    A noninvasive molecular approach: exploiting species-locus-specific PCR primers in defeating numts and DNA cross-contamination of cercopithecidae

    The lack of a standardized, noninvasive molecular approach to studying genetic aspects of primates has made it hard for primatologists to decode the evolutionary history of these species. Researchers must optimize their own techniques to fully exploit the available samples. Lack of species-locus-specific primers also contributes to difficulties in using noninvasive genetic samples. Thus, the objectives of this study were to develop a standardized technique to collecting samples noninvasively, propose newly designed species-locus-specific primers, and optimize conditions for polymerase chain reaction (PCR) for Macaca fascicularis, M. nemestrina, Trachypithecus cristatus, and T. obscurus. Nine new species-locus-specific primers for three different loci of mitochondrial DNA, namely D-loop, cytochrome oxidase subunit I (COI), and cytochrome b, were successfully designed. These primers proved to be efficient in amplifying larger datasets (up to ~1,000 bp) of the targeted species in the optimized PCR conditions. The species-locus-specific primers are able to anneal to host DNA alone in highly contaminated feces of highlighted species. They can also offer alternatives measures in avoiding contamination related to nuclear insertion of mitochondrial pseudogenes (numts)

    "Olisin voinut kierähtää selän kautta koska vaan" — Osallistavan tanssitaiteen ja yleisötyön etiikka postmodernin aikakauden viitekehyksessä on Helmi Räisäsen opinnäytetyö. Opinnäytetyön tavoitteena oli tutkia yleisöä osallistavaa nykytanssiteosprosessia etiikan näkökulmasta. Aiheen pariin syvennyttiin suhteessa ohjaajakoreografi Johanna Partion ROUDAUS-nykytanssiteosprosessiin ohjaajakoreografin assistentin näkökulmaa hyödyntäen. ROUDAUS oli teos, jossa osallistavuus ilmeni yhteisenä tilallisena muutoksena. ROUDAUS esitettiin Kuopiossa ITAK-näyttämöllä (tunnetaan myös nimellä Sotku-teatteri). Esityksiä oli yhteensä neljä, aikavälillä 25.–27.5.2023. Lopputuloksena syntyi kirjallinen työ, joka käsittelee taiteen etiikkaa ja esittelee näkökulmia, joista sitä esimerkiksi voi tarkastella. Näitä näkökulmia ovat muun muassa kestävä kehitys, ekososiaalinen sivistys, reilu taide, inhimillinen turvallisuus ja turvan sekä sosiaalisen liittymisen mekanismit. ROUDAUS-teosprosessi ja assistentin näkökulma olivat lopulta konkreettinen keino maadoittaa dialogia kirjallisen opinnäytetyön kanssa. Työ syvensi Helmi Räisäsen käsitystä taiteen eettisistä rajapinnoista ja samalla se tarjoaa näitä rajapintoja koskevia reflektiomahdollisuuksia myös muille tanssialan toimijoille.This thesis aimed to explore the ethics of participatory and interactive art through contemporary dance processes. It focused on ROUDAUS — a contemporary dance production, directed and choreographed by Johanna Partio — from the perspective of the author in the role of the director-choreographer's assistant. ROUDAUS was presented at the ITAK stage in Kuopio (also known as Sotku Theatre), with a total of four performances between May 25 and May 27, 2023. The production aimed to involve the audience via an interactive aspect that manifested as a shared spatial transformation. One of the outcomes of the project is this thesis, which discusses the ethics of art-making and presents various perspectives from which those ethics may be examined. These perspectives include: sustainable development; eco-social education; fair art; human security; and the mechanisms of safety and social integration. The production process for ROUDAUS and the assistant's perspective ultimately served as a practical grounding for the dialogue with the written part of the project. The thesis deepened the author's understanding of the ethical frameworks within contemporary dance art, while simultaneously providing opportunities for further reflection and practices regarding ethical frameworks for other professionals in the field of contemporary dance

The purpose of this study is to describe some intrinsic substance and explain the Social Values that reflected in the novel entitled Kaze no Uta wo Kike. The data used for this study was obtained from the novel Kaze no Uta wo Kike by Japanese author, Haruki Murakami. The method used in this study is literature review by reading relevant references such as books, theses, and scientific journals. This study is a kind of qualitative research which uses sociological approach of literature to find out the social values that reflected in the novel entitled Kaze no Uta wo Kike. This theory is to reveal social values, such as dominant value (the desire from “Aku”’s Parents to see their children heal and live normally) and ingrained value which according to Notonagoro is divided into material value, moral value, and spritual value (but in this studyonly focuses in vital value and spirtual value). The vital value that reflected in the novel Kaze no Uta wo Kike are psychiatrist, beer and raison d’etre. Meanwhile, spiritual value is divided into truth value (luck can’t be bought by money, human will die), beauty value (beautiful Saturday night, beautiful song), moral value (forgive others, returning the favor), religius value (People’s will die, funeral procession, heaven).     Päättötyön tekijä on töissä it-suunnittelija Haapaveden kaupungilla. Hän vastaa sosiaali- ja terveyspiiri Helmen tietotekniikasta terveydenhuollon osalta. Organisaatiossa oli selvästi tarve kehittää käyttäjätunnusten hallintaa. Työn tavoitteena oli tutkia mahdollisia ongelmia nykyisessä käyttäjätunnusten anomisprosessissa ja hallinnassa. Ongelmia tutkimalla oli tarkoituksena luoda kehitys- ja parannusehdotuksia uudesta toimintatavasta. Tavoitteena oli siis kehittää nykyistä prosessia. Terveysalan ollessa kyseessä tietoperustaan vaikuttaa hyvin voimakkaasti säädetetyt lait ja asetukset. Nämä määrittelevät myös osan teknisistä ratkaisuista, kuten salasanan monimutkaisuudesta, pituudesta ja vaihtovälistä. Salasanoista ja salasanojen muodostamisesta on olemassa paljon hyviä käytöntöjä ja muistisääntöjä. Kehittämistehtävään soveltui hyvin laadullisen tutkimuksen menetelmät. Tekijä on tehnyt organsiaatiossa työskennellessään omia havaintoja ja haastatellut epävirallisest työkavereita. Tätä tiedonkeruuta täydennettiin kyselytutkimuksilla esimiehille ja verkko- ja Efficatunnuksia tekeville ylläpidon henkilöille. Kerätty aineisto tuki tehtyjä havaintoja. Lisäksi esimiehiltä tuli muutamia erittäin hyviä ja toteuttamiskelpoisia kehittämisehdotuksia. Paras toteutusmalli tekniselle ratkaisulle löytyi naapurikunnan vastaavista käytännöistä. Tämän teknisen ratkaisun käyttöönottoa ja hintaa lähdetään selvittämään.Author is working at town of Haapavesi. He is responsible for healthcare ict in Sote-Helmi. There were clear need for develop better ways to manage user rights in the organization. Object for the thesis was find out current problems in the process to request username for user. Taking closer look for possible problems that lies in the process and would help to make suggestions to improve current process. Healthcare is strongly regulated by laws and decrees. These defines some of the technical solutions such as complexity and length of password and how often you need to change your password. There are lot of best practices and thumb rules how to create a good password and how to remember your password. Qualitative research methods suits well for this case. While the author has been working in the organization he has done observation and unofficially interview his colleges. More data was also collected with survey for supervisors and people in the charge of ict. Collected data supported notes made by observation. Supervisors gave few very good suggestions how to develop future system. The best solution to improve current system found from neighbor town. Technical solution they are using will be monitored and costs calculated

    The essays collected together in this book reflect the author's varied experiences in the realms of politics and social struggle; he notes that they cannot be separated from his other experiences in his country, Egypt, over the years. These experiences extend from popular culture or folklore, through the wider political world of African liberation politics, to the Committee for the Defense of National Culture. This book is like a long trip through African culture from the 1950s to the beginning of the 21st century. These essays will most likely provoke a lot of memories, sweet and bitter; with maybe the bitter ones as the more lasting. The author notes that it appears as if the only relationship that seems to have mattered, for a long time, for the Egyptians with the rest of Africa was the river Nile, which joins the country to ten other countries, while a vast desert stands in-between. Such separation ignores the ancient relations between Pharaonic Egypt and the rest of Africa, and the role of Egypt in supporting many liberation movements on the continent. The author has set himself some tough questions in this book: Is it legitimate today to use race to sub-divide the African continent? Can this, moreover, be simply done as if race is ahistorical or an idealistic concept of identities? Or are we going to talk about Arabism in Egypt, Libya or Maghreb as if it were an identity gained with the advent of the Arabs, implying that these were 'lands with no people' - a sort of 'No Man's Land?' Or was this a fragile space that could not confront the invading empire? Or will Arabism equate with Bantuism or negroism sometimes, and Hausa and Swahili cultures at other times? These are the types of issues that Helmi Sharawy examines in this very important book. Experiences that inform this book began with the author's first encounter in March 1956, with some African youths who were in Cairo for higher studies or as representatives of liberation movements with whom he worked as an intermediary with the Egyptian national state, which work left on him an everlasting impression

ADiTV have various types of broadcasting programs ranging from information program until entertainment programs that are beneficial to society. One of them SinemA program, the program information with the soft news magazine program format. In Program Production Magazine has a production process such as pre-production, production, and post production. In the production process, especially program magazine program, production assistant has an important role. Where a production assistant secretary and spokesman for PD. The task of a production assistant is responsible for the production process. For the production assistants shall determine the course of the production process from pre-production, production, and postproduction. During the performance of the Working Class Media (KKM) for 2 months at the television station ADiTV, authors can feel how the real world of broadcasting. The author also know and understand how to work in local television which is different than the way it works with the national television station. Moreover, I also understand how the role and duties of a production assistant in a production program. With the KKM author more confidence in giving direction to the crew and the writers also be sociable and can adapt quickly. All of this can’t be separated from the guidance and direction provided by the agency. Conclusion according to the author of the work as a production assistant is a production assistant one key to the success of the production process of a program, and a production assistant is someone who responsive and knowledgeable and has a creative thinking. The suggestions for ADiTV that every program ADiTV requires innovation - innovation that program doesn’t look boring. ADiTV also must maintain a competent crew and should increase the human resources to improve the quality of programs.     Mediated music makers : Constructing author images in popular music

    The subject of the thesis is the mediated construction of author images in popular music. In the study, the construction of images is treated as a process in which artists, the media and the members of the audience participate. The notions of presented, mediated and compiled author images are used in explaining the mediation process and the various authorial roles of the agents involved. In order to explore the issue more closely, I analyse the author images of a group of popular music artists representing the genres of rock, pop and electronic dance music. The analysed material consists mostly of written media texts through which the artists authorial roles and creative responsibilities are discussed. Theoretically speaking, the starting points for the examination lie in cultural studies and discourse analysis. Even though author images may be conceived as intertextual constructions, the artist is usually presented as a recognizable figure whose purpose is to give the music its public face. This study does not, then, deal with musical authors as such, but rather with their public images and mediated constructions. Because of the author-based functioning of popular music culture and the idea of the artist s individual creative power, the collective and social processes involved in the making of popular music are often superseded by the belief in a single, originating authorship. In addition to the collective practices of music making, the roles of the media and the marketing machinery complicate attempts to clarify the sharing of authorial contributions. As the case studies demonstrate, the differences between the examined author images are connected with a number of themes ranging from issues of auteurism and stardom to the use of masked imagery and the blending of authorial voices. Also the emergence of new music technologies has affected not only the ways in which music is made, but also how the artist s authorial status and artistic identity is understood. In the study at hand, the author images of auteurs, stars, DJs and sampling artists are discussed alongside such varied topics as collective authorship, evaluative hierarchies, visual promotion and generic conventions. Taken altogether, the examined case studies shed light on the functioning of popular music culture and the ways in which musical authorship is (re)defined.Väitöskirjassani Mediated music makers. Tarkastelen tekijäkuvia rakenteina, joita artistien lisäksi määrittävät media sekä yleisö. Tutkimuskysymysten tarkastelu tapahtuu konkreettisten esimerkkitapausten analysoinnin kautta. Käsiteltävät esimerkkitapaukset rajautuvat rockin, popin sekä elektronisen tanssimusiikin genreihin. Tutkimusmateriaali koostuu populaarimusiikkia käsittelevistä mediateksteistä, kuten levyarvosteluista sekä artistien biografioista ja haastatteluista. Tutkimuksen teoreettiset lähtökohdat pohjautuvat kulttuurintutkimukseen ja diskurssianalyysiin. Tekijäkuvien intertekstuaalisesta rakentumisesta huolimatta artisti esitetään yleensä tunnistettavana hahmona, jonka kautta musiikkia markkinoidaan. Tutkimuksen päähuomio kohdistuukin siihen, kuka esitetään tekijänä miten tekijäkuvia rakennetaan, levitetään ja kulutetaan. Populaarimusiikin kollektiivisten tekotapojen ohella median ja markkinointikoneiston roolit vaikeuttavat yksittäisen artistin tekijällisen vastuun selvittämistä. Populaarimusiikkikulttuurin tekijäkeskeisyydestä seuraten usko luovaan yksilötekijään vaikuttaa yhä vahvasti siihen, miten populaarimusiikin tekijyyttä määritetään ja arvotetaan. Lähestyn populaarimusiikin tekijyyden problematiikkaa neljän pääteeman kautta, jotka määrittävät tutkimuksen pääluvut. Käsiteltävät teemat jakautuvat (1) artistin tähteyden ja julkisuusarvon sekä toisaalta julkisen näkymättömyyden tarkasteluun, (2) auteur-teorian ja autenttisuuden käsitteen pohdintaan, (3) median roolin ja genrekohtaisten konventioiden analysointiin sekä (4) musiikillisen luovuuden uudelleenmäärittelyyn suhteessa uuden musiikkiteknologian ja poststrukturalismin vaikutuksiin.     The Influence of Online Game on the Learners’ Arabic Vocabulary Achievement

    Online game is suggested as a medium that can encourage learners' acquisition of Arabic vocabulary. This research aimed to find out the learners’ vocabulary achievement before applying online game to the learners and to describe the usage of online game in improving learners’ vocabulary achievement effectively. This research used Classroom Action Research. There were 30 learners involved. Teaching processes consisted of two-cycle. Interviews, observation, tests, and documentation were the instruments of this research. The results of the tests were analyzed by using SPSS 20; the results of the interview and observation were analyzed qualitatively. The author found that online game improves learners’ vocabulary. The average score in cycle I was better than the average score in cycle II. The Pretest results in cycle I showed the greatest score obtained by the learners was 76. While in the Post test of the cycle I, the greatest score obtained by the learner was 90. Whereas in cycle II, the greatest score obtained by the learners was 100. Based on the observation, the learners were more excited on studying and focus on finishing their task; the learners could understand well the subject was given, and the learners were more enthusiastic in memorizing vocabulary. The author had successfully provided a new experience for learners who always learn conventionally by involving into the appealing and interactive online game-based learning atmosphere. The author hoped this research could be guidance for the effectuality of the next Arabic language learning through the online game in the class
