The Workplace of the Future : The Fourth Industrial Revolution, the Precariat and the Death of Hierarchies /
The Fourth Industrial Revolution is a global development that shows no signs of slowing down. In his book, The Workplace of the Future: The Fourth Industrial Revolution, the Precariat and the Death of Hierarchies, Jon-Arild Johannessen sets a chilling vision of how robots and artificial intelligence will completely disrupt and transform working life. The author contests that once the dust has settled from the Fourth Industrial Revolution, workplaces and professions will be unrecognizable and we will see the rise of a new social class: the precariat. Studies on proteins from Methylococcus capsulatus Bath : expression in Lactobacillus plantarum WCFS1, purification and large-scale fermentation
The work described in this thesis is part of a larger project investigating the non-commensal methanotroph Methylococcus capsulatus Bath. This species has shown anti-inflammatory and obesity-reducing effects in animal models when given as an extract in the diet. In silico studies of the M. capsulatus Bath genome revealed four genes (mam, tir, mif and sim) that encode proteins which are homologous to proteins with known immunomodulatory functions. They might therefore contribute to the observed effects. These proteins (Mam, Tir, Mif and Sim) were the focus of this thesis.
The genetic sequences from M. capsulatus Bath were cloned into the pSIP401 vector of the pSIP inducible gene expression system. They were successfully transformed into the lactic acid bacteria Lactobacillus plantarum WCFS1. Inducible protein expression was successfully accomplished as verified with Western blot. The genetic sequences were also cloned into the pNIC-CH vector for purification of protein. The proteins were successfully produced in Escherichia coli BL21 and purified with immobilized metal ion affinity chromatography.

After successfully producing purified protein and lysate from L. plantarum harbouring the pSIP401-derivatives, in vitro assays were performed. In the first assay, effects of these bacterial stimuli on interleukin (IL)-1b-induced production of IL-8 was investigated. In the second assay, effects on transepithelial electrical resistance when co-incubated with tumour necrosis factor (TNF)a. Neither of these assays were successful, however.
Finally, L. plantarum harbouring pSIP401_MAM or empty vector were cultured in 12-liter batch fermentations by using a 15-liter bioreactor. The resulting broth was lysed via French pressing and lyophilized before being shipped away to Copenhagen, to be used in a murine study.Arbeidet som ble gjennomført i denne masteroppgave er en del av et større prosjekt som undersøker den metanotrofe bakterien Methylococcus capsulatus Bath. Denne bakterien, som ikke finnes i tarmfloraen, har fått påvist betennelsesdempende og fedmereduserende effekter i dyreforsøk når den blir gitt som et ekstrakt i fôret. stimuliene sammen med tumour necrosis factor (TNF)-a på elektrisk motstand over et epitelcellelag undersøkt. Ingen av disse to assayene var vellykket.
Til slutt ble L. plantarum med pSIP401_MAM eller tom vektor dyrket i 12-liters batchfermenteringer
i en 15-liters bioreaktor. Bakteriemassen ble deretter lysert med French press,
før den ble frysetørket og sendt til København for å brukes i et museforsøk.M-MA
Combining mathematical programming and machine learning in electricity price forecasting
The future electricity market will have a higher share of generation from unregulated renewable energy sources because of binding targets for reducing CO2 emissions. Energy reserves for regulating the power production will be needed, and there will be an increased flow of power across country borders. As a result, electricity prices are expected to become more volatile. Electricity price forecasting models will thus need a higher level of detail and temporal resolution to capture the increased variation in the market.
With increased resolution and complexity, the computational times will increase. This thesis investigates how the complexity of a prototype hydrothermal scheduling model named PriMod under development at SINTEF Energy Research can be reduced, and thus saving computational times, by exploiting patterns in the input data.
The potential for reducing the problem size has been assessed by removing constraints with a dual value of zero in the optimal solution of the linear programming (LP) problems produced by PriMod. Findings show that the number of constraints in the current implementation of the prototype model on two different datasets can be reduced with 77% and 93%, respectively. A genetic algorithm was able to further reduce the number of constraints, while having an insignificant impact on the modeled total costs.
An algorithm for reducing the number of constraints by combining water balances is presented. The algorithm was used with an LP problem from PriMod and proved effective in reducing the number of water balances while having an insigniffcant impact on the total system cost. After combining the water balances, the number of water balances were reduced by 64%.
Finally, a framework for reducing the problem sizes is presented. In this framework, constraints that can be removed are identified and used as target data to train a machine learning model. The framework was tested using an artificial neural network to predict which constraints can be removed, based on data on inflow, wind power production and load. The mean computation time of 1092 LP problems generated by PriMod was reduced by 55%, while the mean percentage error in total system cost was 0.14%. The machine learning task is a multi-label classification problem, which is complex and requires more training data and models with more parameters than the one presented in thesis, to achieve higher prediction accuracies
Combining Machine Learning and Optimization for Efficient Price Forecasting
We present a framework based on machine learning for reducing the problem size of a short-term hydrothermal scheduling optimization model applied for price forecasting. The general idea is to reduce the optimization problem dimensions by finding patterns in input data, and without compromising the solution quality. Mallarmé’s presence
Dans sa thèse intitulée La présence de Mallarmé, Arild Michel Bakken interroge la présence de Mallarmé dans son œuvre. Contre l’idée dominante qui fait de Mallarmé le poète de l’absence, Bakken montre que la figure du poète est très présente dans le texte, et que c’est en réalité cette figure qui assure la cohérence de l’œuvre. Utilisant une méthode rhétorique centrée sur l’expérience du lecteur, la thèse montre comment Mallarmé se met en scène. La figure de l’auteur est bien ancrée dans la société de son temps, comme Français, comme petit-bourgeois, comme mari et père. Cette figure apparaît clairement comme une figure de poète, mais aussi comme « homme de lettres » et comme journaliste. La thèse montre aussi que le lecteur peut suivre la vie intérieure de la figure comme voyant et comme penseur. Même dans les textes les plus impersonnels, l’auteur n’est pas absent, mais assume la posture de la mystérieuse « Figure que Nul n’est », qui représente l’essence de la subjectivité humaine. Par la mise en scène de sa figure dans l’œuvre, Mallarmé cherche à séduire le lecteur, à obtenir une « gloire », à conférer à son œuvre une valeur. Fleksibelt inflasjonsmålsregime og pengepolitiske utfordringer i Norge : 2001 – 2005
Denne utredningen omhandler pengepolitikk med særlig vekt på innføringen av fleksibelt inflasjonsmål i Norge i tiden etter våren 2001. Først prøver jeg å etablere en forståelse for hva pengepolitikk er. Deretter gir jeg en kort innføring i pengepolitiske mål og knytter dette til ulike pengepolitiske regimer.
Siden utredningens hovedvekt knytter seg til pengepolitikk under et fleksibelt inflasjonsmålsregime, har jeg videre fokusert på en enkel modell for hvordan pengepolitikken bør innrettes dersom sentralbanken styrer etter et eksplisitt inflasjonsmål. Modellen er hentet fra en artikkel av Røisland og Sveen (2004) fra Norges Bank.
Deretter prøver jeg gjennom pengepolitisk teori og modellen for pengepolitikk under et inflasjonsmål å vise at overgangen til et fleksibelt inflasjonsmål var riktig, men også at et slikt regime ikke gir et fasitsvar for hvordan pengepolitikken bør utformes. Jeg ser også på den konjunkturmessige utviklingen i Norge de siste fem årene og gir en gjennomgang av noen av valgene Norges Bank har gjort denne perioden.
