staging proposals to which we had access
Gα proteins Gvm2 and Gvm3 regulate vegetative growth, asexual development, and pathogenicityon apple in Valsa mali.
In fungi, heterotrimeric guanine-nucleotide binding proteins (G-proteins) are key elements of signal transduction pathways, which control growth, asexual and sexual development, as well as virulence. In this study, we have identified two genes encoding heterotrimeric G protein alpha subunits, named Gvm2 and Gvm3, from Valsa mali, the causal agent of apple Valsa canker. Characterization of Gvm2 and Gvm3 mutants indicates that Gvm3 may be a crucial regulator of vegetative growth. Deletion of the corresponding gene results in a 20% reduction in growth rate. Besides, Gvm2 and Gvm3 seem to be involved in asexual reproduction, and mutants are hypersensitive to oxidative and cell membrane stresses. Interestingly, both G protein alpha subunits were most probably involved in V. mali virulence. In infection assays using Malus domestica cv. 'Fuji' leaves and twigs, the size of lesions caused by deletion mutants △Gvm2, or △Gvm3 are significantly reduced. Furthermore, many genes encoding hydrolytic enzymes-important virulence factors in V. mali-are expressed at a lower level in these deletion mutants. Our results suggest that Gvm2 and Gvm3 play an important role in virulence probably by regulation of expression of cell wall degrading enzymes. △Gvm2, and △Gvm3 mutants were further analyzed with respect to their impact on the transcript levels of genes in the cAMP/PKA pathway. The expression of the genes encoding adenylate cyclase VmAC, protein kinase A (PKA) regulatory subunit VmPKR, and PKA catalytic subunit VmPKA1 are down-regulated in both mutants. Further analyses indicated that intracellular cAMP level and PKA activity are down-regulated in the △Gvm3 mutant, but are basically unchanged in the △Gvm2 mutant. Overall, our findings indicate that both Gvm2 and Gvm3 play diverse roles in the modulation of vegetative growth, asexual development, and virulence in V. mali

Abstract: The piece of writing Raízes do Brasil (1936) by Sérgio Buarque de Holanda, in which the author reflects about the disparities between Brazilian society of the nineteenth century and the ideas of European liberalism, provides a basis to Roberto Schwarz’s criticism of Brazilian Romanticism in Ao vencedor as batatas (1977). According to Schwarz, different from its European correlate, Brazilian Romanticism would have been based on an ideological comedy, thus representing an idealization that did not correspond to reality. This article revisits such concepts from the perspective of the ballroom dancing scene in Senhora (1875), by José de Alencar. Based on the presence of the waltz in nineteenth century Brazilian literature we analyzed how the European phenomenon manifested in Brazilian territory and how the imaginary constructed around ballroom dancing was appropriated and re-read by Alencar. This work establishes a connection between Brazilian Literature and the literature produced in Europe during the same period. In addition to Roberto Schwartz’s point of view, we included Maria Cecília de Moraes Pinto’s study (1999), which investigated Alencar’s European influences and his efforts in creating a national literature. Keywords: waltz in Europe and Brazil; Romanticism; national literature.</jats:p

Clear cell calcifying epithelial odontogenic tumor: A rare case report
The calcifying epithelial odontogenic tumor (CEOT) is a benign epithelial odontogenic lesion that accounts for less than 1% of all odontogenic tumors. Clear cell variant of CEOT is extremely rare with only fifteen cases documented till date. The occurrences of clear cells may prove to be a sign of increased tumor aggressiveness, indicating the need for a more radical surgical approach
