    Significantly decreased hypoxanthine levels in bone marrow plasma of aplastic anemia patients revealed by 800 MHz NMR based metabolomics analysis

    Background: Compared to plasma, the hypoxanthine (HX) concentration is three times higher in the human bone marrow [R1] and its availability is crucial to regulate the de novo purine synthesis in human bone marrow in vivo [R1]. Studies have shown that circulatory HX levels in the plasma increase in patients with solid tumors and leukemia [R2]. Aplastic anemia (AA) is rare disorder of bone marrow failure characterized by hypocellularity and peripheral cytopenias. As purine biosynthesis is essential for normal cell growth and proliferation, we hypothesized that bone marrow HX levels will be decreased in aplastic anemia (AA) patients and may reset back partially in AA patients showing clinical improvement. Aim: To compare the HX levels in bone marrow plasma samples of AA patients and control subjects and further to check its metabolic reprogramming in AA patients showing clinical improvement. Methods: The HX levels of bone marrow plasma metabolites were estimated for 56 AA patients and 20 control subjects using 800 MHz NMR spectroscopy. The NMR spectra were analyzed using NMR suite of commercial software (CHENOMX). The levels of altered metabolites in AA patients w.r.t control were estimated and the values are reported as Mean ± SD (SD is standard deviation). The diagnostic potential is evaluated employing standard receiver operating characteristic (ROC) curve analysis. Results: The present study aims to compare the bone marrow plasma metabolic profiles of hypoxanthine between age and sex matched AA patients (N=40; mean age 32.31±16.03, male/female ratio of 22/18) and follow-up AA patients (N=16) with respect to age matched control subjects (N=20, mean age=39.5±18.48 years, male/female ratio of 11/9). The comparison revealed that the HX levels are significantly decreased in AA patients (HX=3.62 ± 3.17) compared to NC subjects (HX = 20.89 ± 12.73) and improved slightly in follow-up patients (HX=6.21 ±4.77, however no statistically significant change compared to AA patients). Further, the circulatory HX levels exhibit significant diagnostic potential with area under ROC (AUROC) curve values equal to 0.96 [95%CI = 0.92–1.00]. Concluding remarks: The depleted HX levels in the bone marrow plasma suggested altered/compromised purine biosynthesis/metabolism in AA patients and future studies are warranted to underscore its role in the disease pathobiology.     Model data and post-processed data supporting the creation of the manuscript "Estimating the Meridional Extent of Adiabatic Mixing in the Stratosphere using Age-of-Air" submitted to JGR:Atmospheres in August 2022. 1) The netCDF files created through post-processing of full model data in FORTRAN are shared in the /data/ directory. These file contains the zonal mean circulation statistics based on Gupta et al. (2020), age-of-air transport diagnostics based on Linz et al. (2021), and the novel \Gamma-\Theta circulation streamfunction introduced in this study. The /data/ directory also contains MATLAB .mat data files for the transport diagnostics obtained from WACCM. 150 days of actual GFDL-FV3 model data in the northern hemisphere, between 0.1 hPa-500 hPa pressure levels is also provided to support external computations and validation. 2) The Jupyter notebook used for final computation and figures production is provided in .ipynb, .html and .pdf formats in /code/.     An application of the ABS LX algorithm to multiple sequence alignement

    Il problema del multiple sequence alignement porta alla soluzione di sistemi lineari malcondizionati, per il cui trattamento si è usato con esiti positivi il metodo implicito LOX della classe ABSWe present an application of ABS algorithms for multiple sequence alignment (MSA). The Markov decision process (MDP) based model leads to a linear programming problem (LPP), whose solution is linked to a suggested alignment. The important features of our work include the facility of alignment of multiple sequences simultaneously and no limit for the length of the sequences. Our goal here is to avoid the excessive computing time, needed by dynamic programming based algorithms for alignment of a large number of sequences. In an attempt to demonstrate the integration of the ABS approach with complex mathematical frameworks, we apply the ABS implicit LX algorithm to elucidate the LPP, constructed with the assistance of MDP. The MDP applied for MSA is a pragmatic approach and entails a scope for future work. Programming is done in the MATLAB environment

    Serum albumin predicts survival in Indian adult diffuse large B cell lymphoma patients in the rituximab era

    Objective: The present study was done to evaluate the prognostic impact of the National Comprehensive Cancer Network-International Prognostic Index (NCCN-IPI) and serum albumin levels in the treatment outcome of Indian diffuse large B-cell lymphoma (DLBCL) patients in the rituximab era. Patients and Methods: We retrospectively analyzed the data (2013–2016) of 135 newly diagnosed DLBCL cases ≥18 years of age. All patients received Rituximab-Cyclophosphamide, Adriamycin, Vincristine, Prednisone (R-CHOP) chemotherapy. The analysis was carried out to assess the overall survival (OS) and progression-free survival (PFS) and the prognostic factors predicting the outcome. Results: Of the 135 patients in the study, 89 (65.9%) had B-symptoms, 20 (14.8%) had bulky disease, 79 (58.5%) had advanced disease (Stage III and IV), and 29 (21.5%) had primary extranodal involvement. Serum albumin ≤3.5 g% was present in 71 (52.6%) patients. About 74 (54.8%) cases were risk stratified to NCCN-IPI high-intermediate-risk group, while 18 (13.3%) patients were categorized into high-risk group. The median PFS and OS of our study cohort were 19 months (95% confidence interval [CI] = 2.59–35.4) and 38 months (95% CI = 9.02–55.68), respectively. Serum albumin ≤3.5 g/dl was significantly associated with poor OS (hazard ratio [HR] = 3.99, 95% CI = 2.25–7.07, P < 0.001) and PFS (HR = 3.71, 95% CI = 2.20–6.26, P < 0.002). Similarly, low NCCN-IPI (<4) was significantly associated with improved OS (HR = 0.21, 95% CI = 0.09–0.47, P < 0.005) and PFS (HR = 0.19, 95% CI = 0.09–0.41, P < 0.001), respectively. These two factors (serum albumin and NCCN-IPI) retained their prognostic significance with respect to OS and PFS in the multivariate analysis. Conclusion: The NCCN-IPI prognostic model and serum albumin levels have independent prognostic significance in Indian DLBCL patients. Serum albumin is a readily available, easy to standardize, and cheap investigation requiring no specialized expertise and holds promise for being incorporated in future DLBCL prognostic risk models

    We start from the two-dimensional Gross-Pitaevskii equation (GPE) and develop algorithms for the ab-initio determination of the temperature (T) dependence of the mutual-friction coefficients, α and α, and the normal-fluid density Pn, which appear as parameters in the Hall-Vinen-Bekharevich-Khalatnikov (HVBK) two-fluid model for a superfluid. In the second part of our study, we elucidate the statistical properties of two-dimensional, homogeneous, isotropic superfluid turbulence in the simplified HVBK model, with values for the mutual-friction coefficients that are comparable to those we obtain from the first part of our study
