HDAC2-deficient CD4+ T cells in a psoriasis-like mouse model of skin autoimmunity
Psoriasis ist eine Autoimmunerkrankung der Haut, von der 2–4 % der Weltbevölkerung betroffen sind und wogegen derzeit spezifische Medikamente noch fehlen. Die Haut ist ein wichtiges Barrieregewebe des Körpers, in dem Effektor-T-Zellen (Teff) und Foxp3+-regulatorische T-Zellen (Treg) die Immunhomöostase bestimmen. Daher ist das Verständnis der molekularen Mechanismen, die die Differenzierung und Funktion kutaner CD4+ T-Zellen steuern, von entscheidender Bedeutung für die Entwicklung neuartiger T-Zell-basierter Therapien für T-Zell-vermittelte Autoimmunerkrankungen der Haut. Viele Mausmodelle versuchten, menschliche Psoriasis zu reproduzieren, doch das komplexe molekulare Netzwerk, das die Krankheit charakterisiert, macht es schwierig, die Krankheit zu rekapitulieren. Histondeacetylasen (HDACs) haben unterschiedliche Funktionen bei der Regulierung der Biologie und Funktion von T-Zellen. Allerdings ist die Rolle von HDAC2 in kutanen T-Zellen noch immer nicht geklärt. Die Ziele meines Doktoratsprojekts bestehen darin, das Immunmilieu und die Wechselwirkung zwischen Immunzellen und Strukturzellen in einem etablierten Modell der Autoimmunerkrankung der Haut zu charakterisieren und die Auswirkungen des HDAC2-Mangels auf CD4+ T-Zellen bei Autoimmunerkrankungen der Haut zu untersuchen. Zu diesem Zweck verwendeten wir ein Mausmodell einer experimentellen Autoimmunentzündung der Haut (bezeichnet als K5/TGO). In diesem Modell haben wir WT- oder HDAC2-defiziente naive Ova-spezifische OTII CD4+ T-Zellen adoptiv in K5/TGO-Empfängermäuse übertragen, die eine induzierbare Expression von Ova in der Epidermis aufweisen. Adoptiv übertragene T-Zellen reagieren auf in der Haut exprimierte Ova-Antigene und lösen eine Autoimmunerkrankung aus, die hauptsächlich durch Th1- und Th17-Zellen reguliert wird. Wir beobachteten das Immunmilieu der Haut und histologische Merkmale, die Hautläsionen von Psoriasis-Patienten ähnelten. Bei der Untersuchung der Rolle von HDAC2 in CD4+-T-Zellen stellten wir bei K5/TGO-Mäusen, die HDAC2-defiziente T-Zellen erhalten hatten, eine erhöhte Überlebensrate und eine verringerte Hautentzündung fest. Darüber hinaus zeigten unsere Ergebnisse, dass das Fehlen von HDAC2, den Phänotyp (z. B. die Expression von CTLA-4) von Foxp3+ Treg reguliert. Alice in Wonderland, published in 1865, is 150 years old and continues to be one of the most popular masterpieces that is appreciated by children and adults. The article explores the polysemy of the text, its fine hermeneutical approaches, the symbolic ramifications of its visual reinterpretations – especially in the illustrations. This novel continues to fuel our imagery and, at same time, it continues to discuss the biographers of Carroll on the complex personality of this author. In particular, the article emphasizes the original and unexpected turn that Alice in Wonderland gave to children's literature, through the storytelling of a point of view – "another look" – that definitely belongs to childhood

Reading between the blurred lines: A discussion into the representation of rape and rape culture in contemporary fiction
In this dissertation, the focus will be on the representation of rape and rape culture within contemporary fiction; the aim of this is to discover how prevalent rape and rape culture is within this particular area. The thesis is split into three chapters, the first of which discusses the depictions of male rape and gender shifts in contemporary fiction, focusing on Lisbeth Salander as a rapist in The Girl with the Dragon Tattoo. The second chapter follows on from this, discussing the portrayal of victims of rape within contemporary fiction; and the last chapter debates whether or not a perpetuating rape culture means that women cannot be as sexually experimental as they wish to be. The aim of the dissertation is to focus on how authors depict rape, and whether or not this depiction is perpetuating rape culture, or simply addressing the issue within fiction. The introduction is a larger part of the dissertation, setting up exactly what rape and rape culture is, and how rape myths prevail in modern western society. There are certain areas that the dissertation has not addressed, such as race, because they are such complex issues that merely by giving them a single chapter within the dissertation would not be enough. The dissertation’s main purpose, and main area of focus, is to illustrate the perpetuating rape culture in western societies through gender inequality
