    Background Enhanced activity of histone deacetylases (HDAC) is associated with more aggressive tumour behaviour and tumour progression in various solid tumours. The over-expression of these proteins and their known functions in malignant neoplasms has led to the development of HDAC inhibitors (HDI) as new anti-neoplastic drugs. However, little is known about HDAC expression in renal cell cancer. Methods We investigated the expression of HDAC 1, 2 and 3 in 106 renal cell carcinomas and corresponding normal renal tissue by immunohistochemistry on tissue micro arrays and correlated expression data with clinico-pathological parameters including patient survival. Results Almost 60% of renal cell carcinomas expressed the HDAC isoforms 1 and 2. In contrast, HDAC 3 was only detected in 13% of all renal tumours, with particular low expression rates in the clear cell subtype. HDAC 3 was significantly higher expressed in pT1/2 tumours in comparison to pT3/4 tumours. Expression of class I HDAC isoforms correlated with each other and with the proliferative activity of the tumours. We found no prognostic value of the expression of any of the HDAC isoforms in this tumour entity. Conclusion Class I HDAC isoforms 1 and 2 are highly expressed in renal cell cancer, while HDAC 3 shows low, histology dependent expression rates. These unexpected differences in the expression patterns suggests alternative regulatory mechanisms of class I HDACs in renal cell cancer and should be taken into account when trials with isoform selective HDI are being planned. Whether HDAC expression in renal cancers is predictive of responsiveness for HDI will have to be tested in further studies

    Die vorliegende Diplomarbeit befasst sich mit dem Re-Engineering einer bestehenden Web-Anwendung zur Datenbeschaffung, -verarbeitung sowie -präsentation und wurde durch das Team 4 der ISW/Auto &MI 2 der Firma T-Systems vergeben. Das primäre Ziel dieser Arbeit bestand darin, die Trennung von Darstellung und Verarbeitungs-Logik der Altanwendung in einem Prototyp mittels einer Mehrschichtarchitektur umzusetzen. Die Lösung der Aufgabe erfolgte in drei Phasen. Die erste Phase stellte die Analyse dar, in der die Altanwendung untersucht und Anforderungsdefinitionen für den zu schaffenden Prototyp erstellt wurden. In der anschließenden Entwurfsphase erfolgte die Anfertigung eines Grobentwurfs der Anwendungsarchitektur, unter Berücksichtigung der zu realisierenden Schichten, wie auch eines Modells für die Anwendungsoberfläche. Die letzte Phase der Arbeit stellte die Implementierung der Entwürfe dar. Dies erfolgte in der Programmiersprache Java sowie unter Verwendung der Java-Webframeworks Struts 2 und Apache Tiles. Weiterhin wurden aktuelle Web-Technologien als auch Bibliotheken wie AJAX und Dojo Toolkit eingesetzt, um das Layout und die Usability der Anwendungsoberfläche zu verbessern

    Multicentre validation study of nucleic acids extraction from FFPE tissues.

    Contains fulltext : 89289.pdf (Publisher’s version ) (Open Access)In most pathology laboratories worldwide, formalin-fixed paraffin embedded (FFPE) samples are the only tissue specimens available for routine diagnostics. Although commercial kits for diagnostic molecular pathology testing are becoming available, most of the current diagnostic tests are laboratory-based assays. Thus, there is a need for standardized procedures in molecular pathology, starting from the extraction of nucleic acids. To evaluate the current methods for extracting nucleic acids from FFPE tissues, 13 European laboratories, participating to the European FP6 program IMPACTS (www.impactsnetwork.eu), isolated nucleic acids from four diagnostic FFPE tissues using their routine methods, followed by quality assessment. The DNA-extraction protocols ranged from homemade protocols to commercial kits. Except for one homemade protocol, the majority gave comparable results in terms of the quality of the extracted DNA measured by the ability to amplify differently sized control gene fragments by PCR. For array-applications or tests that require an accurately determined DNA-input, we recommend using silica based adsorption columns for DNA recovery. For RNA extractions, the best results were obtained using chromatography column based commercial kits, which resulted in the highest quantity and best assayable RNA. Quality testing using RT-PCR gave successful amplification of 200 bp-250 bp PCR products from most tested tissues. Modifications of the proteinase-K digestion time led to better results, even when commercial kits were applied. The results of the study emphasize the need for quality control of the nucleic acid extracts with standardised methods to prevent false negative results and to allow data comparison among different diagnostic laboratories.01 september 201

    1.この報文は、琉球列島内の奄美群島に属する徳之島のサビ菌についてまとめたものである。2.この島のサビ菌類については、1955年に、平塚直秀、島袋俊一、新納義馬氏らが10属28種記録したのが最初である。3.その後著者の一人田盛正雄は1961年3月本島に採集して新しく39種を追加したので、結局16属67種のサビ菌を産することが認められた。新しく加わったものは次のとおりである。すなわち、Milesina属1種、Phakopsora属2種、crossopsora属1種、Coleopucciniella属1種、Coleosporium属1種、Hamaspora属1種、Phragmidium属2種、Uromyces属7種、Puccinia属19種、Uredo属2種、Aecidium属2種。4.この報文において、つぎの10種が奄美群島のサビ菌フロラに新しく加わった。すなわち、1)Uromyces galli Dietel,2)Puccinia allii(DC.) Rudolphi,3)Puccinia caricis-macrocephalae Dietel,4)Puccinia lactucae-repentis Miyabe et T.     Inverse energy cascade in ocean macroscopic turbulence: Kolmogorov self-similarity in surface drifter observations and Richardson-Obhukov constant

    We combine two point velocity and position data from surface drifter observations in the Benguela upwelling region off the coast of Namibia. The compensated third order longitudinal velocity structure function Δu3/s\left\langle{\Delta u_{\ell}^{\rm 3}}\right\rangle/s shows a positive plateau for inertial separations ss roughly between 9 km9~\rm{km} and 120 km120~\rm{km} revealing an inverse energy cascade with energy transfer rate ε1.2±0.1107m3/s2\varepsilon\simeq 1.2 \pm 0.1 \cdot 10^{-7} m^3/s^2. Deviations from Gaussianity of the corresponding probability distribution P(Δus)P(\Delta u_{\ell} |s) of two-point velocity increments Δu\Delta u_{\ell} for given pair separation ss show up in the nth^{th} antisymetric structure functions S(n)(r)=un(P(u)P(u)duS_{-}^{(n)}(r)=\int u^n(P(u)-P(-u)d u, which scale in agreement with Kolmogorov's prediction, S(n)(r)r(n/3)S_{-}^{(n)}(r)\sim r^{(n/3)}, for n=2,4,6n=2,4,6. The combination of ε\varepsilon with Richardson dispersion s2(t)=gεt3\left\langle s^2(t)\right\rangle=g\varepsilon t^3, where s2(t)\left\langle s^2(t)\right\rangle is mean squared pair separation at time t t, reveals a Richardson-Obhukov constant of g0.11±0.03g\simeq 0.11\pm 0.03.Comment: 6 pages, 5 figure
